Abstract:The dry seeds of four varieties of seedy watermelon were used as experimental materials. The seed cucurbitin from seedy watermelon were analyzed by SDS-PAGE and IEF-SDS-PAGE. The results showed that the cucurbitin has a molecular mass of 55 kD, it can be dissociated into six subunits of MW. 37kD, 35kD, 33kD, 23kD, 19 kD, and 18 kD in SDS-PAGE under reducing conditions. The 37 to 33 kD proteins are acidic subunit and their isoelectric points lay between pH 5.5 and 6.2, and the 23 to 18 kD proteins are basic subunit and their isoelectric points lay between pH 8.9 and 9.6. The three acidic subunits and the three basic subunits linked by disulphide bonds were also found in the seed cucurbitin. Therefore it is proposed that cucurbitin of seedy watermelon are homologous to leguminlike protein.
