血清胸腺因子(STF)的合成

本文报道了用液相法合成血清胸腺因子(STF)的结果。由于羧端L-天门冬酰胺羧基采用了对硝基苄基保护,每步均可方便地得到纯度和收率较高的中间肽结晶,最后经催化氢解温和地去除全部保护基获得有生物活性的STF纯品。

Synthesis of Serum Thumic Factor

In this paper,the synthesis of a thymic nonapeptide designated as STF "Serum Thymic Factor" with the sequence of Gln·la·Lys·er·ln·ly·ly·er·sn· OH is reported. In this synthesis the conventional method in solution has been used with the majority of coupling reactions carried out by the mix anhydride method.By using nitrobenzyl group as the protecting group for the C-terminal as paragine, the intermediate peptides could be easily crystallized and the recoveries were satiafactory in all steps. In the last step of the synthesis, all the protecting group, such as Cbz (the N-terminal protecting group) , Bzl (the side chain protecting group) and Onb (the C-terminal protecting group) were cleaved by catalytic hydrogenation. The final product was purified by gel filtration chromatog-raphy.The characterization and identification of purity of the synthetic nonapeptide were established by elementary analysis, melting point,optical rotation determination together with T.L.C.and amiao acid analysis of acid hydrolyaates. The final product, the synthetic nonalpeptide behaved like the isolated natural STF in high voltage electrophoresis and was proved to be biologically active in the sheep red cell rosette assay.
