Relationship between Cell Growth and the Core Fucose in N-glycans of Cell Surface Glycoproteins

Abstract:H-Mannose labeled N-lonked glyeopeptides were prepared from the cell surface of human hepatocareinoma cell SMMC-7721, and the contents of core moose (c-Fuc) in the N-glyeans were analyzed by means of LCA affinity chromatography. It was found that after the cells were treated with two differention inducers, retinoic acid(RA) and dibntyry1-3', 5'-eyelio AMP (db-cAMP) for 1 to 5 days, the percentage of c-Fue in biantennary (bi) complex type N-glycans was decreased day by day, and the c-Fuc percentage of C2C2,6 triantennary (tri 226) or tetraantennary(tetra) N-glythens was also deoreased at 5 days culture of cells. Conversely, when cells were treated with the Proliferation stimulator,phorbol misstate acetate (PMA) for 5 days, the percentage of c-Fue in (bi)fractions was inereased. These readlts suggest that the c-Fuo eontent in N-glycans of surface glyooproteins is closely related to cell growth,differentiation and proliferation.
