尿激酶及其抑制物对未成年小鼠排卵的影响

摘    要:利用未成年小鼠超数排卵方法,研究了纤溶酶原激活因子——尿激酶和它的抑制物——6氨基己酸对排卵的作用。实验结果表明:18日龄小鼠在注射PMSG和hCG条件下,增注尿激酶800U,排卵动物数及平均排卵数与对照组比较有显著性差异。如果仅用PMSG和尿激酶,不能引起动物排卵。21日龄小鼠在注射PMSG后再注射不同剂量hCG情况下增注尿激酶,平均排卵数均比相应对照组有显著性差异,但hCG注射量为l.25IU组,增注尿激酶后,实验组与对照组平均排卵数无显著性差异。用尿激酶抑制剂6氨基己酸可以有效抑制超数排卵的效果。实验结果提示,尿激酶只有在hCG存在条件下对小鼠排卵有促进作用,而尿激酶本身不具有诱发排卵的作用。

Abstract:The effects of urokinase and 6-aminocaproic acid (an inhibitor of urokinase) on ovulation were studied by using superovulatory technique in immature mice.Eighteen days old female mice were divided into three groups. All animals were injected with a superovulatory dose (10 IU) of PMSG. Fourty eight hours later, in group 1, each mouse was given hCG 20 IU as control. In group 2, each of the animal was given urokinase 800 U, but not injected with hCG in group 3. The ova ovulated by animals in three groups were 5.6++++++++++++3.9, 23.5++++++++++++8.0 and 0, respectively. There is a significant difference was found between group 1 and 2 (P<0.01). No ovulation was found in all the animals of group 3.Mice aged twenty one days were separated into four groups. Does of PMSG for each mouse is the same as used in 18 days old mice. In group 1, 2 and 3, the control animals were respectively given hCG 5, 2.5 and 1.25 IU. All experimental mice not only injected with hCG, but also were given urokinase 800 U. Ova ovulated by control animals were 27.4+++++++++++4.0, 23.2+++++++++++++++6.9 and 5.4++++++++++++++2.4, respectively. The experimental animals ovulated 49.3+++++++++++++++3.9, 47.3++++++++++++4.7 and 11.4+++++++++++++++4.1 ova, respectively. In group 1 and 2, the experimental animals had significant difference when compaired with controls (P<0.01). However, in group 4, the mice were injected PMSG and urokinase 800 U, no ovulation was found in all of the animals.Ovulation was significantly reduced when the mice received 6-aminocaproic acid (P< 0.01).The results auggest that the urokinase is capable of increasing ovulation rate for 18 and 21 days old mice. This promoting effect of urokinase on ovulation can only be maintained with the presence of hCG.
