肾缺血再灌流损伤过程中肾小管上皮细胞色素氧化酶活性变化及TAD防治作用的研究

摘    要:本实验选用大鼠一侧肾切除,对侧肾缺血60min动物模型,用组织化学方法观察再灌流15min,24h髓质外带肾小管上皮细胞色素氧化酶活性的变化及还原型谷胱甘肽(TAD)对它们的影响。结果发现60min肾缺血后再灌流可致细胞色素氧化酶活性呈进行性降低。给TAD能一定程度地保护细胞色素氧化酶活性。提示氧自由基可能损害细胞色素氧化酶活性,细胞色素氧化酶活性降低在肾缺血再灌流损伤中可能起重要作用。

THE EXPERIMENTAL STUDIES ON THE CHANGES OF CYTOCHROME OXIDASE ACTIVITY OF THE RENAL TUBULAR EPITHELIA AND THE PROTECTIVE EFFECTS OF TAD DURING THE INJURY OF RENAL ISCHEMIA AND REPERFUSION
Abstract:The rat renal ischemia model with right nephrorectomy and occlusion of the left renal pedicle for 60 min was used in this experiment. The changes of cytochrome oxidase activity in the renal tubular epithelia at out strip of the medulla and the effects of TAD (reduced glutathione) on the cytochrome oxidase were studied at the time of 15 min and 24 h reperfusion after 60 min renal ischemia by means of light microscopic histochemistry. We found that 60 min ischemia and reperfusion could result in a progressive reduction of cytochrome oxidase activity. TAD could protect cytochrome oxidase activity in some degree. These results suggest that oxygen free radicals may damage the activity of cytochrome oxidase, the reduction of which may play an important role in the injury of renal ischemia and reperfusion.
