Abstract:[14C]Phenylalanyl-tRNA bound to Escherichia coli ribosomes in the presence of poly U and varying levels of deacylated tRNA was isolated by adsorption to cellulose nitrate filters. The extent of binding of the radioisotope was inhibited by deacylated tRNA in a manner suggesting two binding sites for phenylalanyl-tRNA one sensitive to inhibition by deacylated tRNA and the other insensitive. The data did not fit a model that assumed identical sites with equal affinity for deacylated or acylated tRNA. Combinations of either tetracycline or streptomycin and deacylated tRNA gave additive inhibitions, while a mixture of both antibiotics did not produce a greater effect than either tested alone. Thus the antibiotics may inhibit the binding of phenylalanyl-tRNA at one type of ribosomal site, whereas deacylated tRNA may inhibit binding at the other.
