糖化酶一个主要组分的纯化和有关光谱的测定

摘    要:采用糖化酶粗酶制剂,经抽提、硫铵分级和DEAE-纤维素层析等程序纯化了糖化酶一个主要组分,并经电泳鉴定被纯化至均一状态,分子量为89000。于不同条件下,测定了该组分的圆二色性谱和荧光发射谱,取得了关于该组分构象的一些重要结果。

Abstract:in this study, a primary component of Glucoamylase was purified by extraction, ammonium sulfate fractionation, DEAE-cellulose 52 chromatography, purity examined by PAGE was shown by the presence of a single band. In the presence of different concentrations of Ca2 ,iodoacetic acid or urea, circular dichroism spectra and nuorescence spectra of this component were measured. In the experiment conditions, each ligands could change the conformation of this component. These results could offer some important information about the conformation of this component.
