虫草素联合阿霉素干预乳腺癌细胞增殖及转移作用

摘    要:研究了虫草素联合阿霉素在体外抑制三阴性乳腺癌MDA-MB-231细胞增殖及转移作用,评估了联合用药的作用效应,为虫草素在临床应用上增强抗乳腺癌作用提供了科学数据。研究结果表明,联合用药比单独用药作用效果更明显,根据Chou-Talalay法显示出在80μmol/L虫草素联合1μmol/L阿霉素的条件下,联合用药协同作用最优,CI值为0.665,细胞抑制率达到60.31%±1.06%;与对照组相比,平板克隆形成实验证明联合用药显著抑制细胞增殖,克隆形成率仅为7.03%±1.19%;显微观察细胞形态变化表明联合用药明显影响细胞生长;Hochest 33258染色、DNA Ladder发现联合用药对细胞凋亡诱导作用更显著,细胞凋亡率可达78.52%±11.18%;细胞划痕愈合实验检测联合用药显著抑制细胞迁移,细胞迁移率仅为18.82%±2.43%。本研究确证虫草素可协助阿霉素治疗乳腺癌的增敏作用。

Effects of cordycepin combined with doxorubicin on proliferation and metastasis of breast cancer cells

Abstract:The inhibitory effect of cordycepin combined with doxorubicin on proliferation and metastasis of triple negative breast cancer MDA-MB-231 cells in vitro was studied, and the effect of compound remedy was evaluated, for the purposes of enhancement of anti-breast cancer effect of cordycepin in clinical application. The results showed that cordycepin and doxorubicin compound was more obviously effective than single drug treatment. According to Chou-Talalay method, 80μmol/L cordycepin combined with 1μmol/L doxorubicin manifests the best synergistic effect, and the CI value is 0.665, and the cell inhibition rate reaches 60.31%±1.06%. Compared with the experimental control, such a drug combination significantly inhibited cell proliferation, and the plate clone formation rate was only 7.03%±1.19%. Microscopic observation showed that the compound remedy significantly affected cell growth. Hochest 33258 staining and DNA Ladder indicate that the compound remedy has a more significant effect on the induction of apoptosis, and the apoptosis rate can reach 78.52%±11.18%. The cell scratch healing test detected that the compound significantly inhibited cell migration, and the cell migration rate was only 18.82%±2.43%. This study confirms that cordycepin can assist doxorubicin in the sensitization of breast cancer.
