摘    要:目的探讨EZH2在胃癌组织中表达的意义及与幽门螺杆菌L型(Helicobacter pylori-L,Hp-L)感染的关系。方法 (1)应用免疫组织化学Elivision法和革兰染色法检测80例胃癌组织及30例癌旁组织(对照组)中EZH2蛋白的表达和Hp-L型的感染情况;(2)采用逆转录多聚酶链反应(RT-PCR)技术检测30例新鲜胃癌组织及对应切缘正常胃黏膜组织(对照组)中EZH2的mRNA表达。结果胃癌组EZH2蛋白表达的阳性率高于对照组(P<0.05),且EZH2表达水平升高与肿瘤大小、浸润深度、淋巴结转移和TNM分期有关(P<0.05),与性别、年龄无关(P>0.05);RT-PCR显示,肿瘤组织、远端正常对照组织的EZH2表达量差异明显(P<0.01)。胃癌组Hp-L型检出率78.8%(63/80)与对照组23.3%(7/30)有显著性差异(P<0.05),与免疫组化Hp-L型抗原表达率73.8%(59/80)无显著性差异(P>0.05),Hp-L检出阳性率为71.3%(57/80);癌组中Hp-L型感染阳性组的EZH2表达阳性率高于Hp-L型阴性组(P<0.05),且Hp-L型阳性率和EZH2蛋白的表达呈正相关(r=0.250,P<0.05)。结论 EZH2蛋白和mRNA在胃癌中的表达增加,且与胃癌的浸润、转移相关,其机制可能与幽门螺杆菌L型(Hp-L型)感染有关。

Abstract:Objective To investigate the signiiicance ot EZHZ protein expression m gastric carcinoma (GC)and its relationship with the Helicobacler pylon L-form (Hp L) infection. Methods (1) Gram,s and immunohistochemical staining were used to deled the Hp L infection and the expression of EZH2 protein in 80 samples of GC tissue and 30 corresponding adjacent tissues (control group). (2) The expression of EZH2 mRNA in 30 cases of fresh GC tissue and adjacent normal gastric mucosa (control group) was detected by scmi quantitative reverse transcriplase polymerase chain reaction (RT-PCR). Results The positive rate of EZH2 protein expression in the GC group was significantly higher than in the control group (P〈0. 05). EZH2 protein expression increased with tumor size, depth of invasion, lymph node metastasis and TNM slage (P〈0.05), but had no correlation with patients' sex and age (P〉0.05). RT PCR showed that the EZH2 mRNA expression was significantly different between GC and distal normal control tissues (P~-.0.01). The positive rate of Hp-L infection in GC group (78.8%) was significantly higher than that in the control group (23.3%) (P〈0.05). The antigenof Hp-I. was detected in 73.8 % cases byimmunohis-tochemical techniques, and there There was no difference between the results obtained from each staining method respectiwdy (P)0.05). The positive rate of Hp-I. infection was 71.3 % (57/80). In GC group, the expression of EZH2 was significantly higher in Hp-I. infection positive cases than that in negative cases (P〈0.05). Conclusion The expression of EZH2 protein and mRNA in gastric cancer is increased and cor-related with tumor's invasion and metastasis, and the mechanism may be related to Helicobacter pylori I7 form infection.
