摘 要: | 目的研究布拉酵母菌对胃黏膜上皮细胞损伤的保护作用。方法建立人胃黏膜上皮细胞GES-1单层细胞模型,并将其分为对照组,布拉酵母菌上清培养组,阿司匹林处理组(18.32mmol/L),阿司匹林+布拉酵母菌上清液组。采用流式细胞术检测各组细胞凋亡率;采用免疫荧光、Western blot方法检测胃黏膜上皮细胞中Occludin和ZO-1蛋白、JNK蛋白、ERK蛋白、磷酸化JNK(p-JNK)蛋白、磷酸化ERK(p-ERK)蛋白表达水平。结果流式细胞术显示阿司匹林+布拉酵母菌上清液组凋亡率低于阿司匹林处理组,差异有统计学意义(P0.05);免疫荧光结果显示,阿司匹林+布拉酵母菌上清液组Occludin蛋白、ZO-1蛋白表达较阿司匹林处理组增加,Western blot结果显示,阿司匹林+布拉酵母菌上清液组的该两种蛋白表达量较阿司匹林处理组增加,阿司匹林+布拉酵母菌上清液组p-ERK、p-JNK蛋白表达较阿司匹林处理组减少。结论布拉酵母菌对阿司匹林造成的胃黏膜上皮细胞损伤有保护作用,其机制可能通过抑制MAPK信号通路中ERK、JNK的激活而上调Occludin和ZO-1蛋白的表达。

Protective effect of Saccharomyces boulardii on
injury of human GES-1 cells and the possible mechanisms |

Abstract: | Abstract: Objective To investigate the protective effect of Saccharomyces boulardii on the injury of human GES-1 cells induced by Asprin, its possible mechanisms and clinical usage. Methods GES-1 cells were cultured in vitro, and divided into four groups: Control group, Saccharomyces boulardii group, Aspirin group, and Aspirin+Saccharomyces boulardii group. The apoptosis rates of GES-1 cells in the four groups were tested with Flow cytometry. The levels of Occludin as well as ZO-1, JNK, ERK, p-JNK and p-ERK were determined by using immunofluorescence and Western blot. Results Flow cytometry showed that the apoptosis rate was 48.30%±1.63% in Asprin group, significantly higher than the 25.83%±2.00% in Aspirin+Saccharomyces boulardii group (P<0.05). Immunofluorescence revealed that the levels of ZO-1 and Occludin were increased in Saccharomyces boulardii+Aspirin group, which was confirmed by Western blot. The expression of p-ERK and p-MAPK was lower in Saccharomyces boulardii+Asprin group than in Aspirin group, according to Western blot. Conclusion Saccharomyces boulardii has a protective effect on the injury of human GES-1 cells induced by aspirin, the mechanism of which is probably the inhibition of both ERK and JNK signaling pathways and the up-regulation of the expression of Occludin and ZO-1 proteins. |
