1,4‐Dithiothreitol (DTT) has wide applications in cell biology and biochemistry. Development of effective methods for monitoring DTT in biological systems is important for the safe handling and study of toxicity to humans. Herein, we describe a two‐photon fluorescence probe (Rh‐DTT) to detect DTT in living systems for the first time. Rh‐DTT showed high selectivity and sensitivity to DTT. Rh‐DTT can be successfully used for the two‐photon imaging of DTT in living cells, and also can detect DTT in living tissues and mice.  相似文献

天冬氨酰蛋白酶(β-site amyloid precursor protein cleaving enzyme 1, BACE1)作为治疗阿尔兹海默症的潜在靶点,其抑制剂的开发已成为医学领域的重要研究方向。本文以59个氨基恶唑啉呫吨类BACE1抑制剂为研究对象,运用比较分子相似性指数(comparative molecular similarity index, CoMSIA)和分子对接方法,深入挖掘影响抑制剂活性的特征结构,以及抑制剂与BACE1间的结合模式和作用力类型,并以此为基础设计新型抑制剂并预测其活性。CoMSIA模拟结果表明,由立体场、静电场、疏水场和氢键供体场4个场组合建立的构效关系模型具有较强的预测能力,交叉验证相关系数Q2=0.48, 非交叉验证相关系数Rncv2=0.94, 外部预测相关系数Rpre2=0.85;通过分子对接,发现抑制剂占据了靶标的S3、S1和S2'位点,与BACE1之间的结合主要是通过氢键作用力和π-π堆积作用实现的;占据S2'位点的R取代基是立体场、静电场和疏水场影响的敏感区域,氨基恶唑啉核心官能团是氢键供体场的敏感区域。基于以上分析获得的抑制剂特征结构信息及其与蛋白质受体的作用机制,成功设计出了新的分子并预测了抑制活性。实验所得模型和信息,为后续新型BACE1抑制剂的结构优化和改造提供了重要理论依据  相似文献

Chlorpromazine forms charge-transfer complexes with xanthene dyes in bacteria. These complexes permit the differentiation of Gram-positive and Gram-negative bacteria in both light and polarization microscopy. The birefringence induced by the charge-transfer complex might explain the molecular basis of bacterial staining.The charge-transfer complexes formed between chorpromazine and xanthene dyes accumulate in the bacterial cell, mainly inside the bacterial cell wall. The complexes give the cells a color, which depends on the chemical composition of the staining structure, and in particular the polysaccharides of the cell wall in bacteria.Metachromatic granules were seen inside Gram-positive bacteria after chlorpromazine and rose bengal staining. Although the nature of these granules remains unclear, this type of binding may have a role in the inhibition of biochemical processes in the bacterial cells.  相似文献

We have demonstrated the synthesis of regioisomerically pure unsymmetrical xanthene derivatives consisting of three units which can be independently modified to control their physical properties. The photochemical properties of the synthetic unsymmetrical xanthene derivatives were investigated in solution by UV–vis absorption and fluorescence measurements, and their cell imaging properties were examined by confocal laser-scanning microscopy.  相似文献

The use of photoactive substances for controlling adult or immature stages of insect pests is an attractive alternative to chemical insecticides. Phloxine B is an environmentally friendly xanthene derivative that is safe for mammals but toxic for dipterans. In this study we tested the effect of phloxine B as a phototoxic larvicide against immature stages of the blood-sucking horn fly, Haematobia irritans (L.). The mortality rate of phloxine B was very low in the dark during the larval stage (100 h) unless a 0.5-mM dye concentration was used. However, a high mortality rate was attained when larvae III were transferred to containers exposed to 5000 lux during the last 2 h before pupariation. This was concentration-dependent up to 0.1-mM phloxine B. After a 2-h larval exposure to light the phloxine B 50% lethal concentration was 0.043 mM. These results indicate that H . irritans larvae are very sensitive to this dye, which in turn seems a promising component for larvicide formulations to control horn flies.  相似文献

Summary Erythrosin and light flashes induce voltage transients across bilayer membranes in the presence of transmembrane pH gradients. Fast voltage transients, which rise in <50 nsec and fall in 500 nsec, are attributed to photo-deprotonation of dye sorbed in the glycerol region of phospholipid membranes. Six other halogenated xanthene dyes induce similar effects, which apparently resulted from triplet states of monoanionic dye. No photo-effects were observed with fluorescein.  相似文献

Breast cancer is the most diagnosed type of cancer among women for which an exhaustive cure has not been discovered yet. Nowadays, tamoxifen still represents the gold standard for breast cancer therapy; it acts on both estrogen receptor-positive and estrogen receptor-negative breast cancers. Unfortunately, its toxicity and the related chemoresistance undermine its antitumor potential. In this paper, new tamoxifen-based derivatives with a rigid structural motif in their structure were designed, synthesized, and evaluated to assess their antitumor behavior. All the tested compounds affected estrogen receptor-positive tumor (MCF-7) cell growth, even with different extents, among which, the most active ones proved also to induce mitochondria-mediated apoptosis through activation of PARP cleavage, decrease in Bax/Bcl-2 ratio and increase in Bim gene expression levels. Here we found that the compound 1, carrying a rigid xanthene core, turned out to be the most promising of the set showing an activity profile comparable to that of tamoxifen. Furthermore, a more favorable genotoxic profile than tamoxifen made compound 1 a promising candidate for further studies.  相似文献

The xanthene dyes, erythrosine, phloxine, and rose bengal, were adsorbed to charred cellulose granules. The charred cellulose granules were preliminarily steeped in ionic (NaOH, NaCl, KOH, KCl, and sodium dodecyl sulfate (SDS)), nonionic (glucose, sucrose, and ethanol), and amphipathic sucrose fatty acid ester (SFAE) solutions, and adsorption tests on the dye to the steeped and charred cellulose granules were conducted. Almost none of the dye was adsorbed when the solutions of ionic and amphipathic molecules were used, but were adsorbed in the case of steeping in the nonionic molecule solutions. Thin-layer chromatography (TLC) and the Fourier transform infra-red (FT-IR) profiles of SFAE which was adsorbed to the charred cellulose granules and extracted by ethyl ether suggested the presence of hydrophobic sites on the surface of the charred cellulose granules. We confirmed that the xanthene dyes could bind to the charred cellulose granules by ionic and hydrophobic bonds.  相似文献

The initial rate of horseradish peroxidase (HRP)-catalysed chemiluminescence (CL) reaction in an aqueous compartment of liposomes was applied to the estimation of membrane permeability of liposomes. HRP-encapsulated liposomes were prepared by an extrusion method, and a CL reagent and H(2)O(2) were added into the liposomes suspensions. Fluorescein, eosin Y and phloxin B, which are xanthene dyes with different chemical structures, were used as CL reagents. Xanthene dye and H(2)O(2) permeate into the inner phase of liposomes, resulting in initiation of the HRP-catalysed xanthene dye CL reaction with H(2)O(2). The initial rate of the CL reaction was independent of the xanthene dye used. The reproducibility of the initial rate with eosin Y was better than that with fluorescein and phloxin B. When the membrane permeability of the liposomes was changed by altering the concentration of cholesterol in them, the initial rate of the eosin Y CL reaction was dependent on the membrane permeability of the liposomes.  相似文献
