AIMS: Plant growth-promoting (PGP) activity of two Azospirillum strains and their effects on foliar and vascular bacterial diseases were evaluated on fresh market and cherry tomato. METHODS AND RESULTS: Tomato seeds were inoculated with A. brasilense Sp7 or Azospirillum sp. BNM-65. Four-week-old plants were challenge-inoculated with Clavibacter michiganensis subsp. michiganensis (bacterial canker) or with Xanthomonas campestris pv. vesicatoria (bacterial spot). Azospirillum-induced PGP was greater on cherry than on fresh-market tomato. Cherry tomato was more resistant to bacterial canker but more susceptible to bacterial spot than the fresh-market tomato. Canker severity was not affected by Azospirillum seed treatments. However, leaf- and plant-death were delayed on Azospirillum-treated plants compared with nontreated controls. Azospirillum increased the bacterial spot severity on cherry but not on fresh-market tomato. CONCLUSIONS: PGP was observed on both tomato genotypes, although growth effects were larger on cherry tomato. Also, Azospirillum treatments may alter tomato susceptibility to bacterial diseases. SIGNIFICANCE AND IMPACT OF THE STUDY: The interaction between PGP rhizobacteria like Azospirillum spp., not known to induce systemic resistance, with plant pathogens distantly located is frequently overlooked. This work demonstrates the importance of this kind of evaluation.  相似文献

Tomato fruits and seed lots were screened for the presence of Xanthomonas vesicatoria and Ralstonia solanacearum. Yellow colonies of Xanthomonas vesicatoria and white colonies of Ralstonia solanacearum were consistently isolated on yeast extract-dextrose-calcium carbonate agar medium (YDC) from diseased fruits and seed samples. This was confirmed by isolation on semi-selective medium such as Tween B for Xanthomonas and triphenyltetrazolium salt (TTC) medium for Ralstonia solanacearum followed by biochemical tests. The four isolates belonging to Xanthomonas vesicatoria and Ralstonia solanacearum were used to inoculate a local tomato variety. The isolates were found to cause yellowing and wilting of 2-weeks-old seedlings by 8–14 days after inoculation and by 4 weeks all plants had wilted and completely died. Bacteria with the same characteristics as those inoculated were reisolated from the infected plants. Uninoculated plants remained healthy.  相似文献

In our previous studies, we observed the biological control effect of lactic acid bacteria strains (LABs) KLF01, KLC02 and KPD03 against different plant pathogenic bacteria in vitro against Ralstonia solanacearum, and strains KLF01 and KLC02 against Pectobacterium carotovorum under greenhouse and field experiments, respectively. In this study, we observed the efficacy of these bacteria against bacterial spot pathogen (Xanthomonas campestris pv. vesicatoria) and their plant growth-promoting activities in pepper (Capsicum annuum L. var. annuum), under greenhouse and field conditions. LABs significantly (P < 0.05) reduced bacterial spot on pepper plants in comparison to untreated plants in both the greenhouse and the field experiments. The plant growth-promoting effect of LABs on pepper varied; some strains had a significant effect on growth promotion (P < 0.05) compared with untreated plants, while some showed no significant effect in the greenhouse and field experiments. Additionally, LABs were able to colonise roots, produce indole-3-acetic acid (IAA), siderophores and solubilise phosphate. These findings indicate that application of LABs could provide a promising alternative for the management of bacterial spot disease in pepper plants and could therefore be used as a healthy plant growth-promoting agent.  相似文献

The molecular events associated with susceptible plant responses to disease-causing organisms are not well understood. We have previously shown that ethylene-insensitive tomato plants infected with Xanthomonas campestris pv. vesicatoria have greatly reduced disease symptoms relative to wild-type cultivars. Here we show that salicylic acid (SA) is also an important component of the susceptible disease response. SA accumulates in infected wild-type tissues and is correlated with necrosis but does not accumulate in ethylene-insensitive plants. Exogenous feeding of SA to ethylene-deficient plants restores necrosis, indicating that reduced disease symptoms are associated with failure to accumulate SA. These results indicate a mechanism for co-ordination of phytohormone signals that together constitute a susceptible response to pathogens.  相似文献

To control defense and cell‐death signaling, plants contain an abundance of pathogen recognition receptors such as leucine‐rich repeat (LRR) proteins. Here we show that pepper (Capsicum annuum) LRR1 interacts with the pepper pathogenesis‐related (PR) protein 4b, PR4b, in yeast and in planta. PR4b is synthesized in the endoplasmic reticulum, interacts with LRR1 in the plasma membrane, and is secreted to the apoplast via the plasma membrane. Binding of PR4b to LRR1 requires the chitin‐binding domain of PR4b. Purified PR4b protein inhibits spore germination and mycelial growth of plant fungal pathogens. Transient expression of PR4b triggers hypersensitive cell death. This cell death is compromised by co‐expression of LRR1 as a negative regulator in Nicotiana benthamiana leaves. LRR1/PR4b silencing in pepper and PR4b over‐expression in Arabidopsis thaliana demonstrated that LRR1 and PR4b are necessary for defense responses to Pseudomonas syringae pv. tomato and Hyaloperonospora arabidopsidis (Hpa) infection. The mutant of the PR4b Arabidopsis ortholog, pr4, showed enhanced susceptibility to Hpa infection. Together, our results suggest that PR4b functions as a positive modulator of plant cell death and defense responses. However, the activity of PR4b is suppressed by interaction with LRR1.  相似文献

Xanthomonas campestris pv. vesicatoria type III‐secreted effectors were screened for candidates influencing plant cell processes relevant to the formation and maintenance of stromules in Nicotiana benthamiana lower leaf epidermis. Transient expression of XopL, a unique type of E3 ubiquitin ligase, led to a nearly complete elimination of stromules and the relocation of plastids to the nucleus. Further characterization of XopL revealed that the E3 ligase activity is essential for the two plastid phenotypes. In contrast to the XopL wild type, a mutant XopL lacking E3 ligase activity specifically localized to microtubules. Interestingly, mutant XopL‐labeled filaments frequently aligned with stromules, suggesting an important, yet unexplored, microtubule–stromule relationship. High time‐resolution movies confirmed that microtubules provide a scaffold for stromule movement and contribute to stromule shape. Taken together, this study has defined two populations of stromules: microtubule‐dependent stromules, which were found to move slower and persist longer, and microtubule‐independent stromules, which move faster and are transient. Our results provide the basis for a new model of stromule dynamics including interactions with both actin and microtubules.  相似文献
