Summary The amphibian antidiuretic hormone, arginine vasotocin, stimulated osmotic water flow across isolated skin from the pelvic but not the pectoral skin of the toad, Bufo woodhouseii. Changes in the apical membrane capacitance were not observed for either region of the skin following treatment with arginine vasotocin when there was an osmotic gradient across the tissue. In the absence of an osmotic pressure gradient, the apical membrane capacitance of the pelvic skin increased from 2.8±0.5 to 3.3±0.6 F · cm-2 after treatment with 5 · 10-8 M arginine vasotocin. Under these conditions, apical membrane capacitance of the pectoral skin was 1.8±0.1 F · cm-2 and did not change significantly after arginine vasotocin treatment. The amiloride-sensitive short-circuit current across the pelvic skin was stimulated by arginine vasotocin as was the density of channels in the apical membrane as determined by fluctuation analysis. Values for channel density in the pelvic skin also correlated with apical membrane capacitance and increased from 90 to 273 channels per m2 of estimated membrane area following arginine vasotocin treatment. In the pectoral skin the stimulation of short-circuit current following arginine vasotocin treatment was small and an increase in channel density could not be demonstrated. Summary Renal clearance experiments were conducted on feral chickens descended from birds collected from a coral island off the coast of Queensland, Australia. Following a control period when 0.13 M NaCl was used as a vehicle for the renal function markers, a salt load was imposed by infusion of 1 M NaCl. The hypertonic NaCl infusion resulted in increases in glomerular filtration rate (GFR), effective renal blood flow (ERBF), and urine flow rate (V), whereas filtration fraction decreased. Haematocrit was reduced and plasma osmolality, sodium, chloride and potassium concentrations increased. Plasma arginine vasotocin (PAVT) levels increased from 31.4±2.3 pg·ml-1 during the control infusion to 56.0±1.7 pg·ml-1 following a salt load of 12 mmol Nacl·kg-1 The sensitivity of release of AVT was 0.69±0.11 pg·ml-1 per mosmol·kg-1. The concentrations of Na and Cl in urine increased, whereas urine osmolality and potassium concentration decreased. The expansion of the extracellular fluid volume induced by the salt loading would tend to suppress the release of AVT, whereas the osmotic stimulus would provide a stimulus for the release of AVT. Simultaneous measurements of cardio-respiratory variables, oxygen uptake and whole body urea/ammonia/tritiated water effluxes were performed on cannulated gulf toadfish, Opsanus beta, before and after intra-arterial injection of the vasoactive agents, adrenaline, isoproterenol and arginine vasotocin. These experiments were conducted to test the hypothesis that the phenomenon of pulsatile urea excretion might reflect sudden changes in the general diffusive properties of the gill for solute transfer. Injection of isoproterenol (final nominal circulating level = 10−6 mol l−1), was used as a tool to maximise the diffusive and perfusive conditions for branchial solute transfer. This protocol caused a pronounced reduction in arterial blood pressure, an elevation of cardiac frequency and associated increases in whole body urea and tritiated water effluxes; ammonia excretion and oxygen uptake were unaffected. Injection of adrenaline (final nominal circulating level=10−6 mol l−1), caused a significant increase in arterial blood pressure and a tachycardia, yet nitrogen excretion and oxygen uptake were unaffected. Injection of arginine vasotocin, caused a dose-dependent (final nominal circulating levels = 10−11–10−9 mol l−1) increase in arterial blood pressure without affecting cardiac or ventilation frequency. At the two higher concentrations, arginine vasotocin caused large and transient increases in urea excretion without significantly affecting ammonia, water or oxygen fluxes. These results suggest that increased gill diffusive or perfusive conductance, while capable of augmenting urea efflux, cannot fully explain the sudden and massive increases in urea transfer associated with pulsatile urea excretion in toadfish. It is suggested that pulsatile urea excretion in this species may reflect a specific enhancement of urea excretion under the control of the neurohypophyseal hormone, arginine vasotocin. Based on a wide variety of data, it is now clear that birds and teleost (bony) fish possess a core "social behavior network" within the basal forebrain and midbrain that is homologous to the social behavior network of mammals. The nodes of this network are reciprocally connected, contain receptors for sex steroid hormones, and are involved in multiple forms of social behavior. Other hodological features and neuropeptide distributions are likewise very similar across taxa. This evolutionary conservation represents a boon for experiments on phenotypic behavioral variation, as the extraordinary social diversity of teleost fish and songbirds can now be used to generate broadly relevant insights into issues of brain function that are not particularly tractable in other vertebrate groups. Two such lines of research are presented here, each of which addresses functional variation within the network as it relates to divergent patterns of social behavior. In the first set of experiments, we have used a sexually polymorphic fish to demonstrate that natural selection can operate independently on hypothalamic neuroendocrine functions that are relevant for (1) gonadal regulation and (2) sex-typical behavioral modulation. In the second set of experiments, we have exploited the diversity of avian social organizations and ecologies to isolate species-typical group size as a quasi-independent variable. These experiments have shown that specific areas and peptidergic components of the social behavior network possess functional properties that evolve in parallel with divergence and convergence in sociality.  相似文献

Non-peptide antagonists of the oxytocin receptor (OTR) have been developed to prevent pre-term labour. The benzoxazinone-based antagonists L-371,257 and L-372,662 display pronounced species-dependent pharmacology with respect to selectivity for the OTR over the V(1a) vasopressin receptor. Examination of receptor sequences from different species identified Ala(318) in helix 7 of the human OTR as a candidate discriminator required for high affinity binding. The mutant receptor [A318G]OTR was engineered and characterised using ligands representing many different chemical classes. Of all the ligands investigated, only the benzoxazinone-based antagonists had decreased affinity for [A318G]OTR. Molecular modelling revealed that Ala(318) provides a direct hydrophobic contact with a methoxy group of L-371,257 and L-372,662.  相似文献

In a first set of experiments, intracerebroventricular (ICV) treatment of 1 μl 100 g−1 body mass of Cortland saline containing different doses (1–20 nmol μl−1) of arginine vasotocin (AVT) produced after 180 min dose‐dependent changes in levels of brain neurotransmitters in several brain regions and pituitary of rainbow trout Oncorhynchus mykiss . Thus, an enhancement of serotonergic and dopaminergic activity, together with a decreased noradrenergic activity, were observed both in the hypothalamus and pituitary of AVT‐treated fish. In the other brain regions assessed, only increased serotonergic activity in the optic lobes, and decreased dopaminergic activity in the telencephalon of AVT‐treated fish were noticed. Changes observed in monoamine levels resemble those observed during osmotic adaptation of euryhaline fishes. In a second set of experiments, fish were ICV injected with AVT as described above to assess changes in several variables of brain energy metabolism. The results obtained show a dose‐dependent enhancement of brain glycogenolytic potential in the brain of AVT‐treated fish, that again resemble the changes observed in euryhaline fishes during osmotic acclimation.  相似文献

The avian arginine vasotocin (AVT) synthesized in the hypothalamic magnocellular neurons and released from the posterior pituitary is known to be involved in the regulation of uterine contractions for oviposition in chickens. However, regulation of AVT synthesis and release within the magnocellular hypothalamus has not been elucidated. Galanin, the oviposition inducing factor in the oviduct of the hen, has been demonstrated to have sexually dimorphic stimulatory action in oxytocin- and vasopressin neurons in the mammalian hypothalamus. In this study, galanin and AVT immunoreactivity was investigated around the time of oviposition in the supraoptic nucleus (SON) to determine if galanin modulates AVT synthesis and/or release. Within SON neurons increased AVT immunoreactivity before oviposition and the decreased AVT immunoreactivity after oviposition implied function-related peptide release. The significantly increased number of galanin neurons co-localizing with AVT immediately after oviposition suggests that galanin is involved in the negative feedback to limit AVT release in the SON. Thus, these data support the idea that AVT in the SON is involved in central regulation of oviposition and that AVT release could be modulated by the neuropeptide galanin.  相似文献
