Abstract.The stimuli and mechanisms mediating host location and host choice by the bee mite, Varroa jacobsoni (Oudemans), are currently unknown. It is shown that Varroa can use single clean‐air puffs and bee‐odour plumes in a wind tunnel as directional cues. Varroa turned nearly straight upwind in response to single 0.1‐s puffs of clean air directed at 90° to the their anterior‐posterior axis. They turned significantly further to their left side (104°) than to their right (76°), but showed no difference in latency to initiation of the turns (means of 63.3 ms vs. 62.6 ms, respectively). They also followed bee‐odour plumes in a wind tunnel. When released in odour and control plumes mid‐way between the plume's origin and the downwind end of the tunnel, mites responding to bee‐odour walked upwind in, or along the edge of, the odour plume with 38% making contact with the odour delivery tube; mites in clean air did not walk upwind along the air stream, and none made contact with the air delivery tube. Walking speeds were not different between the bee‐odour and control groups (0.28 vs. 0.29 cm s–1); there were also no differences in the turning rates (96.85 vs. 97.16 deg s–1 and 388.08 vs. 379.18 deg cm–1, respectively). Electroantennogram (EAG) measurement of male Cydia caryana moth antennal olfactory response to monounsaturated 12 and 14 carbon alcohols and acetates indicated that the (E)-8-, (E)-10- conjugated double bond system of a dodecadien-1-ol acetate is a critical chemical structural component of the C. caryana sex pheromone. Additionally, EAG measurements implicated (E)-8-dodecen-1-ol acetate, (Z)-8-dodecen-1-ol acetate, (Z)-9-dodecen-1-ol acetate and (Z)-12-tetradecen-1-ol as potential minor pheromonal components. An EAG dosage-response study suggested that there were at least two heterologous populations of pheromone acceptors. Behavioral analysis of male moth response in a flight tunnel to compounds which evoked the stronger EAG responses suggested that (E,E)-8,10-dodecadien-1-ol acetate and (Z)-9-dodecen-1-ol acetate resemble or are C. caryana sex pheromonal components, while (Z)-8-dodecen-1-ol acetate and (E)-10-dodecen-1-ol acetate are either parapheromones or are minor pheromone components. Behavioral significance of (Z)-12-tetradecen-1-ol was difficult to interpret in the flight tunnel.
Résumé Les réponses olfactives antennaires de Cydia caryana, mesurées par électroantennogrammes (EAG), aux alcools et acétates à carbones monounsaturés en positions 12 et 14, ont montré que le système conjugué de double liaison, (E)-8-, (E)-10- du dodecadien-1-ol acétate constitue un composé chimique strutural critique de la phéromone sexuelle de C. caryana.De plus, les acétates: (E)-8-dodecen-1-ol,(Z)-8-dodecen-1-ol,(Z)-9-dodecen-1-ol, et le (Z)-12-tetradecen-1-ol, se sont révélés en AEG comme des composés secondaires de la phéromone. L'étude par AEG de la relation dose-réponse a conduit à l'hypothèse de deux catégories de populations de récepteurs de phéromones. L'analyse comportementale des résponses des papillons mâles dans le tunnel de vol aux composés qui ont provoqués les plus forts AEG, on fait estimer que les acétates (E,E)-8,10-dodécadien-1-ol et (Z)-9-dodecen-1-ol ressemblent (ou sont) les constituants de la phéromone sexuelle de C. caryana; tandis que les (Z)-8-dodecen-1-ol et (E)-10-dodecen-1-ol sont, soit des paraphéromones, soit des constituants mineurs de la phéromone.La signification biologique du (Z)-12-tétradécen-1-ol a été difficile à interprêter avec les expériences en tunnel de vol.

One reason to measure cross-sectional structural properties of primate long bones is to define mechanically relevant complexes of traits that describe the adaptation of bone to different biomechanical environments. This can be effectively accomplished when congeneric species having different postural and locomotor behaviors are compared. This paper compares the cross-sectional geometry of the femur and humerus in three behaviorally different macaque species as a basis for defining such patterns. Cross-sectional moments of inertia in the standard anatomical planes were calculated at five locations along the diaphyses of the femur and humerus in Macaca fascicularis, M. nemestrina, and M. mulatta. The data suggest that the "barrel-shaped" femur is associated with behaviors for which long limbs and small body size are an asset. This may be associated with, but is not restricted to, leaping behaviors. The data also suggest that structural rigidity of the femur and humerus is greater per unit body weight in primates that spend significant amounts of time in terrestrial environments than in those that are more restricted to climbing in arboreal environments.  相似文献

Six compounds were identified from gland extracts of the cotton bollworm, Heliothis armigera(Hubner): (Z)-11-hexadecenal (Z11-16:Ald), (Z)-9-hexa-decenal (Z9-16:Ald), hexadecanal, (Z)-11-hexadecenol (Z11-16:OH), (Z)-7-hexadecenal (Z7-16:Ald), and (Z)-9-tetradecenal (Z9-14:Ald). Each of the compounds that were identified was examined for its ability to elicit sexual responses from male moths in a flight tunnel. Males flew upwind to Z11-16:Ald alone, but greater levels of copulatory responses were evoked with the addition of 2.5% Z9-16:Ald to the Z11-16:Ald. Addition of hexadecanal to the binary mixture had no effect in raising the behavioral response of the males in the flight tunnel. The effect of Z7-16:Ald on male flight depended on the loading. The addition of 1% of this component to 2 mg of the binary mixture reduced levels of copulatory response, but the same addition (1 %) to 10 g of the binary mixture increased copulatory response. The addition of 79-14:Ald or Z11-16:OH to the binary mixture reduced behavioral responses of males. Proteinase K, the extracellular serine endopeptidase (E.C. 3.4.21.14) from the fungus Tritirachium album limber, is homologous to the bacterial subtilisin proteases. The binding geometry of the synthetic inhibitor carbobenzoxy-Ala-Phechloromethyl Ketone to the active site of proteinase K was the first determined from a Fourier synthesis based on synchrotron X-ray diffraction data between 1.8 Å and 5.0 Å resolution. The protein inhibitor complexes was refined by restrained least-squares minimization with the data between 10.0 and 1.8 Å. The final R factor was 19.1% and the model contained 2,018 protein atoms, 28 inhibitors atoms, 125 water molecules, and two Ca2+ ions. The peptides portion of the inhibitor is bound to the active center of proteinase K by means of a three-stranded antiparallel pleated sheet, with the side chain of the phenylalanine located in the P1 site. Model building studies, with lysine replacing phenylalanine in the inhibitor, explain the relatively unspecific catalytic activity of the enzyme.  相似文献

Summary We describe a simple method for determining the overall fold of a polypeptide chain from NOE-derived distance restraints. The method uses a reduced representation consisting of two particles per residue, and a force field containing pseudo-bond and pseudo-angle terms, an electrostatic term, but no van der Waals or hard shell repulsive terms. The method is fast and robust, requiring relatively few distance restraints to approximate the correct fold, and the correct mirror image is readily determined. The method is easily implemented using commercially available molecular modeling software.  相似文献

Quantum chemistry calculations of equilibrium geometry, atomic charges, dipole moment, and frontier orbital energies are carried out on model N-vinyl-2-pyrrolidone, its radical anion and cation, and three protonated derivatives. Attempts to obtain poly(N-vinyl-2-pyrrolidone) films grafted on nickel and platinum electrodes, respectively by cathodic and anodic polarizations, are reported. A thin, covering and insulating film is obtained on the Pt anode, as indicated by surface characterizations carried out with UPS and IRRAS spectroscopies. The fact that the film remains adherent to the metallic surface in spite of a permanent contact with an acetonitrile solution in which poly(N-vinyl-2-pyrrolidone) is soluble suggests that chemisorption has indeed been achieved.  相似文献

The three-dimensional solution-state structure is reported for the zinc-substituted form of rubredoxin (Rd) from the marine hyperthermophilic archaebacterium Pyrococcus furiosus, an organism that grows optimally at 100 degrees C. Structures were generated with DSPACE by a hybrid distance geometry (DG)-based simulated annealing (SA) approach that employed 403 nuclear Overhauser effect (NOE)-derived interproton distance restraints, including 67 interresidue, 124 sequential (i-j = 1), 75 medium-range (i-j = 2-5), and 137 long-range (i-j > 5) restraints. All lower interproton distance bounds were set at the sum of the van Der Waals radii (1.8 A), and upper bounds of 2.7 A, 3.3 A, and 5.0 A were employed to represent qualitatively observed strong, medium, and weak NOE cross peak intensities, respectively. Twenty-three backbone-backbone, six backbone-sulfur (Cys), two backbone-side chain, and two side chain-side chain hydrogen bond restraints were include for structure refinement, yielding a total of 436 nonbonded restraints, which averages to > 16 restraints per residue. A total of 10 structures generated from random atom positions and 30 structures generated by molecular replacement using the backbone coordinates of Clostridium pasteurianum Rd converged to a common conformation, with the average penalty (= sum of the square of the distance bounds violations; +/- standard deviation) of 0.024 +/- 0.003 A2 and a maximum total penalty of 0.035 A2. Superposition of the backbone atoms (C, C alpha, N) of residues A1-L51 for all 40 structures afforded an average pairwise root mean square (rms) deviation value (+/- SD) of 0.42 +/- 0.07 A. Superposition of all heavy atoms for residues A1-L51, including those of structurally undefined external side chains, afforded an average pairwise rms deviation of 0.72 +/- 0.08 A. Qualitative comparison of back-calculated and experimental two-dimensional NOESY spectra indicate that the DG/SA structures are consistent with the experimental spectra. The global folding of P. furiosus Zn(Rd) is remarkably similar to the folding observed by X-ray crystallography for native Rd from the mesophilic organism C. pasteurianum, with the average rms deviation value for backbone atoms of residues A1-L51 of P. furiosus Zn(Rd) superposed with respect to residues K2-V52 of C. pasteurianum Rd of 0.77 +/- 0.06 A. The conformations of aromatic residues that compose the hydrophobic cores of the two proteins are also similar. However, P. furiosus Rd contains several unique structural elements, including at least four additional hydrogen bonds and three potential electrostatic interactions.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

Non-invasive characterization of limb bone cross-sectional geometry would be useful for biomechanical analyses of skeletal collections. Computed tomography (CT) is potentially the method of choice. Additionally, CT images are suitable for automated analysis. CT is here shown to be both accurate and precise in the analysis of cross-sectional geometry of prehistoric femora. Beam hardening artifacts can be reduced by using a water bath. As the availability of CT for research increases, both bone density and geometry could be determined simultaneously with this method.  相似文献
