In this work, we analyzed the kinetics of anti-Trichinella spiralis newborn larva (NBL) antibodies (Ab) and the antigenic recognition pattern of NBL proteins and its dose effects. Wistar rats were infected with 0, 700, 2000, 4000 and 8000 muscle larvae (ML) and bled at different time intervals up to day 31 post infection (p.i.). Ab production was higher with 2000 ML dose and decreased with 8000, 4000 and 700 ML. Abs were not detected until day 10, peaked on day 14 for the 2000 ML dose and on day 19 for the other doses and thereafter declined slowly from 19 to 31 days p.i. In contrast, Abs to ML increased from day 10, peaked on day 19 and remained high until the end of the study. Abs bound strongly at least to three NBL components of 188, 205 and 49 kDa. NBL antigen of 188 and 205 kDa were recognized 10-26 days p.i. and that of 49 kDa from day 10 to day 31 p.i. A weak recognition towards antigens of 52, 54, 62 and 83 kDa was also observed during the infection. An early recognition of 31, 43, 45, 55, 68 and 85 kDa ML antigens was observed whereas the response to those of 43, 45, 48, 60, 64 and 97 kDa (described previously as TSL-1 antigens) occurred late in the infection. A follow-up of antigen recognition up to day 61 with the optimal immunization dose (2000 ML) evidenced a decline of Ab production to the 49 kDa NBL antigen 42 days p.i., which suggested antigenic differences with the previously reported 43 kDa ML antigen strongly recognized late in the infection. To analyze the stage-specificity of the 49 kDa NBL antigen, polyclonal antibodies (PoAb) were obtained in rats immunized with 49 kDa NBL antigen. PoAb reacted strongly with the 49 kDa NBL component in NBL total soluble extract but no reactivity was observed with soluble antigen of the other T. spiralis stages. Albeit with less intensity, the 49 kDa component was also recognized by PoAb together with other antigens of 53, 97 and 107 kDa, in NBL excretory-secretory products (NBL-ESP). Thus, our results reveal differences in the kinetics of anti-NBL and ML Ab responses. While anti-NBL Abs declined slowly from day 19 until the end of the experiment, Abs to ML antigen remained high in the same period. It is remarkable the optimal Ab response to NBL antigens with 2000 ML infective dose and the reduced number of NBL antigens identified throughout the experimental T. spiralis infection, standing out the immunodominant 49 kDa antigen. Interestingly, this antigen, which was prominently expressed in NBL somatic proteins, was also detected in NBL-ESP.  相似文献

Trichinellosis transmission to humans via the consumption of reptile meat is rare worldwide. In Korea, however, 2 such outbreaks, possibly via consumption of soft-shelled turtle meat, have occurred in 2 successive years. In 17 August 2014, 6 patients were admitted to Wonju Severance Christian Hospital complaining of myalgia, fever, and headache. Eosinophilia was the indication of the initial laboratory results, and they were eventually diagnosed as trichinellosis by ELISA. All of the patients worked at the same company and had eaten raw soft-shelled turtle meat at a company dinner 10 days prior to their admission. They were treated with albendazole for 2 weeks, upon which all of their symptoms disappeared. This is the 8th report on human trichinellosis in Korea, and the second implicating raw soft-shelled turtle meat.  相似文献

Trichinellosis is one of the most serious foodborne parasitic zoonoses in Europe. Wild carnivorous and omnivorous hosts are the main reservoirs of Trichinella spp. nematodes in nature. In the winter of 2008-2009, an atypical clinical case of trichinellosis occurred for the consumption of pork from a wild boar (Sus scrofa) hunted in southwestern Alps in Italy. The symptomatic individual showed delayed development of oedemas in the lower limbs and eosinophilia, which appeared three months after infection. Muscle samples harboured 3.8 larvae/g, which were identified as Trichinella britovi. During the epidemiological investigation, anti-Trichinella IgG were detected in five hunters.  相似文献

During an examination, for trichinellosis, of 1489 mammals of 12 different species, in the territory of Chukotka National district (regions of Anadyr', Bering, Iul'tin, Chaun and Chukotka) and in the region of North-Even of Magadan province, Trichinella larvae were detected in 236 (16·4%) of these animals from 6 of the species: in 109 (44·8%) of the blue polar foxes kept in cages, in 12 (15·8%) of the silver-black foxes kept in cages, in 84 (18·2%) of the white polar foxes, in 17 (17·5%) of the common foxes, in 13 (12·1%) of the dogs and in the one European bear examined. Trichinella larvae were not found in 190 marine mammals examined: 178 ringed seals, 7 whales, 4 walruses, and 1 bearded seal, nor in the 8 Siberian squirrels and 159 domestic pigs examined. The high incidence of Trichinella infection amongst blue polar foxes and silver-black foxes on fur-animal farms in Chukotka, where the food ration of these animals is predominately the flesh of marine mammals (walruses and other pinnipeds), suggests that marine mammals may constitute one of the sources of infection to caged animals. In Chukotka, as in other regions of the Extreme North and North-East, the circulation of trichinae in nature may occur between two ecologically different groups of animals: terrestial and marine mammals. A wide distribution of trichinellosis among animals in nature, or kept in cages and among dogs in Chukotka National district and in the North-Even region of Magadan province provides a constant threat of infection to man. Thus the flesh and sub-products of potential hosts of trichinae (including dogs, fur animals, European and polar bears, marine mammals, etc.) should undergo thorough trichinelloscopy examination before being consumed by man. The flesh of sea mammals should be examined for trichinellosis before being used as food for animals; the presence of trichinae in it requires its sterilization.  相似文献

Ahmad R. A. and Harpur R. P. 1982. Trichinellosis in the exercised rat. International Journal for Parasitology12: 59–63. Trichinella spiralis in Fischer (CDF/344) inbred rats was used to investigate the effect of exercise on a parasitic infection. Sixty male rats were trained for 2 months by running on a drum, a distance of 1.26 km day?1 in 60 min (4 days week?1) or 1.5 km in 90 min (5 days week?1). They were then given 0, 100 or 500, and 0,100 or 1000 infective Trichinella spiralis larvae respectively; the numbers of larvae encysted in the muscle of the rats 4 weeks later were determined. Trained animals had a significantly lower body weight which they maintained for 3 weeks after exercise ceased, but the training had no effect on the larval recovery. Likewise, exercise for 4 weeks after inoculation (i.e. during the chronic and invasive phases) had no effect. It is concluded that exercise does not modify the resistance of the rat to T. spiralis infection. It is difficult to separate the effects of exercise from the stress caused by exercise but because the rat-T. spiralis system is shown to be relatively insensitive to stress it is a good model with which to look for the effects of exercise per se.  相似文献
