Carbonic anhydrases (CAs) are a family of widely distributed metalloenzymes, involved in diverse physiological processes. These enzymes catalyse the reversible conversion of carbon dioxide to protons and bicarbonate. At least 19 genes encoding for CAs have been identified in the sea urchin genome, with one of these localized to the skeletogenic mesoderm (primary mesenchyme cells, PMCs). We investigated the effects of a specific inhibitor of CA, acetazolamide (AZ), on development of two sea urchin species with contrasting investment in skeleton production, Paracentrotus lividus and Heliocidaris tuberculata, to determine the role of CA on PMC differentiation, skeletogenesis and on non‐skeletogenic mesodermal (NSM) cells. Embryos were cultured in the presence of AZ from the blastula stage prior to skeleton formation and development to the larval stage was monitored. At the dose of 8 mmol/L AZ, 98% and 90% of P. lividus and H. tuberculata embryos lacked skeleton, respectively. Nevertheless, an almost normal PMC differentiation was indicated by the expression of msp130, a PMC‐specific marker. Strikingly, the AZ‐treated embryos also lacked the echinochrome pigment produced by the pigment cells, a subpopulation of NSM cells with immune activities within the larva. Conversely, all ectoderm and endoderm derivatives and other subpopulations of mesoderm developed normally. The inhibitory effects of AZ were completely reversed after removal of the inhibitor from the medium. Our data, together with new information concerning the involvement of CA on skeleton formation, provide evidence for the first time of a possible role of the CAs in larval immune pigment cells.  相似文献

Microtiter micromass cultures of limb-bud mesenchymal cells   总被引:4,自引:0,他引:4  
Summary A method is described for growing high-density micromass cultures of chick and mouse limb mesenchyme cells in 96-well microtiter plates (μTμM cultures). Rapid quantitative estimates of chondrogenic expression were obtained by automated spectrophotometric analysis of Alcian-blue-stained cartilage matrix extracts performed in the wells in which the cells had been grown. Quantitative estimates of myogenic expression were obtained similarly using anti-sarcomere myosin monoclonal antibody and modified ELISA techniques. This μTμM-ELISA method may be adapted for use with other antigens for which specific antibodies are available. These methods were used to compare cartilage and muscle differentiation in 1 to 4 d μTμM cultures grown in serum-containing (SCM) and defined (DM) media. The DM contains minimal additives (insulin, hydrocortisone, and in some cases, ascorbate or transferrin) and supports both chondrogenesis and myogenesis. The colorimetric analyses agree well with the morphologic appraisal of chondrogenesis and myogenesis. Similar numbers of cartilage nodules formed in all cultures, but in DM the nodules failed to enlarge; explaining the reduced matrix synthesis in DM as compared with SCM, and suggesting that nodule enlargement is a discrete, serum-dependent step. Studies of selected additives to DM show that transferrin enhances myogenesis, ascorbic acid enhances chondrogenesis, and retinoic acid inhibits chondrogenesis. Together, the μTμM system, in situ colorimetric assays of chondrogenesis and myogenesis, and DM will allow rapid prescreening of teratogens and screening of various bioactive compounds (e.g., hormones, growth factors, vitamins, adhesion factors) for effects on limb mesenchymal cell differentiation. Epithelial proliferation in the ventral surface of mouse tongue follows a pronounced circadian rhythm with a peak in mitotic activity at 10.00 a.m., preceded by a wave of DNA synthesis 8 h earlier. Nearly all cells (85%) pass through G2 and mitosis immediately after the S-phase; they subsequently divide again, usually after 2 or 3 days, indicating cohorts of cells with different G1-duration. The fraction of all nucleated cells comprised in one daily proliferation wave is about 20%, indicating a turnover time of the nucleated cell compartment of about 5 days. Cytotoxic injury by a single radiation dose of 20 Gy causes a steep decrease in cell counts, leading to complete denudation after 9–13 days. The difference between the latent period before ulceration and the tissue turnover time is explained by a marked proliferative activity of the doomed cells. The mitotic index increases steeply after day 1 to three times the control level, but most mitotic figures display gross abnormalities such as multipolar spindles or chromosome clumping. As a consequence cells with abnormal or multiple nuclei appear in the basal layers 3 days post irradiation and subsequently migrate to the upper layers. After denudation the epithelium rapidly becomes restored, with a phase of transient hyperplasia on days 13–14. Normal architecture is regained by day 15. Over the whole healing period the mitotic index remains at a high level, with most of the mitoses appearing histologically normal.  相似文献

Abstract. 1. A positive correlation between the tongue length of conspecific workers collecting nectar from seven plant species and the corolla length of the flowers probed was found for B.lapidarius and B.pascuomm but not B.terrestris . No simple relationship was found between the volume, sugar weight or concentration of nectar in flowers and the tongue or wing length of probing bees.
2. B.terrestris workers collecting pollen from four plant species producing pollen only, were found to differ in size according to the type of pollen presentation mechanism and the pollen content per flower. Body size variation was also related to the foraging of pollen plus nectar from two other plant species.  相似文献

Cogley T. P., Anderson J. R. and Cooley L. J. 1982. Migration of Gasterophilus intestinalis larvae (Diptera: Gasterophilidae) in the equine oral cavity. International Journal for Parasitology12: 473–480. Larvae of G. intestinalis pursued a specific migratory pathway within the equine oral cavity en route to the stomach. The larval migration included the following sequence: burrowing in the tongue mucosa, invasion of the interdental spaces, transitory attachment at the root of the tongue and movement to the stomach. The molt from first to second instar did not occur in the tongue, as commonly believed, but between the interdental spaces. Ninety five percent of the larvae invading the interdental spaces were associated with gingiva of the upper molars. SEM analysis revealed further details of the oral migration: (1) air holes excavated in the epithelium which connect with deeper burrows; (2) an intimate association between air holes and posterior spiracles of larvae; (3) precise impressions of larvae in tissue immediately surrounding the most recently formed burrows; and (4) initial larval entry into the tongue through the use of natural disruptions or healing lesions. Factors influencing the development of the oral migration are discussed.  相似文献

The surface structures of the tongues of Dissostichus mawsoni, Trematomus bemacchii, f. borchgrevinki (Nototheniidae), Stemoptyx diaphana (Sternoptychidae), Diretmus sp. (Diretmidae), Cataetyx memorabilis (Ophidiidae), Photoblepharon palpebratus (Anomalopidae), Carapus mourlani (Carapidae) and Salmo gairdneri (Salmonidae) were investigated with the scanning electron microscope. The aim of the study was to test the theory that the degree to which gustatory receptors are developed morphologically in different species of fishes is related to the environment, and in particular to the variey of food present in the latter. It was found that in the two mesopelagic species, S. diaphana and Diretmus sp., taste receptors were poorly developed, but in the benthic deep-water fish C. memorabilii , which lives off South West Africa in a biologically rich environment, well-developed taste receptors occurred in large numbers. The three Antarctic species studied had similar tongue surface structures with rather well developed papillae of a moderate density. Photoblepharon palpebratus , in spite of its small tongue, possessed a large number of taste buds on elevated ridges; Carapus mourlani , however, had only very few obvious receptors and is thought to be a poor taster. Undoubtedly the highest degree of taste receptor development is found in the freshwater trout Salmo gairdneri. The results support the hypothesis that taste receptor development and the variety of food types preseni in a fish's environment are correlated.  相似文献
