Mucor rouxianus produced two forms (isoenzymes) of glucoamylase which could be separated from each other by polyacrylamide gel electrophoresis or by chromatography on SP-Sephadex C-50, and they were designated glucoamylase I and glucoamylase II. Glucoamylases I and II were isolated in crystalline form, and were homogeneous in poly acrylamide gel electrophoresis and in ultracentrifugation, respectively. The sedimentation coefficient () molecular weight of glucoamylase I were 4.39 S and 59,000, and those of glucoamylase II were 4.29 S and 49,000, respectively.  相似文献

Membrane clarification of green tea extract was studied as a treatment to reduce sediments in packaged drinks and as a pretreatment for concentration processes. The flux and variation of components were examined in dead-end and crossflow filtration with several types of membranes. In dead-end ultrafiltration, the flux reduction rate was small, although the initial flux was similar to the final flux in microfiltration. Prefiltration was effective in decreasing the reduction rate of flux. As the pore size of microfiltration membranes became smaller, the dry weight decreased gradually and the optical transmission at 660 nm increased. By ultrafiltration, 30–50% pectin, 3–11% catechins and, 7–20% caffeine were rejected. Crossflow filtration was effective in keeping the flux high. The ultrafiltration spiral membrane (pore size: 0.008 μm) was selected for repeated batch clarification of prefiltered green tea crude extract and showed reproducible performance.  相似文献

l-Sorbose metabolism in Pseudomonas aeruginosa IFO 3898 was studied. When the strain was cultivated in l-sorbose medium, l-idonic and 2-keto-l-gulonic acids were detected in the culture broth.

From the results on the metabolism of various sugars and sugar acids with the cell suspension and the metabolites accumulated, the following pathway was proposed for the l-sorbose metabolism in Ps. aeruginosa IFO 3898.

l-Sorbose → l-idose → l-idonic acid → 2-keto-l-gulonic acid.  相似文献

The activities of carbon metabolism enzymes were determined in cellular extracts of the moderately thermophilic, chemolithotrophic, acidophilic bacteriumSulfobacillus thermosulfidooxidans subsp.asporogenes, strain 41, grown either at an atmospheric content of CO2 in the gas phase (autotrophically, heterotrophically, or mixotrophically) or autotrophically at a CO2 content increased to 5–10%. Regardless of the growth conditions, all TCA cycle enzymes (except for 2-oxoglutarate dehydrogenase), one glyoxylate bypass enzyme (malate synthase), and some carboxylases (ribulose bisphosphate carboxylase, pyruvate carboxylase, and phosphoenolpyruvate carboxylase) were detected in the cell-free extracts of strain 411. During autotrophic cultivation of strains 41 and 1269, the increase in the CO2 content of the supplied air to 5–10% resulted in the activation of growth and iron oxidation, a 20–30% increase in the cellular content of protein, enhanced activity of the key TCA enzymes (citrate synthase and aconitase), and, in strain 41, a decrease in the activity of carboxylases.  相似文献

The composition of the ether lipids of a strain of Caldariella acidophila, with respect to the different numbers of cyclizations of the biphytanyl components, is shown to differ between the various complex lipid classes, but the degree of cyclization increases systematically with the growth temperature in the range 75–89°. The problem of distinguishing adaptive from phyletic features in archaebacterial lipids is considered.  相似文献
