A new design of reaction vessel for simultaneous manual solid-phase synthesis of multiple peptide analogs is described. Simultaneous use of four of these vessels attached to a single rotary mixer has been successfully applied to synthesis of two sets of four decapeptide amide analogs. Efficient coupling was indicated by chemical determination at the end of each synthesis cycle and overall final yields of between 78 and 84% were obtained. The products obtained were of a high quality, as assessed by high-performance liquid chromatography and amino acid analysis. This system allows expeditious synthesis of multiple peptide analogs for structure-function studies with economical use of efficiently ventilated laboratory space.  相似文献

Summary Industrial fermentation media are normally sterilized with steam to destroy the indigenous microbial population prior to inoculation with a specific microorganism. Because biological validation of each sterilization cycle is impractical, an overkill approach is commonly employed on the basis that alteration of heat-sensitive nutrients is less detrimental than survival of indigenous microbes. However, the heat destruction of microbes is known to be a probability function amenable to calculation. A computer has been programmed to calculate the on-line heat input asF 0 values during sterilization of media in stirred bioreactors. The accumulation ofF 0 values is then announced verbally to bioreactor operators by a communications controller with voice synthesizer.  相似文献

We have developed a small benchtop oligonucleotide synthesizer which allows the scientist to prepare, rapidly and economically, up to 24 oligonucleotides in one batch. We have shown that this instrument can be used for peptide synthesis, as well. The instrument is based on the centrifugation method for solid–liquid separation. Dedicated to the memory of Prof. Bruce Merrifield, who was the inspiration behind all of our efforts in building automatic synthesizers.  相似文献
