Transverse sections of immature and mature sugarcane internodes were investigated anatomically with white and fluorescence light microscopy. The pattern of lignification and suberization was tested histo-chemically. Lignification began in the xylem of vascular bundles and progressed through the sclerenchymatic bundle sheath into the storage parenchyma. Suberization began in parenchyma cells adjacent to vascular bundle sheaths and spread to the storage parenchyma and outer sheath cells. In mature internodes most of the storage parenchyma was lignified and suberized to a significant degree, except in portions of walls of isolated cells. The pattern of increasing lignification and suberization in maturing internodes more or less paralleled an increase of sucrose in stem tissue. In mature internodes having a high sucrose concentration, the vascular tissue was surrounded by thick-walled, lignified and suberized sclerenchyma cells. The apoplastic tracer dyes triso-dium 3-hydroxy-5,8,10-pyrenetrisulfonate (PTS) and amido black 10 B, fed into cut ends of the stalk, wereconfined to the vascular bundles in all internodes above the one that was cut — with no dye apparently in storage parenchyma tissue. Thus both structural and experimental evidence is consistent with vascular tissue being increasingly isolated from the storage parenchyma as maturation of the tissue proceeds. We conclude that in mature internodes the pathway for sugars from the phloem to the storage parenchyma is symplastic. The data suggest that an increasingly greater role for a symplastic pathway of sugar transfer occurs as the tissue undergoes lignification/suberization.  相似文献

The absorption of soil water by roots allows plants to maintain their water status. At the endodermis, water transport can be affected by initial formation of a Casparian strip and further deposition of suberin lamellas and regulated by the function of aquaporins. Four Casparian strip membrane domain protein‐like (CASPL; CASPL1B1, CASPL1B2, CASPL1D1, and CASPL1D2) were previously shown to interact with PIP2;1. The present work shows that CASPL1B1, CASPL1B2, and CASPL1D2 are exclusively expressed in suberized endodermal cells, suggesting a cell‐specific role in suberization and/or water transport regulation. When compared with wild‐type plants, and by contrast to caspl1b1*caspl1b2 double loss of function, caspl1d1*caspl1d2 double mutants showed, in some control or NaCl stress experiments and not upon abscisic acid (ABA) treatment, a weak enlargement of the continuous suberization zone. None of the mutants showed root hydraulic conductivity (Lpr) phenotype, whether in control, NaCl, or ABA treatment conditions. The data suggest a slight negative role for CASPL1D1 and CASPL1D2 in suberization under control or salt stress conditions, with no major impact on whole root transport functions. At the molecular level, CASPL1B1 was able to physically interact with PIP2;1 and potentially could influence the regulation of aquaporins by acting on their phosphorylated form.  相似文献

A comparison of peroxidase isoenzymes in skin, cortex and pith tissues of the potato tuber by thin-layer isoelectric focusing in Sephadex revealed major differences in the isoenzyme patterns. Wounding induced several-fold increases in the peroxidase activity which were correlated with the increased amounts of specific isoenzymes. The anodic and cathodic forms with high activity, normally present in large amounts in skin, were found to be preferentially synthesized in suberizing tissues, suggesting a functional role for peroxidase in the suberization process. Cycloheximide treatment prevented the rapid increase in the content and activity of these specific isoenzymes, which indicated that the increase in peroxidase is due to a de novo synthesis of the enzyme. Suberization is not inhibited by gamma irradiation at sprout-inhibiting dose levels.  相似文献

Root growth, development and frost resistance were examined in winter rye ( Secale cereale L. cv. Puma) plants grown under 6 combinations of temperature and photoperiod (20/16°C or 5/3°C, day/night; 8, 16- or 24-h days). Overall root system growth is influenced by the interaction of temperature and photoperiod. Maximum shoot growth occurs at a 24-h photoperiod in 20°C plants and at a 16-h photoperiod in 5°C plants, and is correlated in both treatments with a high root:shoot ratio. Frost resistance of rye roots is affected by short photoperiods in 2 ways. First, short photoperiod and low temperature delay production of new adventitious roots so that newly developing roots are not exposed to freezing temperatures. Second, short photoperiod alone can induce several degrees of frost tolerance in existing roots during the lag phase of growth. Low temperature alone does not decrease the rate of dry weight accumulation in rye root systems, but cold temperature does retard developmental processes within the roots. Rye roots grown at 5°C develop first order lateral roots, differentiate metaxylem vessels and suberize endodermal cell walls more slowly than roots grown at 20°C.  相似文献

BACKGROUND AND AIMS: Akagare and Akiochi are diseases of rice associated with sulfide toxicity. This study investigates the possibility that rice reacts to sulfide by producing impermeable barriers in roots. METHODS: Root systems of rice, Oryza sativa cv. Norin 36, were subjected to short-term exposure to 0.174 mm sulfide (5.6 ppm) in stagnant solution. Root growth was monitored; root permeability was investigated in terms of polarographic determinations of oxygen efflux from fine laterals and the apices of adventitious roots, water uptake, anatomy and permeability to Fe2+ using potassium ferricyanide. KEY RESULTS: Both types of root responded rapidly to the sulfide with immediate cessation of growth, decreased radial oxygen loss (ROL) to the rhizospheres and reduced water uptake. Profiles of ROL measured from apex to basal regions of adventitious roots indicated that more intense barriers to ROL than normal were formed around the apices. Absorption of Fe2+ appeared to be impeded in sulfide-treated roots. In adventitious roots, deposition of lipid material (suberisation) and thickenings of walls within the superficial cell layers were obvious within a week after lifting the treatment and could prevent the emergence of laterals and commonly result in their upward longitudinal growth within the cortex. Death of laterals sometimes occurred prior to emergence; emergent laterals eventually died. In adventitious roots, blockages formed within the vascular and aeration systems in response to the sulfide. CONCLUSIONS: In both adventitious and lateral roots, sulfide-induced cell wall suberization and thickening of the superficial layers were correlated with reduced permeability to O2, water and Fe2+. This study sheds light on some of the symptoms of diseases such as Akiochi. The results correlate with the authors' previous findings on the effects on roots of sulfide and lower organic acids in Phragmites and of acetic acid in rice.  相似文献

The effects of local soil water content (SWC) alone on the development of hydrophobic deposits and on diffusivity of root tissues were studied in primary roots of maize seedlings whose top regions were growing at a constant rate in damp soil. Corn primary roots were grown through 30 cm-profiles of soil with pre-set dry (8% SWC), or wet (23% SWC), middle layers, and moist (15% SWC) top and bottom layers. When the root tissues in the soil of variable SWC were maturing (approximately 17% of the total root length), the tips of the roots were in damp soil several centimeters away and growing at the rate characteristic of this environment. Histochemistry and fluorescence microscopy on root sections of similar age showed that in these roots local SWC had no effect on hypodermal and endodermal suberization: both had equally developed Casparian bands and suberized lamellae. However, local SWC did alter the hydrophobic deposits in the walls of the epidermis; they increase in dry soil. To study tissue diffusivity, root portions from dry or wet soil were divided in the transverse direction, and half was dipped in sulphorhodamine G (SRG), sectioned, and observed with fluorescence optics. The other half was studied with a cryo-scanning electron microscope to observe the contents of the cortical intercellular spaces. The rate of SRG diffusion in a portion of root from dry or wet soil was independent of local SWC but was positively correlated with numbers of fluid-filled intercellular spaces. The implications of these observations for the movements of ions through the root cortex are discussed.  相似文献
