Single crystals were grown from affinity-purified stinging nettle lectin and from its complex with the specific trisaccharide NNN″ -triacetylchitotriose by vapor diffusion at room temperature. The lectin crystallizes in space group P212121 with unit cell dimensions a = 54.3 (1) Å, b = 62.2 (1) Å, and c = 92.4 (2) Å, and diffracts to 3.0 Å resolution. The asymmetric unit contains three lectin monomers. The crystals of the lectin-trisaccharide complex have space group P212121 with cell constants a = 37.69 (4) Å, b = 48.97 (6) Å, and c = 57.32 (4) Å. These crystals diffract to at least 2.0 Å resolution and the asymmetric unit contains one lectin monomer. Nettle root is recommended for complaints associated with benign prostatic hyperplasia (BPH). We therefore conducted a comprehensive review of the literature to summarise the pharmacological and clinical effects of this plant material. Only a few components of the active principle have been identified and the mechanism of action is still unclear. It seems likely that sex hormone binding globulin (SHBG), aromatase, epidermal growth factor and prostate steroid membrane receptors are involved in the anti-prostatic effect, but less likely that 5alpha-reductase or androgen receptors are involved. Extract and a polysaccharide fraction were shown to exert anti-inflammatory activity. A proprietary methanolic nettle root extract and particular fractions inhibited cell proliferation. Isolated lectins (UDA) were shown to be promising immunomodulatory agents, having also anti-viral and fungistatic effects. However, despite these in vitro studies it is unclear whether the in-vitro or animal data are a surrogate for clinical effects. The clinical evidence of effectiveness for nettle root in the treatment of BPH is based on many open studies. A small number of randomised controlled studies indicate that a proprietary methanolic extract is effective in improving BPH complaints. However, the significance and magnitude of the effect remains to be established in further confirmatory studies before nettle root treatment may be accepted in the guidelines for BPH treatment. The risk for adverse events during nettle root treatment is very low, as is its toxicity. Pre-clinical safety data remain to be completed.  相似文献

Mountain gorillas (Gorilla beringei beringei) in Karisoke, Rwanda, feed on the stinging nettle Laportea alatipes by means of elaborate processing skills. Byrne [e.g. Philosophical Transactions of the Royal Society of London, Series B, Biological Sciences 358:529–536, 2003] has claimed that individuals acquire these skills by means of the so‐called program‐level imitation, in which the overall sequence of problem‐solving steps (not the precise actions) is reproduced. In this study we present western lowland gorillas (Gorilla gorilla gorilla) with highly similar nettles. Twelve gorillas in three different groups (including also one nettle‐naïve gorilla) used the same program‐level technique as wild mountain gorillas (with differences mainly on the action level). Chimpanzees, orangutans, and bonobos did not show these program‐level patterns, nor did the gorillas when presented with a plant similar in structural design but lacking stinging defenses. We conclude that although certain aspects (i.e. single actions) of this complex skill may be owing to social learning, at the program level gorilla nettle feeding derives mostly from genetic predispositions and individual learning of plant affordances. 芙新姬小蜂和潜蝇姬小蜂均为强卵育型(synovigenic)寄生蜂,成虫具有产卵寄生致死、寄主取食致死和寄主叮蛰直接致死3种寄主致死行为。为了明确成虫补充非寄主食物营养对2种姬小蜂控害潜力/控害行为的影响,本文比较研究了分别提供寄主食物和寄主食物+非寄主食物(10%葡萄糖)条件下的2种姬小蜂在初羽化前期(前6 d)内对美洲斑潜蝇幼虫的致死行为的影响。结果显示:在仅提供寄主食物时,芙新姬小蜂的寄生、取食和总体致死量均显著的高于潜蝇姬小蜂,但是直接致死能力却弱于后者;补充非寄主食物和仅提供寄主食物处理相比,两种寄生蜂的寄生量,取食量和总致死量均呈下降趋势。该研究显示:在初羽化前期,取食寄主食物的雌蜂比取食寄主食物+葡萄糖的雌蜂具有更强的致死能力。  相似文献

Roots of stinging nettle (Urtica dioica L.) were sampled at different sites around Basel (Switzerland) and examined under the microscope. They were completely devoid of mycorrhizal structures. Similarly, stinging nettle plants grown in the greenhouse in the presence of the arbuscular mycorrhizal fungusGlomus mosseae did not show any signs of mycorrhiza formation. Spread ofG. mosseae through the rhizosphere of stinging nettle plants was inhibited, and application of extracts of stinging nettle roots and rhizomes to hyphal tips ofG. mosseae reduced hyphal growth.Urtica dioica agglutinin, an antifungal protein present in the rhizomes of stinging nettle, inhibited hyphal growth in a similar way as the crude root extract. The possibility thatUrtica dioica agglutinin is at least partially responsible for the inability of stinging nettle to form the arbuscular mycorrhizal symbiosis withG. mosseae is discussed.  相似文献

Water extract of stinging nettle (Urtica dioica) has a growth stimulating effect on plants. This investigation elucidated effects of nettle water on uptake and transport of N, P and K. Tomato plants (Solanum lycopersicum L. cv. Dansk export) were grown in sand culture 6–8 weeks. Plants were supplied with nettle water and nutrient solution was used as a control medium. Uptake and transport of N, P and K+ were determined with isotopes (15N,32P and86Rb+ as a tracer for K+) and ion-selective electrodes and in exudation experiments. A 15% higher uptake of nitrogen (15N assay) was found after nettle water treatment compared with the nutrient solution control. The total amount of nitrogen was also higher in plants cultivated with nettle water. Transport of inorganic and organic nitrogen, measured in exudation experiments, was more than 50% higher for plants supplied with nettle water compared with plants supplied with nutrient solution. In contrast, nettle water had no effect on uptake, transport or total amount of phosphorus and potassium in the plants. Experiments in hydroculture showed that nettle water had a strong pH-elevating effect. Uptake of NH 4 + was strongly stimulated by nettle water compared with nutrient solution. By holding pH at a constant level during the uptake period for 6 h, the uptake of NH 4 + from nettle water was significantly lower when no adjustment of pH was made. Consequently a good deal of the NH 4 + uptake enhancement by nettle water could be explained by pH-stimulation. Assays with the uncoupler/inhibitor 2,4-dinitrophenol (DNP) and dichlorophenyl-dimethyl-urea (DCMU) showed that uptake of nitrogen from nettle water was less metabolically-linked than uptake from a corresponding nutrient solution. All together, nettle water seems to stimulate the uptake of nitrogen, but not phosphorus or potassium.  相似文献

The common nettle (Urtica dioica L.) is a perennial and cosmopolitan plant species and is known to be the source of food for a great diversity of insects. To understand the importance of the nettle in agro-ecosystems, a field experiment was carried out in an experimental farm at Gembloux (Belgium) to study the effect of nettle margin strips on aphid and aphidophagous populations in close field crops, namely wheat (Triticum aestivum L.), green pea (Pisum sativum L.) and rape (Brassicae napus L.). The aphids and related beneficial populations were weekly assessed, from March to August 2005, by visual observations in two plots per field crop. A higher abundance of aphidophagous beneficials was collected in nettle strips when compared to the field crops. Particularly, the presence of predatory anthocorids, mirids and green lacewings was observed on nettle only. Nevertheless, the most abundant aphid predatory family, the Coccinellidae, was distributed in both environments, in nettle strips and in crop fields. The field margin supported a significantly higher density ofHarmonia axyridis than the field crops. In contrast, the field crops, green pea particularly, supported a higher density of Coccinella septempunctata. The distribution of the aphidophagous species, mainly the ladybirds, was discussed in relation to the host plant and related aphid species and their potential effect on integrated pest management.  相似文献

Polyphenol oxidase (PPO) of nettle (Urtica dioica L.) was extracted and purified through (NH4)2SO4 precipitation, dialysis, and CM-Sephadex ion-exchange chromatography and was used for its characterization. The PPO showed activity to catechol, 4-methylcatechol, L-3,4-dihydroxyphenylalanine (L-DOPA), L-tyrosine, p-cresol, pyrogallol, catechin and trans-cinnamic acid. For each of these eight substrates, optimum conditions such as pH and temperature were determined and L-tyrosine was found to be one of the most suitable substrates. Optimum pH and temperature were found at pH 4.5 and 30°C respectively and Km and Vmax values were 7.90?×?10?4?M, and 11290?EU/mL for with L-tyrosine as substrate. The inhibitory effect of several inhibitors, L-cysteine chloride, sodium azide, sodium cyanide, benzoic acid, salicylic acid, L-ascorbic acid, glutathione, thiourea, sodium diethyl dithiocarbamate, β-mercaptoethanol and sodium metabisulfite were tested. The most effective was found to be sodium diethyl dithiocarbamate which acted as a competitive inhibitor with a Ki value of 1.79?×?10?9?M. In addition one isoenzyme of PPO was detected by native polacrylamide slab gel electrophoresis.  相似文献
