The surfaces of miracidia of Schistosoma mansoni were examined ultrastructurally during in vitro transformation to sporocysts. Before transformation, the surface was composed of ciliated epithelial plates (EP) that were set into a reticulum of narrow syncytial ridges (SR). The EP were attached to SR by extensive pleated septate junctions that had 18-24 strands of intramembrane particles (IMP) on the protoplasmic faces and complementary pits on the ectoplasmic faces. These junctions also appeared to separate the EP plasma membrane into apical and basolateral domains with a larger number of IMPs on the latter. Transformation was induced by placing the miracidia in salt containing medium which also halted ciliary beating. In 2-5 hr, the SR expanded until they formed a syncytium covering the parasite surface, while the EP retracted and rounded up. During this time, the EP and SR were held in contact with one another by the septate junctions which became progressively convoluted. Subsequently, the EP detached from the parasite. When transforming miracidia were returned to fresh water, the cilia resumed beating and the EP detached from the parasite surface and exposed the underlying basement membrane. Those EP that remained attached were connected only by septate junctions to the expanded SR. These studies demonstrate that the formation of the syncytium occurs gradually with contact maintained between EP and SR via the septate junctions. Further, the septate junctions are similar to occluding junctions in mammalian epithelia since they segregate the plasma membrane of the EP and they have an adhesive function.  相似文献

Eimeria ornata n. sp. is described from the feces of 6/16 (37.5%) ornate box turtles, Terrapene ornata ornata , in northcentral Texas. Endogenously sporulated oocysts are ellipsoid 17.9 × 15.7(16-21 × 14-18) μm, with a thin, single-layered wall; shape index 1.14 (1.0-1.3). A micropyle is absent but a polar granule was present in one third of the oocysts. An oocyst residuum was present, consisting of numerous small globules situated either in a distinct mass or scattered within the oocyst. The sporocysts are elongate, 11.1 × 5.4 (9-13 × 5-6) μm, with an indistinct Stieda body at 1 pole. A sporocyst residuum is present, consisting either as a compact mass or as scattered granules. The sporozoites are elongate, 9.5 × 2.0 (8-12 × 2) μm, in situ, with spherical anterior and posterior refractile bodies. The new species is distinguished from the similar Eimeria carri Ernst & Forrester, 1973, from eastern box turtles, T. Carolina , by slight differences in oocyst morphology and endogenous sporulation.  相似文献

SYNOPSIS. Oocysts of Eimeria nieschulzi from the laboratory rat, Rattus, norvegicus , were studied by scanning and transmission electron microscopy. Oocysts had a rough outer wall with apparent random depressions. The oocyst wall is composed of 2 layers: an osmiophilic outer layer consisting of a rough external and smooth internal surface, and a relatively thick, electron-lucent inner layer. The outer layer is composed of a dense, coarsely granular matrix. The inner layer consists of homogeneous fine granular material interspersed with coarse osmiophilic granules and contains one closely applied membrane on the outermost surface. Several raised lenticular areas are seen on the coarse outer surface of the inner layer. These layers are 102 (75–128) and 176 (135–204) nm thick, respectively.
The sporocyst wall is thin, consisting of 3 to 4 unit membranes, and measures 27 (18–34) nm thick.  相似文献

SYNOPSIS. Oocysts and endogenous stages of new species of Eimeria and Isospora from the house lizard, Gehyra mutilata, are described. The ellipsoid to subspherical 2-layered oocysts of E. cicaki averaged 24.0 × 21.0 μm. Polar granules are present. Micropyle and oocyst residuum are absent. Ellipsoid sporocysts average 12.2 × 9.0 μm. A sporocyst residuum is present, but the Stieda body is absent. Endogenous stages are in epithelial cells of the small intestine. The subspherical single-layered oocysts of I. thavari average 23.8 × 22.8 μm. The polar granule is present; micropyle and oocyst residuum are absent. Ellipsoid sporocysts average 12.8 × 9.4 μm. Stieda body and sporocyst residuum are present. There are endogenous stages in epithelial cells of the small intestine.  相似文献

Explants and swabs from the pericardium and mantle of three strains of Biomphalaria glabrata, two of them resistant to infection with Schistosoma mansoni, have yielded small amoebae, 3–5μm in diameter, in culture. These amoebae have been grown axenically through > 50 passages to date. The amoebae form cysts in dense cultures. When mixed with S. mansoni mother sporocysts in vitro, the amoebae adhere to and kill the trematodes within several hours. For 1–2 days thereafter, the amoebae proliferate rapidly at a generation time of about 5 hr, then return to normal growth. Sonically disrupted sporocysts also induce proliferation. Live sporocysts do not attract the amoebae or emit soluble substances which influence amoebal growth. Amoebae also adhered to and killed S. mansoni daughter sporocysts and cells derived from B. glabrata embryos; however, they did not harm S. mansoni cercariae or rediae of other trematode species. The proportion of mantle explants yielding amoebae was significantly higher (P<0.05) in one of the resistant snail strains than in the susceptible strain; however, whether amoebae contribute to snail resistance is unknown. Exposure of snails to S. mansoni miracidia did not influence the proportion of snails yielding amoebae.  相似文献

The outer covering of digenetic trematodes undergoes a number of changes in the course of the life cycle in adaptation to parasitism and to the host-seeking requirements of the various stages. Using Schistosoma mansoni as a model, I review morphological, physiological, and cytochemical features of the coverings of miracidia, sporocysts, cercariae, and adults. Such features should be used with caution in evaluating phylogenetic relationships of any flatworms since the environment is itself a powerful determinant.  相似文献

SYNOPSIS. Isopora gallicolumbae sp. n. is described from Beccari's ground dove, Gallicolumba beccarii Salvadori, in Papua New Guinea. The ellipsoidal oocysts average 16.0 × 20.0 μm. The oocyst wall is light green, single-layered (∼ 0.8 μm), and becomes distorted within 10 min in sugar solution. Micropyle and oocyst residuum are absent; one polar granule is present. Sporocysts are ovoid, 8.0 × 12.0 μm. with prominent conical Stieda body. Sporocyst residuum is a spherical mass (∼ 5.0 μm) of lighter and darker granules. This is the first species of Isospora reported from a host in the avian order Columbiformes.  相似文献

The oocyst wall is severed by means of mechanical injury or chemical agents. This study reports the percentage of in vitro sporocyst release following mechanical shaking in the presence of varying sizes of glass beads. Glass beads measured 0.5, 1, and 3 mm in diameter and were shaken with the oocysts for different times ranging from 5 sec to 5 min. Approximately 80% of sporocysts were released with 5 min of shaking in the presence of 3 mm glass beads, as well as 30 sec with 0.5 mm beads and 1 mm glass beads. The release of sporocysts of E. tenella was most efficient using 1 mm glass beads and treatment times of 30 sec to 1 min. Therefore, the use of 1 mm glass beads with 30 sec to 1 min of agitation is recommended in order to maximize sporocyst release and recovery and to improve the yield of viable sporozoites for use in biochemical, tissue culture, and immunological applications of coccidia.  相似文献
