Gluco-oligosaccharides were synthesized through the enzymatic condensation of D-glucose at high concentration using a commercial almond beta-glucosidase. The synthesis reactions were carried out with both free and immobilized enzyme, with or without sorbitol, an efficient depressor of water activity (a(w)) in the presence of different glucose concentrations. The yield and the composition of the gluco-oligosaccharides produced changed with the reaction mixture and the form of the enzyme used (free or immobilized). The use of 5 M glucose solution permitted only disaccharides to be obtained, whereas with a glucose concentration of 7.5 M glucose, di-, tri-, and tetrasaccharides were produced. A 7.5 M glucose solution used with 4.4 M sorbitol gave three times more disaccharides than the same solution without sorbitol. Moreover, the immobilized enzyme was much more active in synthesis. The synthesis yield (oligomers mg/mL . mg of enzyme) after immobilization was 573% compared to that of the free enzyme, when a 7.5 M glucose solution was tested. The effect of the addition of sorbitol on the activity and stability of enzymes was examined by monitoring transesterification reactions performed in organic media at various water activities (a(w) = 0.08 to 0.97). Lipases from Chromobacterium viscosum and Candida rugosa immobilized on celite, and chymotrypsin, free or immobilized on celite, were used. When the sorbitol-containing enzymes were employed, higher reaction rates and less hydrolysis were observed. Immobilization of chymotrypsin resulted in high activity and operational stability, while the nonimmobilized enzyme was stable only in the presence of sorbitol. The activity of all preparations diminished after washing them with pyridine to remove sorbitol. Furthermore, severe stability problems occurred in the preparations lacking sorbitol. Sorbitol treatment, even after removal of the sorbitol itself, improved the activity of nonimmobilized chymotrypsin relative to the washed control. On the other hand, washing to remove sorbitol had a negative effect on the activity of both coimmobilized lipase and coimmobilized chymotrypsin. Addition of a substrate analogue, N-acetyl-L-phenylalanine, to chymotrypsin yielded a preparation that exhibited higher activity than both the control and its sorbitol-containing counterpart. Differential scanning calorimetry measurements revealed that the chymotrypsin-sorbitol complex was stable against thermal denaturation, undergoing transition at a high temperature (89 degrees C). A ';null' activity variant phenotype for sorbitol dehydrogenase (SDH) was observed in C57BL/LiA mice and used to examine the genetics of this enzyme. Linkage studies of the locus ( Sdh-1 ) with non-agouti (a) and a biochemical Iocus encoding liver L-α-hydroxyacid oxidase ( Hao-1 ) demonstrated that it is coincident with or closely linked to the structural locus, previously localized on chromosome 2. Alcohol dehydrogenase (ADH) isozymes were also examined, since the liver A2 isozyme exhibited some activity as a sorbitol dehydrogenase on cellulose acetate zymograms. It is apparent that SDH activity is not ';essential' in this mouse strain.  相似文献

The locus for sorbitol dehydrogenase (SORD, E.C. 1.1.1.14) has been shown to segregate with hexosaminidase A and mannose phosphate isomerase in a series of human-Chinese hamster somatic cell hybrids. 本文探讨了甲醇流加控制、添加胰蛋白胨和甲醇/山梨醇共混诱导对重组毕赤酵母GS115/pPIC9K-HSA-GCSFm表达HSA-GCSFm的影响.结果显示:甲醇供应充足条件下HSA-GCSFm表达水平仅为37 mg·L-1,而甲醇添加受限条件下HSA-GCSFm表达水平可达到239mg·L-1;甲醇添加受限并添加胰蛋白胨条件下,HSA-GCSFm表达水平可以提高到266 mg·L-1;在此基础上,流加山梨醇作为辅助碳源,表达水平可大幅提高至424 mg·L-1.通过对各诱导条件下OUR、胞外蛋白酶及碳流分配进行分析后发现,将甲醇限制在低浓度同时添加胰蛋白胨与山梨醇,可以改善细胞代谢活性,增加细胞用于HSA-GCSFm合成的碳流分配量,降低胞外蛋白酶活性,从而提高了HSA-GCSFm的表达水平并缓解了HSA-GCSFm的降解.因此,该诱导工艺适于毕赤酵母高效表达HSA-GCSFm.  相似文献

Adventitious shoot regeneration from internodal explants of mature plants of Annona muricata L. was obtained on Nitsch media. Meristems were induced with sorbitol as the sole carbon source supplemented with 2 mg l–1 of benzylaminopurine and 0.5 mg l–1 naphthaleneacetic acid. Adventitious shoots were developed only when the explants were transferred onto growth regulator-free media containing sucrose, galactose, or glucose. A hypothesis is proposed for the involvement of sorbitol in the induction and development of de novo shoots from internodal explants of mature trees of A. muricata.  相似文献

Concentrations of D-glucose, D-fructose and D-sorbitol were quantified in porcine epididymal fluid by spectrofluorimetric assays and aldose reductase (AR) and sorbitol dehydrogenase (SDH) were located immunohistochemically in the epididymal epithelium. Glucose and fructose concentrations were low (<1 mM) and decreased in the cauda whereas sorbitol concentration (4-7 mM) was rather uniform along the duct. AR was luminally located on microvilli in the caput and corpus with less presence distally and was present in the lumen. SDH was present apically and basally in epithelial cells throughout the epididymis and in the lumen. The observations are consistent with diffusion of circulating glucose into the lumen, its conversion via AR to sorbitol which accumulates in the lumen and the action of SDH on sorbitol to produce fructose. Sperm metabolism of glucose and fructose may explain their lower concentrations in the cauda and sorbitol could be a metabolic substrate or osmolyte required for volume regulation.  相似文献
