Molecular biology of wound-inducible proteinase inhibitors in plants   总被引:4,自引:4,他引:0  
Abstract. The techniques of molecular biology are being employed to investigate at the gene level the systemically mediated, wound-induced accumulation of two defensive proteinase inhibitor proteins in plant leaves. These techniques have added a new dimension to biochemical and physiological studies already underway to understand the mechanism of induction by wounding. The acquisition of cDNAs from the RNAs coding for the two inhibitors facilitated studies of mRNA synthesis in leaves in response to wounding, and provided probes to obtain wound-inducible proteinase inhibitor genes from tomato ( Lycopersicon esculentum ) and potato (Solarium tuberosum) genomes. Successful transformations of tobacco plants with fused genes, containing the 5' and 3' regions of the inhibitor genes with the open reading frame of the chloramphenicol acelyltransferase ( cat ) gene, have provided a wound-inducible chloramphenicol acetyltransferase (CATase) activity with which to seek cis- and transacting elements that regulate wound-inducibility to help to understand the interaction of cytoplasmic and nuclear components of the intracellular communication systems that activate the proteinase inhibitor genes in response to wounding by insect pests.  相似文献

The MADS-box APETALA1 genes control plant transition to flowering and the floral morphogenesis proper. The experimental evidence of APETALA1 overexpression presumes that this class of genes can also directly affect time to flowering. We therefore cloned and compared homologs of APETALA1 class genes from potato (Solanum tuberosum cultivars adapted to long day conditions) and its wild relative Solanum demissum, a short-day subtropical species. The homologs isolated from these plants belong to the subclass FRUITFULL. The inconsiderable variations in the primary structure of these homologs cannot explain the diverse photoperiodic reactions of particular Solanum genotypes.  相似文献

本文根据15省30余个县的实地考察,和对采自25省的66份中国龙葵种籽进行种植观察,并与国外的有关研究资料对比分析,在国内首次提出;(1)中国龙葵复合种的四个种,从低纬度到高纬度(18.6-53.5°N),分别属于二倍体,四倍体和六倍体种;(2)从中国南方到北方,自然形成了二倍体,六倍体与二倍体交叉,六倍体与四倍体交叉三个地理分布区。  相似文献

糖苷生物碱(steroidal glycoalkaloids,SGAs)是一类存在于茄科和某些百合科植物的重要次生代谢物,与植物的抗逆性和产品品质有密切关系.茄啶半乳糖基转移酶(solanidine galactosyltransferases,SGT1)是SGAs合成代谢途径的末端关键酶之一,研究其编码基因的启动子序列对于SGAs生物合成代谢调控有重要的作用和意义.研究采用染色体步移技术(Genome walking),首次克隆到马铃薯Sgt1基因起始密码子上游2 183 bp的启动子序列,已注册到GenBank(注册号:KC759163).构建该启动子驱动融合报告基因gfp::gus的植物双元表达载体p1304Sgt1p,转化野生型烟草获得Sgt1p::gfp::gus转基因植株,通过GUS组织化学染色分析Sgt1p::gfp::gus转基因植株中Sgt1p启动子的活性.结果表明,gus基因在转基因烟草的根、茎和叶中均表达,在叶中Sgt1p启动子的活性低于CaMV35S启动子,而在根和茎中二者基本相同;光诱导结果显示,光照处理明显增强了Sgt1p::gfp::gus转基因烟草叶片中Sgt1p启动子的活性,表明庄薯3号马铃薯 Sgt1p启动子是一种光诱导型启动子.  相似文献

A potato crop multimodel assessment was conducted to quantify variation among models and evaluate responses to climate change. Nine modeling groups simulated agronomic and climatic responses at low‐input (Chinoli, Bolivia and Gisozi, Burundi)‐ and high‐input (Jyndevad, Denmark and Washington, United States) management sites. Two calibration stages were explored, partial (P1), where experimental dry matter data were not provided, and full (P2). The median model ensemble response outperformed any single model in terms of replicating observed yield across all locations. Uncertainty in simulated yield decreased from 38% to 20% between P1 and P2. Model uncertainty increased with interannual variability, and predictions for all agronomic variables were significantly different from one model to another (P < 0.001). Uncertainty averaged 15% higher for low‐ vs. high‐input sites, with larger differences observed for evapotranspiration (ET), nitrogen uptake, and water use efficiency as compared to dry matter. A minimum of five partial, or three full, calibrated models was required for an ensemble approach to keep variability below that of common field variation. Model variation was not influenced by change in carbon dioxide (C), but increased as much as 41% and 23% for yield and ET, respectively, as temperature (T) or rainfall (W) moved away from historical levels. Increases in T accounted for the highest amount of uncertainty, suggesting that methods and parameters for T sensitivity represent a considerable unknown among models. Using median model ensemble values, yield increased on average 6% per 100‐ppm C, declined 4.6% per °C, and declined 2% for every 10% decrease in rainfall (for nonirrigated sites). Differences in predictions due to model representation of light utilization were significant (P < 0.01). These are the first reported results quantifying uncertainty for tuber/root crops and suggest modeling assessments of climate change impact on potato may be improved using an ensemble approach.  相似文献

We investigated the effect of temperature during growth and development on the relationship between light-harvesting capacity, indicated by chlorophyll concentration, and light-utilization potential, indicated by light- and bicarbonate-saturated photosynthetic oxygen evolution, in Solanum tuberosum L. cv. Norland. Conal plantles were transplanted and grown at 20°C for 2 weeks before transfer to 12, 16, 20, 24 and 28°C for 6 weeks. After 4 weeks of the temperature treatments, leaf tissue fresh weights per area were one-third higher in plants grown at 12°C vs those grown at 28°C. Conversely, chlorophyll content per area in tissue grown at 12°C was less than one-half of that of tissue grown at 28°C at 4 weeks. Photosynthetic capacity measured at a common temperature of 20°C and expressed on a chlorophyll basis was inversely proportional to growth temperature. Leaf tissue from plants grown at 12°C for 4 weeks had photosynthetic rates that were 3-fold higher on a chlorophyll basis than comparable tissue from plants grown at 28°C. These results suggest that the relationship between light-harvesting capacity and light-utilization potential varies 3-fold in response to the growth temperatures examined. The role of this response in avoidance of photoinhibition is discussed.  相似文献

Glandular secreting trichomes of cultivated tomato (Solanum lycopersicum) and close relatives produce a variety of structurally diverse volatile and non‐volatile specialized (‘secondary’) metabolites, including terpenes, flavonoids and acyl sugars. A genetic screen is described here to profile leaf trichome and surface metabolite extracts of nearly isogenic chromosomal substitution lines covering the tomato genome. These lines contain specific regions of the Solanum pennellii LA0716 genome in an otherwise ‘wild‐type’ M82 tomato genetic background. Regions that have an impact on the total amount of extractable mono‐ and sesquiterpenes (IL2‐2) or only sesquiterpenes (IL10‐3) or specifically influence accumulation of the monoterpene α‐thujene (IL1‐3 and IL1‐4) were identified using GC‐MS. A rapid LC‐TOF‐MS method was developed and used to identify changes in non‐volatile metabolites through non‐targeted analysis. Metabolite profiles generated using this approach led to the discovery of introgression lines producing different acyl chain substitutions on acyl sugar metabolites (IL1‐3/1‐4 and IL8‐1/8‐1‐1), as well as two regions that influence the quantity of acyl sugars (IL5‐3 and IL11‐3). Chromosomal region 1‐1/1‐1‐3 was found to influence the types of glycoalkaloids that are detected in leaf surface extracts. These results show that direct chemical screening is a powerful way to characterize genetic diversity in trichome specialized metabolism.  相似文献

The role of same amendment on phytoremediating different level contaminated soils is seldom known. Soil pot culture experiment was used to compare the strengthening roles of cysteine (CY), EDTA, salicylic acid (Sa), and Tween 80 (TW) on hyperaccumulator Solanum nigrum L. phytoremediating higher level of single cadmium (Cd) or Benzo(a)pyrene (BAP) and their co-contaminated soils. Results showed that the Cd capacities (ug pot?1) in shoots of S. nigrum in the combined treatment T0.1EDTA+0.9CY were the highest for the 5 and 15 mg kg?1 Cd contaminated soils. When S. nigrum remediating co-contaminated soils with higher levels of Cd and BAP, that is, 5 mg kg?1 Cd + 1 mg kg?1 BAP and 15 mg kg?1 Cd + 2 mg kg?1 BAP, the treatment T0.9CY+0.9Sa+0.3TW showed the best enhancing remediation role. This results were different with co-contaminated soil with 0.771 mg kg?1 Cd + 0.024 mg kg?1 BAP. These results may tell us that the combine used of CY, SA, and TW were more useful for the contaminated soils with higher level of Cd and/or BAP. In the combined treatments of Sa+TW, CY was better than EDTA.  相似文献

A protocol is presented for the rapid induction of microtubers on micropropagated, layered potato shoots of Kennebec, Russet Burbank and Superior in medium devoid of growth regulators. Layered shoots microtuberized more rapidly and produced significantly larger microtubers compared with nodal cuttings. The addition of coumarin or (2-chloroethyl)-trimethylammonium chloride and benzyladenine to microtuberization medium, either had no effect or significantly reduced microtuber weight per shoots compared with medium containing only 80 g × 1-1 sucrose and minimally affected the number of microtubers per shoot. Increasing the incubation period from 28 to 56 days did not affect the number but significantly increased the weight of microtubers per shoot and substantially increased the proportion, up to 20%, of microtubers heavier than 1 gram.Abbreviations Ba benzyladenine - ccc (2-chloroethyl) trimethylammonium chloride - coumarin 2h-1-benzopyran-2-one - ga3 gibberellic acid  相似文献
