An improved 13C-density-labeling method was used to study cell wall synthesis in rapidly expanding, slowly expanding and recently mature
internodes of Nitella translucens var axillaris (A.Br.) R.D.W. As cells matured, the rate of wall synthesis slowed and the deposition of cellulose microfibrils changed from
a predominantly transverse direction in the primary wall of rapidly expanding internodes to a helicoidal array in the secondary
wall of mature internodes. The secondary wall was characterized by relatively higher rates of cellulose synthesis and lower
rates of pectin synthesis than the primary wall. The synthesis of xyloglucan also decreased markedly at the transition to
secondary wall synthesis, while the synthesis of mannose-rich hemicellulose increased. Even though structural differences
were striking between the primary and secondary walls of Nitella, compositional differences between the two types of wall were quantitative rather than qualitative.

To identify salt stress-responsive genes, we constructed a cDNA library with the salttolerant rice cultivar, Lansheng. About 15000 plasmids were extracted and dotted on filters with Biomeck 2000 HDRT system or by hand. Thirty genes were identified to display altered expression levels responding to 150 mmol/L NaCl. Among them eighteen genes were up-regulated and the remainders downregulated. Twenty-seven genes have their homologous genes in GenBank Databases. The expression of twelve genes was studied by Northern analysis. Based on the functions, these genes can be classified into five categories, including photosynthesis-related gene, transportrelated gene, metabolismrelated gene, stress-or resistancerelated gene and the others with various functions. The results showed that salt stress influenced many aspects of rice growth. Some of these genes may play important roles in plant salt tolerance. 相似文献

A microplate-microtubule array was observed in Anabaena sp. (B-378). This structure consists of an arched plate, about 8 nm thick, and various microtubules, 12 nm in diameter and 50 nm long, arranged in rows. The microtubules project at right angles from one side of the plate into the cytoplasm or towards the plasma membrane. Up to twelve microplate-microtubule arrays were observed in a single section of a cell.Microfilaments, about 2.8 nm in diameter and of undetermined length, were observed in four isolates of Anabaena. The microfilaments were always found in bundles, which varied in size, up to 0.63 m across and 0.91 long.Microtubules, 10 nm in diameter and about 150 nm in length, were observed associated with one facet of polyhedral bodies in 8 out of 20 isolates of Anabaena. The microtubules occurred in groups of up to 20 or more, and were always oriented with the long axis parallel to a facet of a polyhedral body. In cross section, the microtubules had an electron transparent lumen 5 nm wide and a wall 2.5 nm thick.These structures are compared to previously deseribed microtubules and microfilaments. 相似文献

Epitaxial growth is a potential production process for the new material graphene, where it is grown on silicon carbide (SiC) wafers at high temperatures. We provide first estimates of the life cycle cumulative energy demand, climate change, terrestrial acidification, and eco‐toxicity of this production. For this purpose, we applied prospective life cycle assessment (LCA) for three production scenarios (lab, pilot, and an industrial scenario), which reflect different production scales and technological maturity. The functional unit was one square centimeter of graphene. Results show that the three scenarios have similar impacts, which goes against previous studies that have suggested a decrease with larger production scale and technological maturity. The reason for this result is the dominance of electricity use in the SiC wafer production for all impacts (>99% in the worst case, >76% in the best case). Only when assuming thinner SiC wafers in the industrial scenario is there a reduction in impacts by around a factor of 10. A surface‐area–based comparison to the life cycle energy use of graphene produced by chemical vapor deposition showed that epitaxial graphene was considerably more energy intensive—approximately a factor of 1,000. We recommend producers of epitaxial graphene to investigate the feasibility of thinner SiC wafers and use electricity based on wind, solar, or hydropower. The main methodological recommendation from the study is to achieve a temporal robustness of LCA studies of emerging technologies, which includes the consideration of different background systems and differences in production scale and technological maturity. 相似文献

Redox potentials in microsites of the rhizosphere of flooded rice were continuously measured for several days. Close to the root tips redox potential markedly increased. The highest increase was measured in the rhizosphere of the tips of short lateral roots. Aerobic redox conditions were reached there, except in a very strongly reduced soil. Both the extension of the oxidation zone around the root tips and the maximum redox potential reached were influenced by the reducing capacity of the soil. The radius of the redox rhizosphere varied from less than 1 mm in a strongly reduced soil up to 4 mm in a weakly reduced one. The root-induced oxidation processes in the rhizosphere depended on the atmospheric oxygen supply to the roots. 相似文献

Recent studies have revealed that in higher eukaryotes, several ribosomal proteins are involved in some pathological events or developmental defects, indicating that ribosomal proteins perform unconventional functions other than protein biosynthesis. To obtain an insight into the novel roles of ribosomal proteins, we aimed to analyze the changes in proteome expression in ribosomal protein mutants by using Saccharomyces cerevisiae as a model system. We introduced the rpl35bΔ mutation into the 4159 green fluorescent protein (GFP)-tagged yeast strains by using the synthetic genetic array (SGA) method, and performed quantitative proteomic analysis by using a multilabel microplate reader and flow cytometer. We identified 22 upregulated and 20 downregulated proteins in the rpl35bΔ mutant. These proteins were primarily classified into the Gene Ontology (GO) categories of cellular biosynthetic process, translation, protein or nucleotide metabolic process, cell wall organization and biogenesis, and hyperosmotic response. We also investigated the correlation between the mRNA and protein levels of the identified proteins. Our results show that a ribosomal protein mutation can lead to perturbation in the expression of several proteins, including some other ribosomal proteins. Furthermore, our approach of combining a library of GFP-tagged yeast strains and the SGA method provides an effective and highly sensitive method for dynamic analysis of the effects of various mutations on proteome expression. 相似文献

The purpose of this study was to formulate a gelled self-emulsifying drug delivery system (SEDDS) containing ketoprofen as
an intermediate in the development of sustained release solid dosage form. Captex 200 (an oil), Tween 80 (a surfactant), and
Capmul MCM (a cosurfactant) were used to formulate SEDDS. Silicon dioxide was used as a gelling agent, which may aid in solidification
and retardation of drug release. Effect of concentrations of cosurfactant and gelling agent on emulsification process and
in vitro drug diffusion was studied using 32 factorial design. Multiple regression analysis data and response surfaces obtained showed that liquid crystal phase viscosity
increased significantly with increasing amount of silicon dioxide, which in turn caused an increase in average droplet size
of resultant emulsion and slower drug diffusion. Drug release from the formulation increased with increasing amount of cosurfactant. 相似文献

目的采用液相悬浮芯片系统同时测定实验兔圆小囊中IL-1β、IL-1R1、IL=8、IL-8RA和IL=15各基因的表达情况,并对该方法进行评价。方法利用Affymetrix的Panomics QuantiGene Plex2.0Assay中bDNA信号放大和多磁珠分析技术,来同时检测两种实验兔圆小囊中多重mRNA并定量。建立实验兔免疫相关白介素基因的液相悬浮芯片检测方法。结果可同时检测IL-1β、IL=1R1、IL-8、IL=8RA和IL-15各基因的含量,并发现WHBE兔IL-15基因的相对表达量显著高于JW兔(P〈0.05),IL-1R1基因的相对表达量显著高于JW兔(P〈0.01),IL-8RA基因在WHBE兔中的相对表达量也高于JW兔(P〈0.05)。结论建立了实验兔白介素基因的液相悬浮芯片检测方法,WHBE兔的IL-15、IL-1R1和IL-8RA基因表达量较高,可能与WHBE兔独特的免疫学特性有关。 相似文献
