Scytonemin, the yellow-brown pigment of cyanobacterial (blue-green algal) extracellular sheaths, was found in species thriving in habitats exposed to intense solar radiation. Scytonemin occurred predominantly in sheaths of the outermost parts or top layers of cyanobacterial mats, crusts, or colonies. Scytonemin appears to be a single compound identified in more than 30 species of cyanobacteria from cultures and natural populations. It is lipid soluble and has a prominent absorption maximum in the near-ultraviolet region of the spectrum (384 nm in acetone; ca. 370 nm in vivo) with a long tail extending to the infrared region. Microspectrophotometric measurements of the transmittance of pigmented sheaths and the quenching of ultraviolet excitation of phycocyanin fluorescence demonstrate that the pigment was effective in shielding the cells from incoming near-ultraviolet-blue radiation, but not from green or red light. High light intensity (between 99 and 250 μmol photon · m?2· S?1, depending on species) promoted the synthesis of scytonemin in cultures of cyanobacteria. In cultures, high light intensity caused reduction in the specific content of Chl a and phycobilins, increase in the ratio of total carotenoids to Chl a, and scytonemin increase. UV-A (320–400 nm) radiation was very effective in eliciting scytonemin synthesis. Scytonemin production was physiological and not due to a mere photochemical conversion. These results strongly suggest that scytonemin production constitutes an adaptive strategy of photoprotection against short-wavelength solar irradiance.  相似文献

Immunoelectron microscopic techniques were utilized to characterize the morphology of circumsporozoite protein-containing trails deposited on various substrates by gliding Plasmodium berghei and Plasmodium falciparum sporozoites. The basic components of the trails are beadlike particles, 25 to 90 nm in diameter, which are devoid of unit membrane and have an electronlucent center. Trails were captured on formvar-covered grids coated with anticircumsporozoite protein monoclonal antibodies and compared with trails produced on uncoated formvar; the results suggest that material containing circumsporozoite protein forms the matrix within which the particles are embedded. The trails exhibit morphological features similar to those displayed by circumsporozoite precipitation reactions; of note is the demonstration of sheaths of circumsporozoite protein-containing material that emanate from sporozoites prior to their gliding. The sheaths narrow into accumulations of electron-dense material, which eventually taper to form typical trails. The structural manifestation of sheaths and other morphological details of the formed trails enables us to correlate sporozoite behavior during trail formation with the motile actions of gliding sporozoites observed by light microscopy.  相似文献

Yeasts use different mechanisms to release ascospores of different lengths from bottle-shaped asci. Round to oval-shaped ascospores are enveloped in oxylipin-coated compressible sheaths, enabling ascospores to slide past each other when they reach the narrowing ascus neck. However, more elongated ascospores do not contain sheaths, but are linked by means of oxylipin-coated interlocked hooked ridges on the surfaces of neighboring ascospores, thereby keeping them aligned while they are pushed towards the ascus tip by turgor pressure. In this study, we found elongated, oxylipin-coated sheathed ascospores in Dipodascus geniculatus that are released effectively from bottle-shaped asci without alignment. This is possible because the ascus neck and opening have a diameter that is the same as the length of the ascospore, thus allowing the ascospores to turn sideways without blocking the ascus when they are released. We found that increased concentrations of acetylsalicylic acid inhibit both ascospore release and 3-hydroxy oxylipin production in this yeast, thereby implicating this oxylipin in sexual reproduction.  相似文献

Maize plants, grown in aerated solution cultures, were exposed, at different growth stages, to ACC (1-aminocyclopropane-1-carboxylic acid) applied through the roots for up to 9 d. Total uptake of ACC increased with seedling size. During ACC treatment, ethylene evolution, by the shoots, proceeded at an almost constant rate per unit fresh weight that was up to 40-fold faster than that of untreated plants. This stimulation extended several days beyond the period of ACC uptake. The effects on growth and development were assessed when plants were 50–52-d old. ACC application shortened certain stem internodes, leaf-sheaths and laminae. The location of these effects depended on the time of application. The greatest shortening was induced by application, at the 4-leaf stage (10 d-old), prior to elongation of the cone of the shoot apex. This is ascribed to effects on meristematic tissue, in addition to those on elongating cells. An unexpected response to ACC treatment, at the 4-leaf stage, was an increase of up to four leaf-bearing stem nodes compared to untreated plants. This resulted in a parallel elevation of the uppermost ear-bearing axillary shoot to higher nodal positions. The length of leaves high in the canopy (nodes 11–16) was promoted by treating seedlings with ACC. The only clear effect of the ACC treatments on emergent axillary shoots per se was a retardation of silk elongation.  相似文献

Implantation of pacemakers can be challenging in the context of dilated cardiac chambers and valvular regurgitation. We report a difficult case of single chamber pacemaker implantation in a patient with restrictrive cardiomyopathy resulting in grossly enlarged atria and severe tricuspid regurgitation. In this situation, use of a slittable guiding sheath, more typically used for coronary sinus lead implantation, greatly facilitated rapid and stable deployment of the right ventricular lead.  相似文献

It has been previously shown that soil sheaths cling tightly to some portions of all axile roots and cover all but the growing tips of the young roots of field-grown maize. These sheaths overlie immature regions of the roots which have intact epidermal cells with root hairs, and living, thus non-conducting, late metaxylem elements. Loss of the soil sheath in the proximal region coincides with the opening of these large metaxylem vessels. Now, total, and viable counts have been recorded of bacteria associated with the root surface and adhering soil of sheathed and bare regions. These showed some common features, in that populations of similar size were associated with the two root regions in plants beginning to flower. Each population included about the same numbers of bacteria that were viable on each of three selective media (nitrogen-free, Pseudomonas F or MacConkey). However, more spore-formers capable of growth on nitrogen-free media and more fluorescent bacteria were isolated from the sheathed regions. Actinomycetes were absent from sheathed but plentiful on bare regions.The high numbers of diverse types of bacteria associated with both root surfaces can be related to the previously demonstrated similarity in amounts of organic carbon released from each region. The proliferation of actionomycetes on the bare roots and their exclusion from sheathed roots may in part be due to the lower water status of the bare region, which is related to its greater axial conducting capacity. Thus the distribution of the two types of root surface within an individual root system has important implications for the choice of root and rhizosphere sampling techniques and for root bacterization work.  相似文献

Changes in the activity of oxalate oxidase (OxO) and of the concentrations of oxalate and H2O2 were investigated during the ageing of leaf sheaths of ryegrass (Lolium perenne L.) stubble. The accumulation of H2O2 during ageing coincides with the increases of both oxalate level and OxO activity. Western and Northern blot analyses using protein and RNA extracts of the different categories of leaf sheaths suggested that OxO gene expression, as well as Ca-oxalate synthesis, are crucial events of ageing for leaf sheaths. Immunocytochemistry experiments have revealed that OxO, which is an extracellular enzyme, is nearly always present in the parenchymatous cells surrounding the vascular bundles and in the cells of the lower epidermis. Overall, results suggest that in ryegrass that synthesizes both Ca-oxalate and OxO, the production of H2O2 and Ca2+ during ageing of stubble might be involved in the constitutive defences against pathogens, thus allowing the phloem mobilization of nutrient reserves from the leaf sheaths towards elongating leaf bases of ryegrass.  相似文献

The relative significance of the use of stored or currently absorbed C for the growth of leaves or roots of Lolium perenne L. after defoliation was assessed by steady-state labelling of atmospheric CO2. Leaf growth for the first two days after defoliation was to a large extent dependent on the use of C reserves. The basal part of the elongating leaves was mainly new tissue and 91% of the C in this part of the leaf was derived from reserves assimilated prior to defoliation. However, half of the sucrose in the growth zone was produced from photosynthesis by the emerged leaves. Fructans that were initially present in elongating leaf bases were hydrolysed (loss of 93 to 100%) and the resulting fructose was found in the new leaf bases, suggesting that this pool may be used to support cell division and elongation. Despite a negative C balance at the whole-plant level, fructans were synthesized from sucrose that was translocated to the new leaf bases. After a regrowth period of 28 d, 45% of the C fixed before defoliation was still present in the root and leaf tissue and only 1% was incorporated in entirely new tissue.  相似文献
