Application of salicylate increased the concentration of metallothionein (MT) in liver of pregnant rats as well as of adult male rats, whereas in fetal liver, MT was reduced by salicylate. Induction of MT synthesis by salicylate is an indirect effect because in cultured hepatocytes salicylate did not induce MT synthesis. Salicylate increased MT also in adrenalectomized rats. Indomethacin induced the same concentration of MT in maternal liver as salicylate. However, indomethacin had no effect on MT in fetal liver. Induction of MT in adult liver by salicylate and indomethacin was independent of zinc.  相似文献

Two different genes encoding class II chitinases from peanut (Arachis hypogaea L. cv. NC4), A.h.Chi2;1 and A.h.Chi2;2, have been cloned. In peanut cell suspension cultures, mRNA levels of A.h.Chi2;2 increased after ethylene or salicylate treatment and in the presence of conidia from Botrytis cinerea. The second gene, A.h.Chi2;1, was only expressed after treatment with the fungal spores. Transgenic tobacco plants containing the complete peanut A.h.Chi2;1 gene exhibited essentially the same expression pattern in leaves as observed in peanut cell cultures. Expression characteristics of transgenic tobacco carrying a promoter-GUS fusion of A.h.Chi2;1 are described.  相似文献

Transgenic tobacco plants that express the bacterial nahG gene encoding salicylate hydroxylase have been shown to accumulate very little salicylic acid and to be defective in their ability to induce systemic acquired resistance (SAR). In recent experiments using transgenic NahG tobacco and Arabidopsis plants, we have also demonstrated that salicylic acid plays a central role in both disease susceptibility and genetic resistance. In this paper, we further characterize tobacco plants that express the salicylate hydroxylase enzyme. We show that tobacco mosaic virus (TMV) inoculation of NahG tobacco leaves induces the accumulation of the nahG mRNA in the pathogen infected leaves, presumably due to enhanced stabilization of the bacterial mRNA. SAR-associated genes are expressed in the TMV-infected leaves, but this is localized to the area surrounding necrotic lesions. Localized acquired resistance (LAR) is not induced in the TMV-inoculated NahG plants suggesting that LAR, like SAR, is dependent on SA accumulation. When SA is applied to nahG-expressing leave's SAR gene expression does not result. We have confirmed earlier reports that the salicylate hydroxylase enzyme has a narrow substrate specificity and we find that catechol, the breakdown product of salicylic acid, neither induces acquired resistance nor prevents the SA-dependent induction of the SAR genes.  相似文献

Aqueous Cu2+ and Cu(II) complexes of salicylate, lysine, and tyrosine decrease the rate of benzylamine oxidation by bovine plasma amine oxidase. Bissalicylato Cu(II) and Cu2+ inhibit non-competitively with respect to benzylamine. Lysine, tyrosine, Cu(EDTA)2?, Zn2+, and Co2+ do not inhibit, and erythrocyte Cu, Zn superoxide dismutase shows only slight inhibition of the amine oxidase. The data are most consistent with an inhibitory mechanism involving dismutation of O2? by the Cu(II) complexes within a site relatively inaccessible to the enzyme superoxide dismutase. Excess lysine significantly decreases inhibition by the bis-lysine complex of Cu(II).  相似文献

In barley seedlings (Hordeum vulgare L.) during two days after irradiation of shoots with UV-B (0.5 W/m2, 6 h), the rate of elongation of primary roots decreased 2–3 times compared to that in control plants. The modulus of elasticity of roots (ε) increased at most twofold in 12 h after the onset of irradiation; the hydraulic conductivity (L p) diminished by a factor of two in 12 h, and the root osmotic pressure gradually decreased by 0.08 MPa in 24 h. Changes in ε and L p were shown to be related to oxidative stress in growing roots, which was evidenced from the increase in H2O2 level up to 15-fold increase in 6 h and in activity of guaiacol peroxidase (3.5-fold in 12 h). After 48 h, the characteristics of oxidative metabolism and root characteristics ε and L p became identical in untreated and treated plants. On the third day, the rate of root growth in treated plants reached its initial value. It is concluded that the main causes of retardation of root growth under these conditions were as follows: the increase in cell wall rigidity related to formation of oxidative cross-links in the apoplast and the decrease in root osmotic pressure due to limited transport of assimilates from irradiated leaves. After the intensity of UV-B irradiation applied to shoots was enhanced (1.6 W/m2, 4 h), another physiological status of roots was observed on the 2nd day characterized by twofold increase in L p, tenfold decreased root elongation rate, and by a progressing increase of root diameter in growing roots. The comparison of root responses induced by irradiation of shoots with the root responses to sodium salicylate and ABA suggests that both agents might participate in the transmission of signals from irradiated leaves to roots.  相似文献

Grasses very often form symbiotic associations with Neotyphodium/Epichloë endophytic fungi. These endophytes often allow the host grass to be protected from different pathogens. However, there is little known about the mechanisms of such endophyte influence on the host. Thus, the purpose of this research was to examine the effect of the N. lolii endophyte on the total production of phenolic compounds, VOCs emission and the resistance of three perennial ryegrass genotypes infected by pathogenic Fusarium poae. Analyses of total phenolics content were performed in control (not inoculated) and inoculated plants after 1, 2, 3, 4, 5, and 6 days (DAI) and for VOCs after 0, 3, 6 and 12 DAI. The presence of endophytes significantly reduced the disease index in two of the three genotypes relative to that in E−. Plants infected by N. lolii exhibited higher production of phenolics relative to the E− plants. The highest amounts of phenolics were observed on the second and sixth DAI. Genotype Nl22 showed the strongest effect of the endophyte on the production of phenolics, which increased by over 61%. Both the endophyte infected and non-infected plants emitted most abundantly two GLVs ((Z)-3-hexenal, (Z)-3-hexen-1-yl acetate), three terpenes (linalool, (Z)-ocimene, β-caryophyllene) and three shikimic acid pathway derivatives (benzyl acetate, indole, and methyl salicylate). The endophyte presence and the intervals of VOCs detection were a highly significant source of variation for all emitted volatiles (P < 0.001). The genotype of the perennial ryegrass significantly affected only the emission of methyl salicylate (P < 0.05) and β-caryophyllene (P < 0.05). Most of the VOCs ((Z)-3-hexen-1-yl acetate, (Z)-3-hexenal, linalool and methyl salicylate) reached their highest levels of emission on the sixth DAI, when averaged over genotypes and endophyte status. The results highlight the role of Neotyphodium spp. in the mediation of quadro-trophic interactions among plants, symbiotic endophytes, invertebrate herbivores and plant pathogenic fungi. Our results also confirm the fact that symbiotic plants can activate a defense reaction faster than non-symbiotic plants after a pathogen attack. Thus, N. lolii can be involved in the defense of perennial ryegrass against pathogens and potentially could be central to the host plants’ protection.  相似文献

Sodium alendronate is the first in a pharmacological class known as bisphosphonates, used for treatment of various bone diseases. Assay of bisphosphonates by a spectroscopic technique is very challenging due to the fact that they lack chromophores and none of them are fluorescent. In this work, a simple method is presented for determination of alendronate in bulk and in pharmaceutical tablets using spectrofluorometry by exploiting the ability of alendronate to displace salicylate from the iron(III)–salicylate chelate, forming a non‐fluorescent colorless iron(III)–alendronate complex. The liberated salicylate is fluorescent and is equivalent to the mount of alendronate added. The response was linear over the concentration range 20–90 μM and the proposed method was validated according to the guidelines of the International Conference on Harmonization. The correlation coefficient was found to be 0.995 and the limit of detection was 7.5 μM. The method was successfully applied for determination of alendronate in the commercially available Osteonate® tablets. The average percent recovery ± percent relative standard deviation was found to be 102.118 ± 2.033 which is congruent with the label claim of the dosage form. The results were also compared to a reported method using t‐test and F‐test at 95% confidence level; no significant differences were observed. The presented method is simple, fast, easy, cost‐effective and suitable for routine pharmaceutical analysis.  相似文献

It is known that many social insects and arthropod predators and parasitoids can learn the association between a resource and volatile cues. Although there are various studies on the effect of experience in immature arthropods on behavior later in adult life, not much is known about the effects of such experiences on immature behavior. This was investigated here in the lacewing Ceraeochrysa cubana (Hagen) (Neuroptera: Chrysopidae). Whereas adults of this lacewing feed on plant‐provided food and honeydew, larvae are voracious polyphagous predators of several insect pests, and therefore important for biological control. Hence, studying the foraging behavior and the effects of learning in immatures of this species is important. We exposed immatures to the volatile methyl salicylate (MeSA), which was either associated with food or with the absence of food. Subsequently, their response to this volatile was tested in an olfactometer. Immatures that had experienced the association of MeSA with food were attracted to it and immatures that were exposed to MeSA during food deprivation were repelled. Subsequently, predator immatures that had experienced the association between MeSA and food were released on a plant without food and were found to use this volatile in locating patches with food. In contrast, larvae without such experience were found equally on food patches with and without the volatile. We conclude that these immature predators are capable of learning the association between volatiles and food, or the absence of food, and use this during foraging.  相似文献

The pine weevil is one of the most important pest insects of conifer reforestation areas in Europe. Female pine weevils cover their eggs with chewed bark and feces (frass) resulting in avoidance behavior of feeding conspecifics towards egg laying sites. It has been suggested that microorganisms present in the frass may be responsible for producing deterrent compounds for the pine weevil. The fungi Ophiostoma canum, O. pluriannulatum, and yeast Debaryomyces hansenii were isolated from aseptically collected pine-weevil frass. The isolated fungi were cultured on weevil frass broth and their volatiles were collected by SPME and identified by GC–MS. D. hansenii produced methyl salicylate (MeS) as a major compound, whereas, in addition, O. canum and O. pluriannulatum produced 6-protoilludene. In a multi-choice lab bioassay, MeS strongly reduced pine weevil's attraction to the Pinus sylvestris volatiles. Thus, a fungal metabolite was found that strongly affects the pine weevil host-odor search.  相似文献
