Chronic kidney disease (CKD) is a global problem. Slowing CKD progression is a major health priority. Since CKD is characterized by complex derangements of homeostasis, integrative animal models are necessary to study development and progression of CKD. To study development of CKD and novel therapeutic interventions in CKD, we use the 5/6th nephrectomy ablation model, a well known experimental model of progressive renal disease, resembling several aspects of human CKD. The gross reduction in renal mass causes progressive glomerular and tubulo-interstitial injury, loss of remnant nephrons and development of systemic and glomerular hypertension. It is also associated with progressive intrarenal capillary loss, inflammation and glomerulosclerosis. Risk factors for CKD invariably impact on endothelial function. To mimic this, we combine removal of 5/6th of renal mass with nitric oxide (NO) depletion and a high salt diet. After arrival and acclimatization, animals receive a NO synthase inhibitor (NG-nitro-L-Arginine) (L-NNA) supplemented to drinking water (20 mg/L) for a period of 4 weeks, followed by right sided uninephrectomy. One week later, a subtotal nephrectomy (SNX) is performed on the left side. After SNX, animals are allowed to recover for two days followed by LNNA in drinking water (20 mg/L) for a further period of 4 weeks. A high salt diet (6%), supplemented in ground chow (see time line Figure 1), is continued throughout the experiment. Progression of renal failure is followed over time by measuring plasma urea, systolic blood pressure and proteinuria. By six weeks after SNX, renal failure has developed. Renal function is measured using ''gold standard'' inulin and para-amino hippuric acid (PAH) clearance technology. This model of CKD is characterized by a reduction in glomerular filtration rate (GFR) and effective renal plasma flow (ERPF), hypertension (systolic blood pressure>150 mmHg), proteinuria (> 50 mg/24 hr) and mild uremia (>10 mM). Histological features include tubulo-interstitial damage reflected by inflammation, tubular atrophy and fibrosis and focal glomerulosclerosis leading to massive reduction of healthy glomeruli within the remnant population (<10%). Follow-up until 12 weeks after SNX shows further progression of CKD.  相似文献

Aim To compare bird abundances in woodlands along gradients from the city centre to the peri‐urban area. To evaluate the importance of the proportion of woodland within the city and in the peri‐urban landscape to forest bird communities breeding in urban woodlands. To test whether fragmentation effects on birds were linked to the type of peri‐urban matrix. Location A total of 34 Swedish cities with > 10,000 inhabitants in south‐central Sweden. The study area covered 105,000 km2, in which 84% of the Swedish population of 9.1 million lives. Methods Repeated point count surveys were conducted in 2004 in a total of 474 woodlands. General linear models were used to test for possible differences in abundance along urban to peri‐urban gradients, and to regress bird abundances in local urban woodlands on: (1) total woodland cover in the city, (2) total woodland cover in the peri‐urban landscape, (3) the interaction between woodland cover in the city and in the peri‐urban area, (4) region, and (5) human density. Results More than 12,000 individuals of 100 forest bird species were recorded. Of the 34 most common species detected, 13 bird species had higher abundances in urban than in peri‐urban woodlands, and seven species showed the opposite trend. The bird community of urban woodlands was characterized by species associated with deciduous forests and tree nesters, whereas the bird community of peri‐urban woodlands was characterized by species associated with coniferous woodland and ground nesters. Twelve species were significantly linearly associated with the proportion of urban woodland and/or the proportion of peri‐urban woodland, and a further eight species were associated with the interaction between these two factors. Local breeding bird abundances of four species were significantly positively associated with the proportion of urban woodland only in farmland‐dominated landscapes. Main conclusions Fragmentation effects on some urban birds are linked to the type of peri‐urban matrix. In farmland landscapes, peri‐urban woodlands may have been too scarce to act as a source of bird immigrants to fragmented urban woodlands. To maintain populations of specialized forest birds within cities in landscapes dominated by agriculture, it is of paramount importance to conserve any remaining urban woodlands.  相似文献

Abstract This study reports on the responses of bird assemblages to woodland clearance, fragmentation and habitat disturbance in central Queensland Australia, a region exposed to very high rates of vegetation clearance over the last two to three decades. Many previous studies of clearing impacts have considered situations where there is a very sharp management contrast between uncleared lands and cleared areas: in this situation, the contrast is more muted, because both cleared lands and uncleared savanna woodlands are exposed to cattle grazing, invasion by the exotic grass Cenchrus ciliaris and similar fire management. Bird species richness (at the scale of a 1‐ha quadrat) was least in cleared areas (8.1 species), then regrowth areas (14.6 species), then uncleared woodlands (19.9 species). Richness at this scale was unrelated to woodland fragment size, connectivity or habitat condition; but declined significantly with increasing abundance of miners (interspecifically aggressive colonial honeyeaters). At whole of patch scale, richness increased with fragment size and decreased with abundance of miners. This study demonstrates complex responses of individual bird species to a regional management cocktail of disturbance elements. Of 71 individual bird species modelled for woodland fragment sites, the quadrat‐level abundance of 40 species was significantly related to at least one variable representing environmental position (across a rainfall gradient), fragment condition, fragment size and/or connectivity. This study suggests that priorities for conservation management include: cessation of broad‐scale clearing; increased protection for regrowth (particularly where this may bolster connectivity and/or size of woodland fragments); control of miners; maintenance of fallen woody debris in woodlands; increase in fire frequency; and reduction in the incidence of grazing and exotic pasture grass.  相似文献

对北虫草栽培残基中蛋白质、虫草素等主要营养及活性物质含量分析,为其资源化合理利用研究提供参考。采用反相高效液相色谱法测定氨基酸含量,半微量凯氏定氮法测定粗蛋白,稀酸碱处理法测定粗纤维含量,旋光度法测量淀粉含量,电感耦合等离子体原子发射光谱法测定矿质元素钙及磷含量,苯酚硫酸法测定多糖的含量,高效液相色谱法测定虫草素。结果显示,北虫草栽培残基中主要营养成分氨基酸、粗蛋白、粗纤维、淀粉、含量检测分别为8. 17%、18. 4%、4. 9%、43. 1%;矿质元素钙及磷含量分别为0. 09%、0. 46%;主要活性成分虫草多糖含量可达4. 8%,与子座含量(5. 1%)相当;虫草素含量0. 318%,显著高于子座(0. 166%)中虫草素含量。结果表明,北虫草栽培残基中除富含蛋白质、氨基酸、淀粉等营养物质外,还含有虫草素、虫草多糖等北虫草特有的活性物质,具有较大营养价值。北虫草栽培残基综合利用资源化研究应用前景广阔。  相似文献

Switchgrass (Panicum virgatum) has been planted extensively for habitat restoration across the United States, such as with the Conservation Reserve Program (CRP). However, genetic profiles of these populations have never been studied nor compared with those of remnant prairies or cultivars. In this study, we sampled 16 CRP and 17 prairie populations across eastern Kansas. We assessed ploidy levels of all populations and compared genetic diversity and structure of 10 prairies, 10 CRP areas, and 5 standard cultivars, using nine simple sequence repeat (SSR) DNA markers. All CRP and prairie populations were octaploid (8x), except two prairies with both hexaploid (6x) and octaploid (8x) individuals. Based on the results of SSR analyses, there were no significant differences between CRP and prairie populations in genetic diversity, and 94% of total variation was partitioned within populations. Similarities among prairie and CRP populations were also observed in Bayesian clustering algorithms and principal coordinate analysis, suggesting that they had similar genetic compositions. In addition, positive spatial autocorrelations were detected up to 42 and 46 km among prairie and among CRP populations, respectively. To conclude, the CRP and prairie populations shared similar genetic profiles. However, remnant prairies still harbored unique genotypes and a high level of genetic diversity, highlighting the importance of seed sources for restoration efforts, that is using local wild seeds or cultivars from the same geographical region. A popular tetraploid (4x) cultivar known as “Kanlow” was genetically distinct from the prairie populations and therefore is not recommended for habitat restoration in this region.  相似文献

Questions

What are the most important drivers of plant species richness (gamma‐diversity) and species turnover (beta‐diversity) in the field layer of a forest edge? Does the tree and shrub species richness structure and complexity affect the richness of forest and grassland specialist species?

Location

Southeast Sweden.

Methods

We sampled 50 forest edges with different levels of structural complexity in agricultural landscapes. In each border we recorded trees, shrubs and herb layer species in a 50‐m transect parallel with the forest. We investigated species composition and species turnover in relation to the proportions of gaps in the border and the diversity of trees and shrubs.

Results

Total plant species richness in the field layer was mainly explained by the proportion of gaps to areas with full canopy cover and tree diversity. Increasing number of gaps promoted higher diversity of grassland specialist species within the field layer, resulting in open forest borders with the highest overall species richness. Gaps did however have a negative impact on forest species richness. Conversely, increasing forest species richness was positively related to tree diversity, but the number of grassland specialist species was negatively affected by tree diversity.

Conclusions

Managing forest borders, and therefore increasing the area of semi‐open habitats in fragmented agricultural landscapes, provides future opportunities to create a network of suitable habitats for both grassland and deciduous forest specialist species. Such measures therefore have the potential to increase functional connectivity and support dispersal of species in homogeneous forest/agricultural landscapes.  相似文献

Aim To investigate the application of environmental modelling to reconstructive mapping of pre‐impact vegetation using historical survey records and remnant vegetation data. Location The higher elevation regions of the Fleurieu Peninsula region in South Australia were selected as a case study. The Fleurieu Peninsula is an area typical of many agricultural regions in temperate Australia that have undergone massive environmental transformation since European settlement. Around 9% of the present land cover is remnant vegetation and historical survey records from the ad 1880s exist. It is a region with strong gradients in climate and topography. Methods Records of pre‐impact vegetation distribution made in surveyors’ field notebooks were transcribed into a geographical information system and the spatial and classificatory accuracy of these records was assessed. Maps of remnant vegetation distribution were obtained. Analysis was undertaken to quantify the environmental domains of historical survey record and remnant vegetation data to selected meso‐scaled climatic parameters and topo‐scaled terrain‐related indices at a 20 m resolution. An exploratory analytical procedure was used to quantify the probability of occurrence of vegetation types in environmental domains. Probability models spatially extended to geographical space produce maps of the probability of occurrence of vegetation types. Individual probability maps were combined to produce a pre‐impact vegetation map of the region. Results Surveyors’ field notebook records provide reliable information that is accurately locatable to levels of resolution such that the vegetation data can be spatially correlated with environmental variables generated on 20 m resolution environmental data sets. Historical survey records of vegetation were weakly correlated with the topo‐scaled environmental variables but were correlated with meso‐scaled climate. Remnant vegetation records similarly not only correlated to climate but also displayed stronger relationships with the topo‐scaled environmental variables, particularly slope. Main conclusions A major conclusion of this study is that multiple sources of evidence are required to reconstruct past vegetation patterns in heavily transformed region. Neither the remnant vegetation data nor historical survey records provided adequate data sets on their own to reconstruct the pre‐impact vegetation of the Fleurieu Peninsula. Multiple sources of evidence provide the only means of assessing the environmental and historical representativeness of data sets. The spatial distribution of historical survey records was more environmentally representative than remnant vegetation data, which reflect biases due to land clearance. Historical survey records were also shown to be classificatory and spatially accurate, thus are suitable for quantitative spatial analyses. Analysis of different spatial vegetation data sets in an environmental modelling framework provided a rigorous means of assessing and comparing respective data sets as well as mapping their predicted distributions based on quantitative correlations. The method could be usefully applied to other regions where predictions of pre‐impact vegetation cover are required.  相似文献

We compared early plant succession in four abandoned pastures of differing age since abandonment and a nearby secondary forest site in northwestern Ecuador. Two “Open” pastures had no tree canopy covering, and two “Guava” pastures had a well-developed canopy cover of Psidium guajava. No site had been seeded with pasture grasses. All pastures were compared in a chronological sequence; two were monitored for 18 months. Species richness was consistently higher in Guava sites than in Open sites and it continued to increase over time, whereas it remained static in Open sites. Species richness was highest in secondary forest. Recruitment of tree saplings in Guava sites was lower than in secondary forest; however, it was nearly absent in Open sites. The seed bank contained predominantly herbaceous species at all sites, and was highly dissimilar to aboveground vegetation. Dominance-diversity curves for Guava sites showed a more equitable distribution of species that increased over time. In contrast, dominance-diversity curves for Open sites were static and indicated dominance by a few aggressive species. Soil characteristics among sites were variable; however, a principal components analysis on soils isolated the older Open site from all others. The older Open site had the lowest species richness and was dominated by Baccharis trinervis, an aggressive shrub species. The site appears to be in a state of arrested succession and some form of restorative intervention may be necessary to initiate succession toward a forested condition. Succession in Guava sites appears headed toward secondary forest, whereas it does not in Open sites.  相似文献

Abstract Ecological degradation within areas of remnant forest may be amplified if the effects of fragmentation interact with the effects of other environmental disturbances such as wind storms. We used before–after comparisons to assess the effects of Tropical Cyclone Larry on remnant and continuous rainforest in the Wet Tropics uplands of north‐eastern Australia. Vegetation structure was measured 3 years before the cyclone and 6 months afterwards, at eight continuous forest sites and eight remnants (6–37 ha), within 20 km of the cyclone's track. The cyclone caused extensive defoliation, felling and breakage of stems and branches (greatest among the trees >100 cm diameter which had around 50% stem loss), and increased litter and woody debris. Cyclone effects were strongly influenced by a site's spatial position (P = 0.005, 0.001 in multivariate analyses of overall damage). Maximum damage occurred 10–15 km south of the cyclone track, perhaps because of the additive effects of the west‐moving air at the southern eyewall combined with the cyclone's own rapid westward movement. Most fragments were south of the cyclone track, as a consequence of spatially selective deforestation practices, and therefore, showed greatest damage. However, once the effects of spatial position were considered, the independent differences in cyclone effects between fragments and continuous forest were lost (P = 0.23, 0.41 when north‐south distance was included as a covariate in analyses). The expected protection afforded by a continuous forest canopy seems to have disappeared in the face of extremely strong cyclonic winds and down‐draughts. Nevertheless, an interaction between fragmentation and disturbance may yet occur, during the period of post‐cyclone recovery, owing to the effects of landscape context on plant recruitment. For example, there was a higher diversity of exotic seedling germination in fragments, independent of the extent of cyclone damage.  相似文献

Abstract Species need to disperse at a broad range of spatial scales, the recognition of which has spawned programmes such as Wildlands and WildCountry that aim to restore large‐scale connectivity. To achieve connectivity, a first step is to understand how wildlife uses existing remnants. In this study we examine the effect of remnant isolation and condition on the reptile fauna of fragmented mallee habitats in southern Australia. In three replicate landscapes we use pitfall traps to survey reptiles in five landscape elements: Conservation Park, connected, disconnected and isolated fragments, and the agricultural matrix. Reptile species richness, abundance, abundance of snakes, skinks and the 10 most common species had no significant association with landscape elements, excluding the matrix. This was despite a substantial reduction in plant species richness in the fragments, particularly of shrubs. Only seven individual reptiles were captured in the matrix, most on one site with deep sandy soils. The farmland on clay soils appeared to be relatively impermeable for reptiles, although four species could traverse 100 m of cleared sand‐dune. The lack of an isolation effect suggests that populations in remnants are persistent, or that occasional dispersal by common reptiles maintains populations. In contrast with common species, fewer rare species were captured in remnants compared with the Conservation Park, implying that some species may be entirely excluded from the remnants. Our study suggests that the spatial configuration and condition of the fragments sustain populations of many common reptile species. Remnants will therefore be invaluable as attempts are made to restore landscape‐scale permeability. However, additional conservation effort should be made to restore plant species that have been lost from the agricultural landscape. Future research should aim to better define the suite of reptile species that may not be able to use the remnants at all.  相似文献
