目的测定诺氟沙星、恩诺沙星、环丙沙星、安普霉素和庆大霉素5种抗生素对林麝化脓隐秘杆菌的最小杀菌浓度,为选择治疗林麝脓肿病的有效药物提供指导。方法将菌液与不同浓度的抗生素混合作用30min,再加入中和剂作用10min,吸取300mL作用液均匀涂布在胎牛血清培养基上,38℃培养48h,观察并记录。结果 5种抗生素对化脓隐秘杆菌的最小杀菌浓度不一致,对4株菌杀菌效果相对较好的药物为环丙沙星(CIP),但对TP1需要较高浓度(128μg/mL)。结论喹诺酮类抗生素虽然抑菌效果较明显,但要完全杀死化脓隐秘杆菌却需要较高浓度,这很可能与化脓隐秘杆菌菌膜的存在有关。到目前为止,治疗林麝脓肿病的首选药物为喹诺酮类抗生素,尤以环丙沙星效果最佳。  相似文献

Evolution and ecology of antibiotic resistance genes   总被引:8,自引:0,他引:8  
A new perspective on the topic of antibiotic resistance is beginning to emerge based on a broader evolutionary and ecological understanding rather than from the traditional boundaries of clinical research of antibiotic-resistant bacterial pathogens. Phylogenetic insights into the evolution and diversity of several antibiotic resistance genes suggest that at least some of these genes have a long evolutionary history of diversification that began well before the 'antibiotic era'. Besides, there is no indication that lateral gene transfer from antibiotic-producing bacteria has played any significant role in shaping the pool of antibiotic resistance genes in clinically relevant and commensal bacteria. Most likely, the primary antibiotic resistance gene pool originated and diversified within the environmental bacterial communities, from which the genes were mobilized and penetrated into taxonomically and ecologically distant bacterial populations, including pathogens. Dissemination and penetration of antibiotic resistance genes from antibiotic producers were less significant and essentially limited to other high G+C bacteria. Besides direct selection by antibiotics, there is a number of other factors that may contribute to dissemination and maintenance of antibiotic resistance genes in bacterial populations.  相似文献

AIMS: To study the effect of oral administration of a quinolone on emergence of resistance in an indicator bacterial species from faecal flora. METHODS AND RESULTS: Quinolone resistance was studied in Escherichia coli obtained from the faecal contents of pigs housed in nine commercial farrow-to-finish herds in France after administration of flumequine to sows. The percentage of quinolone-resistant E. coli increased in the faeces of sows after administration of flumequine (mean 21.78% at day 7 vs 6.42% before treatment for nalidixic acid) and then decreased (mean 12.6 and 10.4 at days 30 and 60, respectively for nalidixic acid), being not significantly different from initial values 1 month post-treatment. In young pigs, the proportion of resistant strains was lower and decreased over rearing period. Moreover, changes over time of both total E. coli and the proportion of resistant bacteria exhibited great inter-individual variability. CONCLUSIONS: Restoration of susceptible faecal flora occurred within 2 months after flumequine treatment. SIGNIFICANCE AND IMPACT OF THE STUDY: Effect of flumequine treatment of sows on the quinolone resistance of faecal E. coli of both sows and their progeny is noticeable but transitory.  相似文献

The cathodic electrochemiluminescence (ECL) of peroxydisulphate (S2O82?)–ciprofloxacin (CPF) system at a wax‐impregnated graphite electrode was studied. When CPF was absent, S2O82? was electrochemically reduced to sulphate free radical (SO4??), and dissolved oxygen absorbed on the electrode surface was reduced to protonated superoxide anion radical (HO2?). The HO2? was oxidized by SO4?? to produce molecular oxygen in both singlet and triplet states. Some of the singlet molecular oxygen (1O2) further combined through collision to be an energy‐rich precursor singlet molecular oxygen pair (1O2)2. A weak ECL was produced when 1O2 or (1O2)2 was converted to ground‐state molecular oxygen (3O2). When CPF was present, a stronger ECL was produced, which originated from two emitting species. The main emitting species was excited state CPF (CPF*), which was produced by accepting energy from (1O2)2. The other emitting species was excited singlet molecular oxygen pair [(1O2)2*], which originated from the chemical oxidation of CPF by SO4?? and dissolved oxygen. Based on the stronger ECL phenomenon, an ECL method for the determination of either S2O82? or CPF was proposed. The Arabidopsis thaliana genome contains four genes that were originally annotated as potentially encoding DNA gyrase: ATGYRA, ATGYRB1, ATGYRB2, and ATGYRB3. Although we subsequently showed that ATGYRB3 does not encode a gyrase subunit, the other three genes potentially encode subunits of a plant gyrase. We also showed evidence for the existence of supercoiling activity in A. thaliana and that the plant is sensitive to quinolone and aminocoumarin antibiotics, compounds that target DNA gyrase in bacteria. However, it was not possible at that time to show whether the A. thaliana genes encoded an active gyrase enzyme, nor whether that enzyme is indeed the target for the quinolone and aminocoumarin antibiotics. Here we show that an A. thaliana mutant resistant to the quinolone drug ciprofloxacin has a point mutation in ATGYRA. Moreover we show that, as in bacteria, the quinolone-sensitive (wild-type) allele is dominant to the resistant gene. Further we have heterologously expressed ATGYRA and ATGYRB2 in a baculovirus expression system and shown supercoiling activity of the partially purified enzyme. Expression/purification of the quinolone-resistant A. thaliana gyrase yields active enzyme that is resistant to ciprofloxacin. Taken together these experiments now show unequivocally that A. thaliana encodes an organelle-targeted DNA gyrase that is the target of the quinolone drug ciprofloxacin; this has important consequences for plant physiology and the development of herbicides.  相似文献

A new method based on resonance Rayleigh scattering (RRS) was proposed for the determination of quinolones (QNS) at the nanogram level. In pH 3.3–4.4 Britton–Robinson buffer medium, quinolones such as ciprofloxacin, pipemidic acid (PIP), lomefloxacin (LOM), norfloxacin (NOR) and sarafloxacin (SAR) were protonated and reacted with methyl orange (MO) to form an ion‐pair complex, which then further formed a six‐membered ring chelate with Pd(II). As a result, new RRS spectra appeared and the RRS intensities were enhanced greatly. RRS spectral characteristics of the MO–QNS–Pd(II) systems, the optimum conditions for the reaction, and the influencing factors were investigated. Under optimum conditions, the scattering intensity (∆I) increments were directly proportional to the concentration of QNS with in certain ranges. The method had high sensitivity, and the detection limits (3σ) ranged from 6.8 to 12.6 ng/mL. The proposed method had been successfully applied for the determination of QNS in pharmaceutical formulations and human urine samples. In addition, the mechanism of the reaction system was discussed based on IR, absorption and fluorescence spectral studies. The reasons for the enhancement of scattering spectra were discussed in terms of fluorescence‐scattering resonance energy transfer, hydrophobicity and molecular size. Veterinary pharmaceuticals contained in dead livestock may be ingested by avian scavengers and negatively affect their health and consequently their population dynamics and conservation. We evaluated the potential role of antibiotics as immunodepressors using multiple parameters measuring the condition of the cellular and humoral immune system in griffon (Gyps fulvus), cinereous (Aegypius monachus) and Egyptian vultures (Neophron percnopterus). We confirmed the presence of circulating antimicrobial residues, especially quinolones, in nestlings of the three vulture species breeding in central Spain. Individuals ingesting antibiotics showed clearly depressed cellular and humoral immune systems compared with nestlings from the control areas, which did not ingest antibiotics. Within central Spain, we found that individuals with circulating antibiotics showed depressed cellular (especially CD4+and CD8+T-lymphocyte subsets) and humoral (especially acellular APV complement and IL8-like) immune systems compared with nestlings without circulating antibiotics. This suggests that ingestion of antibiotics together with food may depress the immune system of developing nestlings, temporarily reducing their resistance to opportunistic pathogens, which require experimental confirmation. Medicated livestock carrion should be considered inadequate food for vultures due to their detrimental consequences on health derived from the ingestion and potential effects of the veterinary drugs contained in them and for this reason rejected as a management tool in conservation programmes.  相似文献
