A staining method is described using thionin, for undecalcified deacrylated bone sections. RNA is stained purplish violet, allowing still active osteoblasts to be distinguished from lining cells. Staining intensity of mineralized bone is related to the degree of mineralization. Mineralizing fronts and cement lines are visualized clearly. Lamellae show an alternate pattern. Histomorphometric parameters such as osteon thickness and interstitial bone thickness can be measured without using polarized light. The mineralizing front can be assessed and expressed as a percentage of the osteoblast-covered interface between osteoid and mineralized bone. The stain is also useful for qualitative assessment of metabolic bone disease. Thionin stained sections can be kept for at least one year when stored hi the dark at 7 C.  相似文献

Leaf thionins of barley have been identified as a novel class of highly abundant polypeptides with antifungal activity, which are present in walls and vacuoles of barley leaf cells. Similar thionins occur not only in monocotyledonous but also in various dicotyledonous plants. The leaf thionins of barley are encoded by a complex multigene family, which consists of at least 50–100 members per haploid genome. The toxicity of these thionins for plant pathogenic fungi and the fact that their synthesis can also be triggered by pathogens strongly suggest that leaf thionins are involved in the mechanism of plant defence against microbiol infection.  相似文献

Tomato is one of the most important crop plants; however, attacks by pathogens can cause serious losses in production. In this report, we explore the potential of using the Arabidopsis thionin (Thi2.1) gene to genetically engineer enhanced resistance to multiple diseases in tomato. Potential thionin toxicity in fruits was negated by the use of a fruit-inactive promoter to drive the Thi2.1 gene. In transgenic lines containing RB7/Thi2.1, constitutive Thi2.1 expression was detected in roots and incidentally in leaves, but not in fruits. Disease assays revealed that the transgenic lines that were tested conferred significant levels of enhanced resistance to bacterial wilt (BW) and Fusarium wilt (FW). Further studies indicated that BW disease progression in transgenic lines was delayed by a systemic suppression of bacterial multiplication. By adopting a safe genetic engineering strategy, the present investigation is another step forward demonstrating thionin practicality in crop protection.  相似文献

Two novel chitin-binding peptides, designated Pp-AMP 1 and Pp-AMP 2, which had antimicrobial activity against pathogenic bacteria and fungi, were purified from Japanese bamboo shoots (Phyllostachys pubescens) by a simple procedure based on chitin affinity chromatography. They had the common structural features of the plant defensin family, but they could not be grouped in any type of that family. They showed a high degree of homology to mistletoe toxins.  相似文献

The characterization of fluorescelnphosphatidylethanolamlne (FPE) as a real-time Indicator of the electrostatic nature of a membrane surface is described. The conditions appropriate for the labelling of membranes and the implementation of FPE as a tool to monitor the interactions of various peptides with model membranes are outlined. It is shown that of the membrane-active peptides studied, Naja naja kaouthla cardiotoxin and pyrularia thionin bind to certain model membranes without insertion. Whereas the leader sequence of the nuclear encoded subunit IV of mammalian cytochrome c oxidase (E.C. 1.9.3.1), known as p-25, and melittin appear to bind and then partially insert into the membrane. It seems evident also that melittin does not adopt a fully transmembrane configuration. Melittin is known to promote membrane lysis and by employing a rapid-kinetic technique it is shown that the time-course of such lysis does not appear to correlate with peptide binding, but following binding a significant proportion of melittin must become inserted into the membrane before lysis appears to commence.  相似文献

Summary In etiolated barley seedlings a highly abundant mRNA encoding a 15 000 Mr polypeptide is present whose concentration rapidly declines upon illumination. The amino acid sequence of the 15 000 Mr polypeptide has been deduced from cDNA sequences and the polypeptide has been identified as a high-molecular-weight thionin precursor. Closely related thionins, most of them highly toxic, have been described previously in several higher plants. In cereals the occurrence of these thionins has been thought to be confined to the seeds. Our present data demonstrate that, in additon to endosperm-specific hordothionin mRNA, a closely related but distinct second group of thionin mRNAs is present in the barley leaf during early seedling development. Since the appearance of the bordothionin mRNA is not controlled by light, two different gene families for thionins exist whose expression seems to be under a different mode of developmental control.Dedicated to Prof. W. 丹参EST序列的Blast分析表明,一条序列与硫堇(thionin,THI)基因有较高的同源性,该序列长575bp,包含1个长366bp的开放阅读框(ORF),编码121个氨基酸,命名为SmTHI,GenBank登陆号为DQ212984。在此基础上设计引物,分别从cDNA和gDNA水平上克隆到该基因的全编码区序列的结果表明,该基因无内含子。序列分析表明,该编码蛋白与大多数植物的THI蛋白前体高度同源,并符合植物硫堇类蛋白的序列模式和特征:C—C—x(5).R.x(2)-[FY]-x(2)-C,N端具17个氨基酸的信号肽,中间46个氨基酸为成熟THI部分,C端的58个氨基酸为酸性多肽部分。成熟的THI蛋白带正电荷,偏碱性,推测可能有抗病原微生物活性。实时定量PCR检测SmTHI在丹参不同组织部位的表达以及在黄瓜细菌性角斑病菌(PSL)、NaCl和水杨酸(sA)溶液诱导下的表达结果表明:SmTHI在植物的根、茎和叶中均有不同程度的表达,其表达丰度为叶〉茎〉根:在PSL、NaCl和sA溶液诱导下该基因的表达呈上调趋势。  相似文献

The anthracnose fungus, Colletotrichum gloeosporioides, interacts incompatibly with the ripe fruit of pepper (Capsicum annuum). It interacts compatibly with the unripe-mature fruit. We isolated a defensin gene, j1-1, and a thionin-like gene, PepThi, expressed in the incompatible interaction by using an mRNA differential display method. Both genes were developmentally regulated during fruit ripening, organ-specifically regulated, and differentially induced during the compatible and incompatible interactions. Expression of the PepThi gene was rapidly induced in the incompatible-ripe fruit upon fungal infection. The fungus-inducible PepThi gene is highly inducible only in the unripe fruit by salicylic acid. In both ripe and unripe fruit, it was induced by wounding, but not by jasmonic acid. Expression of the j1-1 gene is enhanced by jasmonic acid in the unripe fruit but suppressed in the ripe fruit. These results suggest that both small and cysteine-rich protein genes are induced via different signal transduction pathways during fruit ripening to protect the reproductive organs against biotic and abiotic stresses.  相似文献

The three-dimensional structure of the Sorghum bicolor seed protein γ-thionin SIα1 has been determined by 2D 1H nuclear magnetic resonance (NMR) spectroscopy. The secondary structure of this 47-residue antifungal protein with four disulphide bridges consists of a three-stranded antiparallel sheet and one helix. The helix is tethered to the sheet by two disulphide bridges which link two successive turns of the helix to alternate residues i, i + 2 in one strand.
