We examine the issue of prochlorophyte origins and provide analyses which highlight the limitations of inferring evolutionary trees from anciently diverged sequences that have markedly different GC contents. Under these conditions we have found that current tree reconstruction methods strongly group together sequences with similar GC contents, whether or not the sequences share a common ancestor. We provide 3psbA termini sequence forProchloron didemni and find it does not have the 7 amino acid deletion that occurs in Chla/b chloroplasts andProchlorothrix hollandica. This is consistent with the recent findings of a Chlc like pigment in the light harvesting system in other prochlorophytes but apparently absent inP. hollandica. From these observations we suggest thatP. hollandica is the prochlorophyte most closely related to Chla/b containing chloroplasts and hence the most appropriate prokaryotic model for higher plant Chla/b photosynthesis.  相似文献

Ingestion and assimilation rates of Euchlanis dilatata lucksiana Hauer, isolated from Lake Loosdrecht (The Netherlands) and cultured on lake water (seston < 33 µm), were measured in the laboratory using the 14C-tracer technique. The five taxa used as tracer foods, together with 6–7 mg C l–1 of lake seston in each case, included four species of filamentous cyanobacteria (Oscillatoria redekei, O. limnetica, Aphanizomenon flos-aquae, Anabaena PCC 7120) and a prochlorophyte (Prochlorothrix hollandica). Except Anabaena, they are all commonly encountered in eutrophic Loosdrecht lakes, including Lake Loosdrecht, and their dimensions ranged between 150 and 250 µm in length and 2 and 3.5 µm in width. The small and large Euchlanis used as experimental animals had mean lengths of 217–223 µm and 276–305 µm, respectively. Euchlanis fed on all the taxa offered as food. Clearance rates ranged from 51 to 99 µl ind–1 d–1 for the large animals and from 22 to 41 µl ind–1 d–1 for the small animals. The highest ingestion rate observed, 1.7 µg ind–1 d–1, was for the large animals feeding on Aphanizomenon. The daily ration for both size classes far exceeded 100% of body weight, reaching up to 690% for the small animals feeding on Aphanizomenon. The small animals generally appeared to assimilate the ingested food more efficiently (assimilation efficiencies: 37–100%) than the large animals (34–77%). Compared with an earlier study in which only lake seston (<33 µm) was used as food, the specific clearance rates of Euchlanis on the cyanobacteria and Prochlorothrix were generally somewhat lower.  相似文献

Apoproteins of the antenna complexes of Prochlorococcus marinus clone SS120 (= CCMP 1375) and Prochlorococcus sp. clone MED4 (= CCMP 1378) cross-reacted with an antibody against the 30 kDa CP 5 complex of Prochlorothrix hollandica antenna. For the MED4 strain, which has a high divinyl-chlorophyll a to divinyl-chlorophyll b (DV-Chl a/b) ratio ranging from 11.4 to 15.0 (w/w), the major antenna proteins had an apparent molecular mass of 32.5 kDa. In contrast for the SS120 strain, which has a low DV-Chl a/b ratio ranging from 1.1 to 2.2, antenna apoproteins were observed in the range 34–38 kDa. For both strains, these apoproteins decreased at high growth irradiance but more markedly in the latter. Partially purified antenna fractions had a DV-Chl a/b ratio ca. 7-fold lower for SS120 than for MED4 at 30 mol photons m-2 s-1. For both strains, the 77 K fluorescence emission spectra of whole thylakoids displayed a major peak at 685 nm and a broad but very low shoulder above 700 nm. Energetic coupling of the antenna to both PS II and PSI reaction centers was demonstrated for SS120 by the strong contribution of DV-Chl b in both the 77 K excitation fluorescence spectra and the oxidized minus reduced absorption difference spectra of P700. The PS I to PS II ratio of Prochlorococcus SS120 was determined as being 0.7 ± 0.1 at low light.  相似文献

A lipid analysis is reported of a symbiont Prochloron sp. associated with the ascidian Lissorclinum patella collected from Rodda Reef, North Queensland, Australia. Phosphatidyl glycerol, monogalacosyl diglyceride, digalacosyl diglyceride and sulfoquinovasyl diglyceride were identified. These lipids together with their fatty acid profiles, the limited range of hydrocargons (nC13-nC17), and sterol analysis (chlesterol 27%) are consistent with a firm relaionship fot the new genus Prochloron with he Cyanophya.  相似文献

The freshwater filamentous green oxyphotobacterium Prochlorothrix hollandica is an unusual oxygenic photoautotrophic cyanobacterium differing from most of the others by the presence of light-harvesting Pcb antenna binding both chlorophylls a and b and by the absence of phycobilins. The pigment-protein complexes of P. hollandica SAG 10.89 (CCAP 1490/1) were isolated from dodecylmaltoside solubilized thylakoid membranes on sucrose density gradient and characterized by biochemical, spectroscopic and immunoblotting methods. The Pcb antennae production is suppressed by high light conditions (> 200 μmol photons m−2 s−1) in P. hollandica. PcbC protein was found either in higher oligomeric states or coupled to PS I (forming antenna rings around PS I). PcbA and PcbB are most probably only very loosely bound to photosystems; we assume that these pigment-protein complexes function as low light-induced mobile antennae. Further, we have detected α-carotene in substantial quantities in P. hollandica thylakoid membranes, indicating the presence of chloroplast-like carotenoid synthetic pathway which is not present in common cyanobacteria.  相似文献

The chlorophyll (Chl) a/b proteins of the photosynthetic prokaryotes appear to have evolved by gene duplication and divergence of the core Chl a antenna family, which also includes CP43 and CP47 and the iron-stress induced Chl a-binding IsiA proteins. We show here that Prochlorothrix hollandica has a cluster of three pcb (prochlorophyte chlorophyll b) genes which are co-transcribed. The major antenna polypeptides of 32 and 38 kDa are encoded by pcbA and pcbC respectively. The pcbC gene is significantly divergent from the other two and may have originated by a gene duplication independent of the one that led to isiA and the other prochlorophyte pcb genes. The distant relatedness of the three prochlorophyte genera implies that not only the ability to make Chl b and use it for light-harvesting arose independently in the three lineages, but also that the pcb genes may have arisen as the result of independent gene duplications in each lineage.  相似文献

A 25 kDa protein associated with Photosystem I (PS I) of the divinyl-chlorophyll a/b-containing oxychlorobacterium Prochlorococcus marinus SS120 (CCMP 1375) was isolated, and the amino acid sequences of the N-terminus and one internal peptide were determined. Polymerase chain reaction (PCR) with degenerate primers yielded a 92 bp fragment, which was used to isolate the complete gene from a genomic library. The corresponding gene was isolated from a library of Prochlorococcus sp. MED4 (CCMP 1378). In both Prochlorococcus strains, the gene encodes a protein of 199 amino acids. The gene products show a strong sequence similarity to the PS I subunit PsaL. The N-terminus contains a hydrophilic domain that has not been found in PsaL proteins from other organisms. In both strains, sequences encoding a protein similar to PsaI were found upstream of the psaL gene. Both genes are transcribed in the same direction.  相似文献

pcb genes, encoding proteins binding light-harvesting chlorophylls, were cloned and sequenced from the Chl d-containing cyanobacterium, Acaryochloris marina, and the Chl b-containing cyanobacterium, Prochloron didemni. Both organisms contained two tandem pcb genes. Peptide fingerprinting confirmed the expression of one of the A. marina pcb genes. Phylogenetic tree reconstruction using distance-matrix and maximum-likelihood methods indicated a single origin of the pcb gene family, whether occurring in Chl b-containing or Chl d-containing organisms. This may indicate widespread lateral transfer of the Pcb protein-based light-harvesting system.  相似文献

The nucleotide sequences of the genes coding for the subunits of the Photosystem I (PS I) core, PsaA and PsaB were determined for the marine prokaryotic oxyphototrophs Prochlorococcus sp. MED4 (CCMP1378), P. marinus SS120 (CCMP1375) and Synechococcus sp. WH7803. Divergence of these sequences from those of both freshwater cyanobacteria and higher plants was remarkably high, given the conserved nature of PsaA and PsaB proteins. In particular, the PsaA of marine prokaryotes showed several specific insertions and deletions with regard to known PsaA sequences. Even in between the two Prochlorococcus strains, which correspond to two genetically different ecotypes with shifted growth irradiance optima, the sequence identity was only 80.2% for PsaA and 88.9% for PsaB. Possible causes and implications of the fast evolution rates of these two PS I core subunits are discussed.
