Although recent studies in patients with paternal uniparental disomy 14 [upd(14)pat] and other conditions affecting the chromosome 14q32.2 imprinted region have successfully identified underlying epigenetic factors involved in the development of upd(14)pat phenotype, several matters, including regulatory mechanism(s) for RTL1 expression, imprinting status of DIO3 and placental histological characteristics, remain to be elucidated. We therefore performed molecular studies using fresh placental samples from two patients with upd(14)pat. We observed that RTL1 expression level was about five times higher in the placental samples of the two patients than in control placental samples, whereas DIO3 expression level was similar between the placental samples of the two patients and the control placental samples. We next performed histological studies using the above fresh placental samples and formalin-fixed and paraffin-embedded placental samples obtained from a patient with a maternally derived microdeletion involving DLK1, the-IG-DMR, the MEG3-DMR and MEG3. Terminal villi were associated with swollen vascular endothelial cells and hypertrophic pericytes, together with narrowed capillary lumens. DLK1, RTL1 and DIO3 proteins were specifically identified in vascular endothelial cells and pericytes, and the degree of protein staining was well correlated with the expression dosage of corresponding genes. These results suggest that RTL1as-encoded microRNA functions as a repressor of RTL1 expression, and argue against DIO3 being a paternally expressed gene. Furthermore, it is inferred that DLK1, DIO3 and, specially, RTL1 proteins, play a pivotal role in the development of vascular endothelial cells and pericytes.  相似文献

We present rapid aneuploidy diagnosis of partial trisomy 3q (3q27.3→qter) and partial monosomy 14q (14q31.3→qter) of paternal origin by aCGH using uncultured amniocytes in a fetus with hypotonia, scoliosis, arthrogryposis, hyperextensible joints, facial dysmorphism, ventricular septal defect, pulmonary stenosis, clenched hands, clubfoot, scalp edema and right hydronephrosis. We discuss the genotype–phenotype correlation of 3q duplication syndrome and terminal 14q deletion syndrome. We demonstrate that fetuses with a paternal-origin deletion of 14q involving the 14q32.2 imprinted region may prenatally present the upd(14)mat-like phenotype such as hypotonia, scoliosis, arthrogryposis and hyperextensible joints.  相似文献

Chromosome 14 is often involved in chromosome rearrangements, although pericentric inversions are rare. Here we report a mother carrying a pericentric inversion of chromosome 14, and her daughter with recombinant chromosome characterized by a partial distal 14q trisomy. Principal clinical findings of the child include facial anomalies, microcephaly, developmental delay, hypotonia and cardiac malformation. Her final karyotype was 46,XX,rec(14)dup(14q)inv(14)(p12q31)mat[20], arr 14q31.3qter(85,427,839–106,356,482)x3. This report brings new data about clinical features of partial 14q trisomy and molecular analysis enables the visualization of genes involved in the segment duplicated.  相似文献

数学形态学是用数学方法描述或分析一个物体图象的形状的理论和方法,是图象处理和图象识别技术的发展,但在生物学当中的应用还很有限。本文介绍了一个新的分支学科——昆虫数学形态学,包括三方面的内容:①昆虫数学形态学技术研究,涉及昆虫图象数字化技术和昆虫图象处理与识别技术;②昆虫数学形态学理论研究,主要以昆虫图象的解释和理解研究及昆虫数学形态学与分类学等学科的关系研究为主;③昆虫和昆虫数学形态学应用基础研究,涉及昆虫数学形态学数据库及其分析软件开发,昆虫图象的机器学习和计算机视觉等内容。昆虫数学形态学理论和方法与计算机视觉技术相结合,在害虫虫情监测、昆虫多媒体专家系统的构建等方面具有广阔的应用前景。  相似文献

Indigenous formulations based on cow dung fermentation are commonly used in organic farming. Three biodynamic preparations viz., Panchagavya (PG), BD500 and ‘Cow pat pit’ (CPP) showed high counts of lactobacilli (109 ml−1) and yeasts (104 ml−1). Actinomycetes were present only in CPP (104 ml−1) and absent in the other two. Seven bacterial isolates from these ferments were identified by a polyphasic approach: Bacillus safensis (PG1), Bacillus cereus (PG2, PG4 PG5), Bacillus subtilis (BD2) Lysinibacillus xylanilyticus (BD3) and Bacillus licheniformis (CPP1). This is the first report of L. xylanilyticus and B. licheniformis in biodynamic preparations. Only three carbon sources—dextrose, sucrose and trehalose out of 21 tested were utilized by all the bacteria. None could utilize arabinose, dulcitol, galactose, inositol, inulin, melibiose, raffinose, rhamnose and sorbitol. All the strains produced indole acetic acid (1.8–3.7 μg ml−1 culture filtrate) and ammonia. None could fix nitrogen; but all except B. safensis and B. licheniformis could solubilize phosphorous from insoluble tri-calcium phosphate. All the strains except L. xylaniliticus exhibited antagonism to the plant pathogen Rhizoctonia bataticola whereas none could inhibit Sclerotium rolfsi. In green house experiment in soil microcosms, bacterial inoculation significantly promoted growth of maize; plant dry weight increased by ~21 % due to inoculation with B. cereus (PG2). Results provide a basis for understanding the beneficial effects of biodynamic preparations and industrial deployment of the strains.

In pots, 25 populations of potato pale cyst-nematode, Globodera pallida Stone, differed significantly in their ability to multiply on potato clones P55/7 and ZC83/ 6, both fully resistant to G. pallida pathotype Pal. Neither clone was fully resistant to any of the populations. For 21 populations common to this and an earlier experiment, increase on the more resistant potatoes (P55/7, ZC83/6, cvs Sante, Paladin and Glenna) was correlated with their increase on less resistant potatoes (cvs Morag, Fingal and Valiant). Variation in virulence on these partially resistant potatoes was not matched by differences in the electrophoretic patterns of the nematodes' proteins. The identification of populations of G. rostochiensis (Woll.) Skarbilovich used in these experiments was confirmed by electrophoresis. All populations of G. pallida Stone, appeared to contain very small numbers of G. rostochiensis after subculture on susceptible potatoes (cv. Désirée).  相似文献
