Summary Fathead minnows, Pimephales promelas, and yellow perch, Perca flavescens, were transferred from moderately soft Lake Superior water (hardness 45mg/l as CaCO3) to very soft diluted Lake Superior water (hardness 4.5mg/l). Sulfuric acid was added in some treatments by means of a multichannel diluter. In very soft water, chloride cells proliferated in the gills, especially in the epithelium of the secondary lamellae. When exposed to acid, chloride cells were damaged and less abundant in the secondary lamellae, and blood osmolality was reduced at pH 5.0 (x = 188 mOsm/kg, 9 days exposure; normal 280 mOsm/kg) for the minnows and pH 4.1 (x = 218 mOsm/kg, 58 days exposure; normal 329 mOsm/kg) for the perch. Certain chloride cells which form gland-like clusters in the primary lamellae of perch gills showed little damage even at pH 4.1. The present study supports the view that chloride cells proliferate in very soft fresh water to help maintain ionic balances, and that damage to these cells in acidified soft water may be related to diminished ionoregulatory capacity. The greater acid tolerance of chloride cells of, and the higher blood osmolality maintained by, perch could help to explain the greater tolerance of this species to low pH. In some cases, a species' ability to acclimate to very soft water and acidified soft water may depend upon the number, distribution, and physiology of its chloride cells.  相似文献

Parasitism by a mermithid nematode, Romanomermis culicivorax, causes severe depletion of haemolymph carbohydrates and proteins in mosquito larvae. We undertook a study to determine if haemolymph osmolality and cation concentrations were affected also by mermithid parasitism. The haemolymph osmolality of R. culicivorax-infected and control Aedes taeniorhynchus and Culex pipiens fourth-instar larvae was not significantly different. However, the haemolymph osmolality decreased significantly in infected Anopheles quadrimaculatus. Each mosquito species demonstrated significant alterations in the haemolymph concentration of at least one cation when infected although the cation concentrations affected differed for each species. The changes observed were statistically significant but the magnitude of change was not great. Overall, despite the severe nutritional burden of the mermithid nematode, these species of mosquito larvae can continue to maintain osmoregulation.  相似文献

The concentrations of taurine in blood and brain regions of the toadBufo boreas have been measured. Most of these values are considerably lower than those found in mammals. Using an antibody prepared against conjugated taurine, the distribution of taurine in three brain regions of the toad has been visualized. The possible osmoregulatory functions of taurine have been investigated by making toads hyper- or hypo-osmotic in vivo. Induction of hypoosmolality is accompanied by a massive taurine tide in blood plasma, but has no immediate effects upon the taurine concentrations in the brain areas studied. However, histochemical visualization indicates a marked redistribution of taurine between cellular components and extracellular space of brain tissues. This may indicate that taurine has an osmoregulatory function in brain tissue under hypo-osmotic conditions. Hyperosmolality results in no elevation of the taurine concentration in blood plasma of toads, but rather in a very gradual decline of total plasma taurine content over a prolonged time period. Histochemical studies reveal little change in frontal cortex after 1 hour but deeper staining of many neurons in optic lobe accompanied by greater staining in the extracellular fluid. By 3 hours there is a depletion of taurine from all compartments of cerebral cortex tissues. No evidence of any prolonged direct osmoregulatory role for taurine is indicated under hyperosmotic conditions. The aim of this study was to investigate the effect of CO2-induced water acidification (pH values: 8.1 ? control, 7.5 and 7.0) on the hemolymph osmolality and total metabolic rate of amphipod Gammarus oceanicus under short-term exposure. The hemolymph osmolality was measured using a vapor pressure osmometer. The metabolic rate was determined based on the heat dissipation measurements using a Calvet-type isothermal twin calorimeter. The hemolymph osmolality remained unchanged after exposure to pH 7.5 and it increased in the lowest treatment. Acidification did not significantly affect the resting or active metabolic rate. The observed results indicate that high osmolality maintained during hypercapnia did not generate additional physiological costs. It may therefore be concluded that the studied species is preadapted for this factor. The long-term adaptation to hypoosmotic conditions in the brackish Baltic Sea might also have preadapted it to hypercapnia because similar physiological mechanisms are responsible for the resistance to both factors.  相似文献

Several factors regulating activation of spermatozoon motility in Eurasian burbot, Lota lota, including osmolality, calcium (Ca2+) ions, and temperature were investigated. Spermatozoon motility in Eurasian burbot, Lota lota was assessed at 4 and 30°C in seminal fluid, isotonic media (with and without Ca2+) and hypotonic media (with and without Ca2+). Spermatozoa were spontaneously activated in seminal fluid at 20°C and the maximum motility was recorded at 30°C, which is out of the spawning temperature range, indicating that no risk of activation occurs during routine semen handling in artificial insemination. Initiation of spermatozoon motility in L. lota is mediated by Ca2+ and sensitivity to Ca2+ is dependent on temperature.  相似文献

Water relations, proline content and gas exchange of leaf were investigated under periodic water stress for two C3 plants (eggplant and tomato) in a greenhouse to study comparative adaptive responses. Although both species showed reduced water content of leaf and increased osmolality and proline content under low soil water potential, the recovery capacity after the stress was better in eggplant than tomato. Both species over-accumulated proline under low soil water potential and returned to its initial concentration during the recovery, indicating that proline may act as an osmoprotectant during drought. Proline was directly corresponding with osmolality during stress, and dehydration stress reduced the gas exchange parameters such as transpiration rate (ET), stomatal conductance (GS), and photosynthesis rate (Pn). At the final stage of the experiment both species showed 2.6 and 3.3 times lower Pn and 27 and 19 times lower GS for eggplant and tomato, respectively, as compared to control. But after stress was relieved by rewatering, both plants increased GS for 2 to 3 times and Pn for 4.5 times. Eggplant showed better water use efficiency (WUE) in relation to fruit production under the stress than tomato. Higher biomass allocation at root and fruit parts in eggplant indicated more efficient recovery than that of tomato. These findings inferred that both C3 plants developed internal complementary drought survival mechanism by lowering relative water content, increasing proline, and decreasing stomatal conductance but eggplants withstood the periodic draughting better than tomato, mainly due to its ability to recover from a water stress condition.  相似文献

Bird-pollinated flowers are known to secrete relatively dilute nectars (with concentrations averaging 20–25% w/w). Many southern African plants that are pollinated by passerine birds produce nectars with little or no sucrose. Moreover, these hexose nectars are extremely dilute (10–15%). This suggests a link between sugar composition and nectar concentration. Nectar originates from sucrose-rich phloem sap, and the proportion of monosaccharides depends on the presence and activity of invertase in the nectary. Hydrolysis of sucrose increases nectar osmolality and the resulting water influx can potentially convert a 30% sucrose nectar into a 20% hexose nectar, with a 1.56 times increase in volume. Hydrolysis may also increase the gradient for sucrose transport and thus the rate of sugar secretion. When sucrose content and refractometer data were compared, some significant correlations were seen, but the occurrence of sucrose-rich or hexose-rich nectars can also be explained on phylogenetic grounds (e.g. Erythrina and Protea). Hexose nectars may be abundant enough to drip from open flowers, but evaporation leads to much variability in nectar concentration and increases the choices available to pollinators.  相似文献

Ammonia-N toxicity to early Portunus pelagicus juveniles at different salinities was investigated along with changes to haemolymph osmolality, Na+, K+, Ca2+ and ammonia-N levels, ammonia-N excretion and gill Na+/K+-ATPase activity. Experimental crabs were acclimated to salinities 15, 30 and 45‰ for one week and 25 replicate crabs were subsequently exposed to 0, 20, 40, 60, 80, 100 and 120 mg L− 1 ammonia-N for 96-h, respectively. High ammonia-N concentrations were used to determine LC50 values while physiological measurements were conducted at lower concentrations. When crabs were exposed to ammonia-N, anterior gill Na+/K+-ATPase activity significantly increased (p < 0.05) at all salinities, while this only occurred on the posterior gills at 30‰. For crabs exposed to 20 and 40 mg L− 1 ammonia-N, both posterior gill Na+/K+-ATPase activity and ammonia-N excretion were significantly higher at 15‰ than those at 45‰. Despite this trend, the 96-h LC50 value at 15‰ (43.4 mg L− 1) was significantly lower (p < 0.05) than at both 30‰ and 45‰ (65.8 and 75.2 mg L− 1, respectively). This may be due to significantly higher (p < 0.05) haemolymph ammonia-N levels of crabs at low salinities and may similarly explain the general ammonia-N toxicity pattern to other crustacean species.  相似文献

Volume regulation in cortical collecting duct cells: role of AQP2   总被引:2,自引:0,他引:2  
BACKGROUND INFORMATION: The renal CCD (cortical collecting duct) plays a role in final volume and concentration of urine by a process that is regulated by the antidiuretic hormone, [arginine]vasopressin. This hormone induces an increase in water permeability due to the translocation of AQP2 (aquaporin 2) from the intracellular vesicles to the apical membrane of principal cells. During the transition from antidiuresis to diuresis, CCD cells are exposed to changes in environmental osmolality, and cell-volume regulation may be especially important for the maintenance of intracellular homoeostasis. Despite its importance, cell-volume regulation in CCD cells has not been widely investigated. Moreover, no studies have been carried out till date to evaluate the putative role of AQPs during this process in renal cells. RESULTS: In the present study, we have studied the regulatory cell-volume responses to hypo-osmotic or hyperosmotic challenges in two CCD cell lines: one not expressing AQPs and the other stably transfected with AQP2. We have used a fluorescent probe technique in which the acquisition of single-cell kinetic data can be simultaneously recorded with the intracellular pH. Experiments with hyperosmotic mannitol media demonstrated that, independent of AQP2 expression, CCD cells shrink but fail to show regulatory volume increase, at least under the studied conditions. In contrast, under hypo-osmotic shocks, regulatory volume decrease occurs and the activation of these mechanisms is more rapid in AQP2 transfected cells. This regulatory response takes place in parallel with intracellular acidification, which is faster in cells expressing AQP2. The acidification and the initial regulatory volume decrease response were inhibited by glibenclamide and BaCl2 only in AQP2 cells. CONCLUSIONS: These results suggest that increases in the osmotic water permeability due to the expression of AQP2 are critical for a rapid activation of regulatory volume decrease mechanisms, which would be linked to cystic fibrosis transmembrane conductance regulator and to barium-sensitive potassium channels.  相似文献

The land snail Helix pomatia (Gastropoda: Helicidae) is widely distributed in Northern and Central Europe where it may experience subzero temperatures during winter months. Its supercooling ability was studied in two populations of H. pomatia. One population originated from Southern Sweden (Gotaland) and the other from Central France (Auvergne). In the experimental design, they were acclimated, over 2 weeks, to artificial winter conditions (hibernation, T=5 degrees C). The Swedish snails showed a rather limited supercooling ability (temperature of crystallization, T(c)=-6.4+/-0.8 degrees C), significantly greater, however, than the supercooling capacity of the population from France (T(c)=-4.6+/-1.4 degrees C). In artificial spring conditions (3 months of hibernation followed by a progressive acclimation, over 2 weeks, to activity at T=20 degrees C), both populations exhibited a similar high T(c) (-2.0+/-1.0 degrees C). The lower T(c) of hibernating Swedish snails could be due to a greater loss of body water, accompanied by a higher concentration of solutes in the hemolymph. In both populations, the variation in hemolymph osmolality measured between hibernating (250-270 mOsm kg(-1)) and active (165-215 mOsm kg(-1)) snails may be explained by the variation in body water mass and did not suggest the production of colligative cryoprotectants. Moreover, the three bacterial strains, Buttiauxella sp., Kluyvera sp., and Tatumella sp. (Enterobacteriaceae) which were isolated from fed snails, but absent in starved snails, did not show any ice-nucleating activity at temperatures higher than -9 degrees C. Only the strain Kluyvera sp. initiated nucleation at -9 degrees C. This strain, therefore, is a weak, also termed a Type III or Class C ice-nucleating active bacterium, but with no influence on the supercooling ability of individual snails. In summary, fluctuations in body water mass of hibernating snail populations, triggering changes in osmolyte concentration, rather than the presence of endogenous ice-nucleating-active bacteria, accounts for fluctuations in their T(c).  相似文献
