Abstract

Cultures of filamentous fungi that secrete significant amounts of exopolysaccharides are among the most difficult of fermentation fluids, presenting difficulties in the areas of aeration, agitation, mixing, and control that may in turn impact the physiology of the microorganism in an undesirable manner. The fungus Sclerotium glucanicum, which produces a potentially useful exopolysaccharide known as scleroglucan, illustrates many such difficulties. This review discusses in detail the range of physiological studies on the producing microorganism itself, including those concerning formation of “undesirable” byproducts, principally oxalate, but also, under certain conditions, other TCA cycle acids. In addition, the bioreactor technology in use for production of this type of biopolymer is discussed in relation to the difficulties such fluid types present. The potential of pneumatically agitated reactors for such production is evaluated, and the lack of fundamental studies on such reactors and on the hydrodynamics and mixing behavior of such complex fluids is pointed out.  相似文献

In total, 366 birds representing 55 species in 24 families and eight orders, were examined for chewing lice (Phthiraptera: Amblycera, Ischnocera) in two high‐altitude localities in Yunnan Province, China. In Ailaoshan, almost all of the birds examined were resident passeriforms, of which 36% were parasitized by chewing lice. In Jinshanyakou, most birds were on migration, and included both passerine and non‐passerine birds. Of the passerine birds caught in Jinshanyakou, only one bird (0.7%) was parasitized by chewing lice. The prevalence of Myrsidea and Brueelia‐complex lice on birds caught in Ailaoshan was higher than in previous reports. Of the chewing lice identifiable to species level, three represent new records for China: Actornithophilus hoplopteri (Mjöberg, 1910), Maculinirmus ljosalfar Gustafsson & Bush, 2017 and Quadraceps sinensis Timmermann, 1954. In total, 17 new host records are included, of which we describe two as new species in the Brueelia‐complex: Guimaraesiella (Cicchinella) ailaoshanensis sp. nov. ex Schoeniparus dubius dubius (Hume, 1874) and G. (C.) montisodalis sp. nov. ex Fulvetta manipurensis tonkinensis Delacour & Jabouille, 1930. A new cumacean genus and species, Ithyleucon sorbei gen. et sp. n., was described from material collected in the southern margin of the Cap Ferret Canyon (Bay of Biscay, NE Atlantic). Although the new genus resembles Pseudoleucon Zimmer, 1903, in terms of the general aspect of the carapace and the pseudo-rostrum position, it shows important differences in the uropod structure and in the size of the antenna 1 accessory flagellum. In addition, some comments regarding the morphology of certain rare species (Mesolamprops denticulatus Ledoyer, 1983, Hemilamprops normani Bonnier, 1896 and Schizocuma spino-culatum (Jones, 1984)) are also provided.  相似文献

Closely related living mammal species present numerous difficulties of recognition and delineation, not least because populations as well as individual organisms fulfill both genetic and economic functions. However, since species are often distributed geographically across multiple ecosystems or environments, it is evident that species as wholes, except where coterminous with the local populations of which they are composed, cannot be said to play unitary ecological roles. Aspects of the economic activity of organisms thus may not be admitted as elements in species recognition. Those who study the behavior of mammals in their natural habitats must therefore focus upon behaviors which preserve the genetic integrity of species if they wish to contribute to the systematic question of species identification. «Isolation» concepts of species are not served by data of this kind, since they emphasize interspecies discontinuities; the «recognition» concept of species, however, specifically focuses upon behaviors of this kind as they contribute to the «Specific-Mate Recognition System». Among primates such behaviors are likely to involve signalling of various kinds; where this is visual it is reasonable to expect to find morphological or chromatic correlates; where such cues are auditory or, to a lesser extent, olfactory, it is less probable that such correlates will be found to exist.  相似文献

Calli were induced from 300,000 embryos isolated from immature to mature stage of seeds collected on late September from 14 elite trees. When the embryos were cultured onto plastic Petri-dish containing 20 mL of modified B5 basal medium supplemented with 3% (w/v) sucrose, 500 mg/L casein hydrolysate, 250 mg/L myo-inositol, 0.5% (w/v) polyvinyl polypyrrolidon (PVPP), 2×MS vitamins, 0.5 mg/L gibberellic acid, and 10 mg/L 2,4-D after 2 weeks of culture, yellowish-white calli were immediately formed on the surfaces of embryos, and subcultured for 4 weeks in same culture medium. Because most of calli maintained for more than 3 months were revealed differences in their colors, surface texture, and growth rate, visual selection was made for first round screening. When the size of visually selected calli larger than 19 mm in their diameter were inoculated, persistent proliferation was observed. Among the plating methods tested for the selection of rapid growing cell lines at single cell and/or small cell aggregate level, 2-layer spread plating revealed as the best for single cell cloning. To enhance cell growth and maintain high rate of viability for long-term culture of yew cells in bioreactor, final cell volume less than 50% in SCV seemed to be the best. Time course study revealed that 30% of inoculum density was suitable for fed batch culture. Among the tested conditional media, the rate of 1∶2 (old medium: fresh medium) was recorded at the best for cell growth.  相似文献
