Abiotic factors are thought to be primarily responsible for the loss of bacteriophages from the environment, but ingestion of phages by heterotrophs may also play a role in their elimination. Tetrahymena thermophila has been shown to ingest and inactivate bacteriophage T4 in co-incubation experiments. In this study, other Tetrahymena species were co-incubated with T4 with similar results. In addition, T. thermophila was shown to inactivate phages T5 and lambda in co-incubations. Several approaches, including direct visualization by electron microscopy, demonstrated that ingestion is required for T4 inactivation. Mucocysts were shown to have no role in the ingestion of T4. When (35)S-labeled T4 were fed to T. thermophila in a pulse-chase experiment, the degradation of two putative capsid proteins, gp23(*) and hoc, was observed. In addition, a polypeptide with the apparent molecular mass of 52 kDa was synthesized. This suggests that Tetrahymena can use phages as a minor nutrient source in the absence of bacteria.  相似文献

ABSTRACT. By use of a monoclonal antibody directed against purified lectin from the sponge Geodia cydonium it was demonstrated that the mucocysts of Tetrahymena pyriformis contain a substance immunologically similar to that found in G. cydonium . In extracts of T. pyriformis the monoclonal antibody recognizes a 36 kDa protein; binding could be abolished by adsorption of the antibody with (i) crude extract, (ii) purified lectin from G. cydonium and (iii) a 29 aa long peptide. In addition the data show that 10-6 M of insulin causes first the release of mucocyst material, which reacts with the lectin antibody, and second its subsequent redistribution on the surface of the somatic cilia and the oral field.  相似文献

The characteristic stalk of Colacium calvum Stein is formed from carbohydrate extruded by granules in the anterior portion of the cell. These granules exhibit the characteristics of mucocysts and their contents have two electron densities. A narrow extension of each granule extends past neighboring muciferous bodies to attachment sites in the posterior notch of the pellicle strip. The granule contains neutral polysaccharide which resembles that in the peripheral cisternae of the Golgi apparatus. The attachment site is structurally complex.  相似文献

Even though many aspects of Dictyocha fibula and D. speculum have long been studied, very little is known about D. octonaria. For the first time a clonal culture derived from a single cell of D. octonaria from Wellington Harbour was studied in detail. In the skeleton‐bearing stage three morphotypes were observed – skeleton bearing, mucocyst‐bearing and amoeboid, while in the naked stage only the naked form was studied. In this study the mucocyst‐bearing form was described as a new morphotype. Vegetative reproduction of the skeleton‐bearing form in the exponential growth phase was by both direct binary fission and by first forming a doublet and then two separate daughter cells, while that of naked form was by simple binary fission. Occasionally double skeletons were observed as end products of both the vegetative and sexual reproductions. In sexual reproduction all three forms in the skeleton‐bearing stage exhibited the same polymorphic life history involving a multinucleate stage. The newly formed daughter cells of all three forms developed individual siliceous skeletons prior to being released from the parent cell. The naked form in the naked stage, however, exhibited a separate polymorphic life history that produced only skeleton‐free daughter cells. For the first time both vegetative and sexual reproduction of D. octonaria were documented.  相似文献

Euglena viridis (subgenus Euglena) serves as the type species for the genus Euglena. In this study, molecular phylogenetic analyses using a small subunit (SSU) and a combined SSU–partial large subunit rDNA data set for members of the genus Euglena showed that strains identified as E. viridis on the basis of morphology are distributed between two separate nonsister clades. Although all the E. viridis strains examined were morphologically indistinguishable and possessed spherical mucocysts and stellate chloroplasts with one paramylon center, there was a high degree of sequence divergence between the E. viridis strains in different clades, making this a cryptic species. Like E. viridis, all taxa from the subgenus Euglena are characterized by having one or more stellate chloroplasts with paramylon grains clustered around the center of the chloroplast. These additional taxa were divided into four clades in all the molecular analyses. Strains of Euglena stellata formed two nonsister clades whose members had a single aggregate chloroplast with paramylon center and spindle‐shaped mucocysts. A geniculata clade included species with one or two stellate chloroplasts with paramylon centers and spherical mucocysts, and the cantabrica clade had members with one stellate chloroplast with paramylon center and spherical mucocysts often arranged in spiral rows. Interspersed among these were three additional clades bearing taxa from the subgenus Calliglena that contains members with discoid plastids and pyrenoids that may or may not be capped with paramylon. These taxa formed a laciniata clade, mutabilis clade, and gracilis clade. This study demonstrates that E. viridis and E. stellata are cryptic species that can only be distinguished at the molecular level. Because E. viridis is the designated type species for the genus Euglena, we designated an epitype for E. viridis.  相似文献

Under appropriate conditions, Alcian Blue-induced exocytosis of Tetrahymena mucocysts leads to formation of a capsule that surrounds the cell. This phenomenon is an example of regulated secretion, a mechanism of fundamental significance in eukaryotic cells. In order to dissect genetically the mechanism of mucocyst biogenesis and regulated exocytosis, mutants unable to form capsules (Caps–) were isolated. In this paper we report a genetic characterization of Caps– mutants in this collection. The mutations in mutants SB255 and SB281 behave as single recessive Men-delian mutations. The mutation in SB251 is restricted to the macronucleus, and could not be further characterized by the genetic methods we used. Complementation tests suggest the existence of at least 2 genes, named exoA and exoB; additional mutant loci are likely to be included in the mutant collection. Deletion mapping using nulli-somic strains showed that exoA and exoB are located on the left arm of chromosome 4. The exo-3 mutation, which behaves as recessive and complements with exoA1 in SB255 and exoB2 in SB281, maps to chromosome 3. These Caps– mutants may be useful for the elucidation of the developmental pathway of mucocyst biogenesis and the control of regulated secretion in eukaryotic cells. © 1992 Wiley-Liss, Inc.  相似文献
