An experiment is described in which the magnitude of N transferred from damaged white clover roots to perennial ryegrass was determined, using 15N labelling of the grass plant. There was no effect on the growth and N-fixation of the clover plants after removing part of the root system. The 15N data suggested that N had been acquired by all grass plants, even in plants grown alone with no further N supplied after labelling. However, after quantifying the mobile and stored N pools of the grass plants it was evident that significant transfer of N from clover to grass only took place from damaged clover roots. Dilution of the atom% 15N in the roots of the grass plants grown alone, and in association with undamaged clover roots, was explained by remobilisation of N within the plant. 相似文献
Left-handed polyproline II (PPII) helices commonly occur in globular proteins in segments of 4-8 residues. This paper analyzes the structural conservation of PPII-helices in 3 protein families: serine proteinases, aspartic proteinases, and immunoglobulin constant domains. Calculations of the number of conserved segments based on structural alignment of homologous molecules yielded similar results for the PPII-helices, the alpha-helices, and the beta-strands. The PPII-helices are consistently conserved at the level of 100-80% in the proteins with sequence identity above 20% and RMS deviation of structure alignments below 3.0 A. The most structurally important PPII segments are conserved below this level of sequence identity. These results suggest that the PPII-helices, in addition to the other 2 secondary structure classes, should be identified as part of structurally conserved regions in proteins. This is supported by similar values for the local RMS deviations of the aligned segments for the structural classes of PPII-helices, alpha-helices, and beta-strands. The PPII-helices are shown to participate in supersecondary elements such as PPII-helix/alpha-helix. The conservation of PPII-helices depends on the conservation of a supersecondary element as a whole. PPII-helices also form links, possibly flexible, in the interdomain regions. The role of the PPII-helices in model building by homology is 2-fold; they serve as additional conserved elements in the structure allowing improvement of the accuracy of a model and provide correct chain geometry for modeling of the segments equivalenced to them in a target sequence. The in vitro uptake of zinc by erythrocytes was measured under near-physiological conditions, using65Zn as a radioactive tracer. Because of the presence of serum albumin—a strong zinc ligand—a low concentration of medium free
zinc was maintained. Under these conditions a high-affinity carrier for zinc transport over the cell membrane was identified.
With human erythrocytes, a Michaelis constant (Km) of 0.2 nM with respect to free medium zinc was measured and aVmax of 4.5 nmoles Zn transported per h/g dry wt. TheKm for medium Zn increases when the size of the internal erythrocytic Zn pool is augmented, whereasVmax remains virtually unchanged. A model to explain this phenomenon is proposed. It is suggested that this phenomenon could underlie
observations, confirmed here, that the in vitro uptake of Zn by animal erythrocytes depends on the Zn status of the animal. 相似文献
The Mutator transposable element system is exceptional in many of its basic attributes. The high frequency and low specificity of mutant induction are both unusual and useful characteristics of the Mutator system. Other basic features are at least equally fascinating: the existence of multiple Mu element subfamilies with apparently unrelated internal sequences; the lack of correlation between Mu element transposition and excision; the complex inheritance of Mutator activity; the tight developmental regulation of Afufaror‐conditioned events; and the coordinated processes of element modification/inactivation, to name a few.
Molecular and genetic studies over the last 10 years have begun to explain many of these interesting properties and have uncovered new mysteries of Mutator biology. Both positive and negative regulators of the system have been identified and characterized to varying degrees. Insertion specificity has been observed at several levels. Recent accomplishments include the isolation of an autonomous Mu element and the discovery of maize lines with altered developmental regulation of Mutator‐derived mutability. This review defines the Mutator system, describes the status of current experimentation in the Mutator field, proposes models that may explain some aspects of Mutator behavior, and details future studies that will help elucidate the nature of the Mutator phenomenon. 相似文献
Inner ear disorders' treatment remains challenging due to anatomical barriers. Robotic assistance seems to be a promising approach to enhance inner ear treatments and, more particularly, lead to effective targeted drug delivery into the human cochlea. In this paper we present a combination of a micro-macro system that was designed and realized in order to efficiently control the navigation of magnetic nanoparticles in an open-loop scheme throughout the cochlea, considering that the magnetic particles cannot be located in real time.In order to respect the anatomical constraints, we established the characteristics that the new platform must present then proceeded to the design of the latter. The developed system is composed of a magnetic actuator that aims to guide nanoparticles into the cochlea. Mounted on a robotic manipulator, it ensures its positioning around the patient's head. The magnetic device integrates four parallelepiped-rectangle permanent magnets. Their arrangement in space, position and orientation, allows the creation of an area of convergence of magnetic forces where nanoparticles can be pushed/pulled to. To ensure the reachability of the desired orientations and positions, a 3 DOF robot based on a Remote Centre of Motion (RCM) mechanism was developed. It features three concurrent rotational joints that generate a spherical workspace around the head. The control of the latter is based on kinematic models.A prototype of this platform was realized to validate the actuation process. Both magnetic actuator and robotic manipulator were realized using an additive manufacturing approach. We also designed a virtual human head with a life-size cochlea inside. A laser was mounted on the end effector to track the positioning of the actuator. This permitted to experimentally prove the capacity of the robotic system to reach the desired positions and orientations in accordance with the medical needs.This promising robotic approach, makes it possible to overcome anatomical barriers and steer magnetic nanoparticles to a targeted location in the inner ear and, more precisely, inside the cochlea. 相似文献
Elucidating the factors underlying the origin and maintenance of genetic variation among populations is crucial for our understanding of their ecology and evolution, and also to help identify conservation priorities. While intrinsic movement has been hypothesized as the major determinant of population genetic structuring in abundant vagile species, growing evidence indicates that vagility does not always predict genetic differentiation. However, identifying the determinants of genetic structuring can be challenging, and these are largely unknown for most vagile species. Although, in principle, levels of gene flow can be inferred from neutral allele frequency divergence among populations, underlying assumptions may be unrealistic. Moreover, molecular studies have suggested that contemporary gene flow has often not overridden historical influences on population genetic structure, which indicates potential inadequacies of any interpretations that fail to consider the influence of history in shaping that structure. This exhaustive review of the theoretical and empirical literature investigates the determinants of population genetic differentiation using seabirds as a model system for vagile taxa. Seabirds provide a tractable group within which to identify the determinants of genetic differentiation, given their widespread distribution in marine habitats and an abundance of ecological and genetic studies conducted on this group. Herein we evaluate mitochondrial DNA (mtDNA) variation in 73 seabird species. Lack of mutation–drift equilibrium observed in 19% of species coincided with lower estimates of genetic differentiation, suggesting that dynamic demographic histories can often lead to erroneous interpretations of contemporary gene flow, even in vagile species. Presence of land across the species sampling range, or sampling of breeding colonies representing ice‐free Pleistocene refuge zones, appear to be associated with genetic differentiation in Tropical and Southern Temperate species, respectively, indicating that long‐term barriers and persistence of populations are important for their genetic structuring. Conversely, biotic factors commonly considered to influence population genetic structure, such as spatial segregation during foraging, were inconsistently associated with population genetic differentiation. In light of these results, we recommend that genetic studies should consider potential historical events when identifying determinants of genetic differentiation among populations to avoid overestimating the role of contemporary factors, even for highly vagile taxa. 相似文献
