Summary A cDNA encoding trypsin inhibitor CMe from barley endosperm has been cloned and characterized. The longest open reading frame of the cloned cDNA codes for a typical signal peptide of 24 residues followed by a sequence which is identical to the known amino acid sequence of the inhibitor, except for an Ile/Leu substitution at position 59. Southern blot analysis of wheat-barley addition lines has shown that chromosome 3H of barley carries the gene for CMe. This protein is present at less than 2%–3% of the wild-type amount in the mature endosperm of the mutant Risø 1508 with respect to Bomi barley, from which it has been derived, and the corresponding steady state levels of the CMe mRNA are about I%. One or two copies of the CMe gene (synonym Itc1) per haploid genome have been estimated both in the wild type and in the mutant, and DNA restriction patterns are identical in both stocks, so neither a change in copy number nor a major rearrangement of the structural gene account for the markedly decreased expression. The mutation at the lys 3a locus in Risø 1508 has been previously mapped in chromosome 7 (synonym 5H). A single dose of the wild-type allele at this locus (Lys 3a) restores the expression of gene CMe (allele CMe-1) in chromosome 3H to normal levels.  相似文献

Summary A locus associated with a severe Minute effect has been mapped at 7C on the X chromosome of Drosophila melanogaster. Previous work has suggested that this Minute encodes ribosomal proteins S14A and S141B. We have made a chromosomal deficiency that removes the S14 ribosomal protein genes, yet does not display the Minute phenotype. These data suggest that the S14 genes do not actually correspond to the Minute locus.  相似文献

Summary The c2 locus of Zea mays, identified as one of the genes affecting anthocyanin biosynthesis, was cloned using the transposable element En (Spm) as a gene tag. The Spm element present at the c2 locus in the autonomously mutating c2-m1 line was isolated using En1 element specific probes. Sequences flanking the element were identified as c2 locus specific and were used to clone the nonautonomous c2-m2 and wild-type alleles. The cloning and analysis of a cDNA complementary to the c2 locus provided evidence that this gene encodes the enzyme chalcone synthase.  相似文献

Summary Under controlled growth chamber conditions of 30 °C, seed set after selfing is possible in normally self-incompatible rye plants. Within selfed progenies produced by this method, plants homozygous at the peroxidase isozyme locus Prx 7 were crossed to heterozygous individuals. Segregation at the Prx 7 locus in progenies of these crosses provides clear evidence of a close linkage between Prx 7 and one of the two incompatibility loci in rye. A recombination fraction in the range of 0–2% was calculated from the segregation data. In rye, Prx 7 is linked with a phosphoglucoisomerase locus (Pgi). The similarity between the observations in Secale cereale and those made in Lolium perenne is discussed.  相似文献

在72只乌拉坦、氯醛糖麻醉兔,静脉注射三碘季铵酚后,在人工呼吸下进行实验。结果观察到,在家兔蓝斑复合核(Lo-So)区微量注射去甲肾上腺素(NE)或可乐宁(Clonidine)能减少刺激下丘脑诱发的室性期前收缩(HVE)数。而微量注射β-肾上腺素能受体阻断剂心得安、α_1肾上腺素激动剂甲氧胺(Methoxamine)或苯肾上腺素(Phenylephrine)、α肾上腺素能阻断剂育亨宾(Yohimbine)或阿片受体阻断剂纳洛酮则对 HVE 无明显影响。在 Lo-So 区微量注射 NE 对 HVE 的抑制效应不能被事先在该区注射心得安所阻断但能被事先静脉或 Lo-So 区注射育亨宾所阻断。NE 或可乐宁的抑制效应也均可被育亨宾所翻转。可乐宁的抑制效应还可被纳洛酮所翻转。电解损毁延髓中缝大核区可消除在 Lo-So 区微量注射 NE 或可乐宁对HVE 的抑制效应。上述结果提示,Lo-So 区α_2受体的兴奋可减少 HVE 数,这些作用可能依赖于延髓中缝大核区的完整。可乐宁对 HVE 的抑制作用可能有阿片受体的参与。  相似文献

Summary Recently, the existence of a vasopressin-immunoreactive cell group was described in the bed nucleus of the stria terminalis (van Leeuwen and Caffé 1983). In the present investigation additional nuclei containing vasopressin-immunoreactive cells were found, after colchicine pretreatment, in the dorsomedial hypothalamus, medial amygdaloid nucleus and the locus coeruleus.Vasopressin-immunoreactive cells in the dorsomedial hypothalamus and medial amygdaloid nucleus are small (8–14 m and 10–14 m, respectively), while those in the locus coeruleus are medium-sized (20–25 m). Incubation with anti-bovine neurophysin II and anti-rat neurophysin revealed staining of the same cell group in the above-mentioned areas. None of these cell groups show stained cells after incubation with anti-oxytocin and anti-bovine neurophysin I. When sections of the homozygous Brattleboro rat, which shows a deficiency in vasopressin synthesis, are incubated with anti-vasopressin, anti-bovine neurophysin II, or anti-rat neurophysin, no immunoreactivity can be observed in these brain regions.The above-mentioned cell groups may contribute to the vasopressinergic innervation of brain sites that have been reported to persist after lesioning of the suprachiasmatic, paraventricular and bed nuclei of the stria terminalis.  相似文献

Summary The locus coeruleus of cat is populated by two types of neurons: medium sized ones, with plump cell bodies and relatively short dendrites; and small ones, with triangular bodies and relatively long dendrites. The former type is regarded here as typical of the centre, whereas the second type could simply represent displaced neurons from the adjacent griseum centrale. Electron microscopy failed to reveal any outstanding richness in pigment granules in kittens up to five weeks old. Very characteristic somatic appendages were found, mostly in the medium sized neurons. These somatic spines communicate with the perikaryon by means of a narrow neck region. A complex, multilayered, glial sheath surrounds the cells. This glial sheath is pierced by the somatic appendages, which are not surrounded by glia and make contact with axonal knobs. Typical dendritic spines appear to be absent. Axodendritic synapses are made on medium sized dendritic trunks. By and large, most of the synaptic vesicles present in the centre are of the small, clear-centered type. However, dense core vesicles extremely variegated in size and appearance were found, both in presynaptic and postsynaptic profiles. The possibility that dense core vesicles should be regarded as atypical lysosomes rich in by-products of the metabolism of catecholamines (melanine) has been considered.Supported by grant MA 4183 from the Medical Research Council of Canada.  相似文献

The locus coeruleus (LC) of the rat was lesioned by microinjection of selective neurotoxins into the brainstem. 6-Hydroxydopamine (6-OHDA), 3 micrograms/microliter, given unilaterally at two sites 0.6 mm apart on the rostro-caudal axis of the LC, was used to lesion catecholamine-containing neuronal elements. Ibotenic acid, 2.5 micrograms/0.5 microliters, administered similarly was used to lesion nerve cell bodies. Two weeks after administration of the neurotoxin, lesion efficacy was determined based on the norepinephrine content of the cerebral cortex ipsi- and contralateral to the lesion. 6-OHDA lesions of the LC caused a 46% reduction in ipsilateral cortical norepinephrine and a 60% reduction in specific 125I-[Sar1, Ile8]-angiotensin II (125I-SIAII) binding in the LC. Ibotenic acid lesions of the LC caused a 73% reduction in ipsilateral cortical norepinephrine and a 81% reduction in specific 125I-SIAII binding in the LC. These results indicate that AII receptor binding sites in the LC are localized on noradrenergic nerve cell bodies or their dendritic and axonal ramifications within the LC.  相似文献

Summary A diploid Allium cepa plant was recovered from the backcross of an interspecific triploid (2 x A. cepa + 1 x A. fistulosum) to an A. cepa diploid which exhibited both A. cepa and A. fistulosum Adh-1 alleles. Cytogenetic analyses revealed a recombinant sub-telocentric chromosome. The ADH-1 locus is believed to be on the long arm of the sub-telocentric A. fistulosum chromosome 5. Meiosis of the triploid progenitor gives strong evidence that recombination occurred.
