Five locally isolated bacterial strains produced extracellular cellulase enzymes, primarily CMCase, when grown on different natural and commercial cellulosic substrates. Extracellular CMCase and avicelase activity was higher with the strain CLS-32, a Cytophaga sp., compared to four other strains. The whole-cell preparations of these isolates were found to saccharify cellulosic substrates to reducing sugars. Maximum release of reducing sugar (5.75 mg ml−1) was obtained with CLS-32 using sugar cane bagasse as growth and hydrolysis substrates.  相似文献

Polysaccharidases adsorbed on commercial amylodextrins were added to environmentally controlled composts of straw plus poultry manure. After 5 days of composting at 48°C, microbial enzyme activities and numbers of bacteria were higher in the treated compost than in the control. During the next phase at 80°C, between days 5 and 6, more C and N were solubilized in the treated compost. After introducing a microbial inoculum on day 6, and maintaining the substrate at 48°C, colonization by bacteria was faster in the treated compost and consequently, more fibre was degraded. Correlating the effect of pretreatment on the enzymatic hydrolysis of straw   总被引:4,自引:0,他引:4  
Avicell, Alkali-treated straw cellulose (ATSC), and wheat straw were ball-milled to reduce crystallinity; wheat straw was delignified by hot (120 degrees C) sodium hydroxide solutions of various concentrations. The physically and chemically pretreated cellulosic materials were hydrolyzed by the cellulases of Fusarium oxysporum strain F3. Enzymic hydrolysis data were fitted by the hyperbolic correlation of Holtzapple, which involves two kinetic parameters, the maximum conversion (X(max)), and the enzymic hydrolysis time corresponding to 50% of X(max) (t(1/2)). An empirical correlation between X(max) and cellulose crystallinity, lignin content, and degree of delignification has been found under our experimental conditions. Complete cellulose hydrolysis is shown to be possible at less than 60% crystallinity indices or less than 10% lignin content.  相似文献

Mild alkaline/oxidative pretreatment of wheat straw   总被引:4,自引:0,他引:4  
A new mild alkaline/oxidative pretreatment of wheat straw prior to enzymic hydrolysis was carried out. It consists of a first alkaline (1% NaOH for 24 h) step, which mainly solubilises hemicellullose and renders the material more accessible to further chemical attack, and a second alkaline/oxidative step (1% NaOH and 0·3% H2O2 for 24 h), which solubilises and oxidises lignin to minor polluting compounds. The entire process was carried out at low temperature (25–40°C) using a low concentration of chemicals, resulting in a relatively low cost and waste liquors containing only trace amounts of dangerous pollutants derived from lignin. Recovery of cellulose after the double pretreatment reached 90% of that contained in the starting material, with a concomitant 81% degradation of lignin. The action of a commercial cellulase on the cellulose obtained produced a syrup with a high concentration of reducing sugars (220 mg/ml), of which a large percentage was glucose.  相似文献

The lignocellulosic biomass from coconut palm (Cocos nucifera Linn.) such as bunch waste (spathe+spadices), leafstalk (petiole), leaflets and coir pith (by-product from coir processing industry) were evaluated as substrates for cultivation of oyster mushroom, Pleurotus sajor-caju (Fr.) Singer. A low-cost mushroom shed built exclusively of coconut materials such as coconut wood and plaited coconut leaves inside a coconut plantation was used for spawn run and cropping. Leafstalk and bunch waste were superior to leaflets and coir pith in producing significantly more edible biomass of mushrooms. Biological efficiency of 58.9% was obtained in leafstalk, followed by bunch waste (56.9%), coir pith (39.7%) and leaflets (38.2%). The yield of sporophore was positively related to cellulose content and the cellulose : lignin ratio of the substrates.  相似文献

Two strains of Pleurotus spp., grown in solid state fermentation on sugar-cane straw, degraded the dry matter by 50% after 60 days. The rate of substrate consumption and the dry weight of fruiting bodies decreased in consecutive flushings. Both strains vigorously attacked hemicellulose (80% of total degradation) and lignin (70%). Fruiting bodies were rich in protein and lipids, and had a low content of carbohydrates and ash.  相似文献

Aims: Improvement of cellulase production of Penicillium decumbens by genome shuffling of an industrial catabolite‐repression‐resistant strain JU‐A10 with its mutants. Methods and Results: After two rounds of genome shuffling, three fusants, GS2‐15, GS2‐21 and GS2‐22, were obtained, showing 100%, 109% and 94% increase in FPase activity than JU‐A10 respectively. The cellulase production of the fusants on various substrates, such as corn stover, wheat straw, bagasse and the corncob residue, was studied. The maximum productivities of GS2‐15, GS2‐21 and GS2‐22 were 92·15, 102·63 and 92·35 FPU l?1 h?1 on the corncob residue at 44 h respectively, which were 117%, 142% and 118% higher than that of JU‐A10 (42·44 FPU l?1 h?1, at 90 h). The improvements of the fusants were possibly because of their enhanced growth rates, earlier cellulase synthesis and higher secretion of extracellular proteins. Conclusions: The fusants obtained after genome shuffling could produce abundant cellulase much earlier, and they could be potential candidates for bioconversion process. Significance and Impact of the Study: This is the first report on the improvement of cellulase production in fungi by genome shuffling, and this is a good technique to improve other important phenotypes in fungi.  相似文献

An original reaction system (the phase separative reaction system) has been designed for derivatizing native lignins to highly phenolic, functional polymers. This system is composed of a phenol derivative and concentrated acid, which are not miscible at room temperature. The key point of the lignin functionalization process, including the phase separative system, is that lignin and carbohdrates, which are totally different in structures and reactivitie, are modified individually in the different phases: lignin is present in the organic phase and carbohydrates in the aqueous phase. Through the process, lignin was modified selectively at Calpha-positions of side chains, the most reactive sites, to give highly phenolic, light-colored, diphenylmethane-type materials which still retained original interunit linkages formed by the dehydrogenative polymerization during the biosynthesis. The carbohydrates were swollen, followed by partial hydrolysis and dissolution in the acid solution, resulting in the perfect decomposition of interpenetrating polymer network structures in the cell wall. Therefore, the functionalization of lignin and the separation of resulting lignin from carbohydrates were quickly achieved at room temperature, independent of wood species. This process would be a powerful tool for estimating strutures and reactivities of lignins as well as the functionalization of lignins, because of the selective structural modifications. The actinomycete strain Streptomyces griseus B1 isolated from soil, when grown on cellulose powder as submerged culture produced high levels of all the three components i.e. filter paper lyase (FPase), CMCellulase and β-glucosidase of the cellulolytic enzyme system. FP activity and CMCellulase were present only extracellularly, while β-glucosidase was both intra- and extra-cellular. It produced highest FPase activity when grown on hardwood powder under submerged culture. It was unable to use lignin monomers (ferulic acid, vanillic acid and syringic acid) as carbon source. While growing on hardwood and softwood powders under solid-state conditions, it depleted them of cellulose (36.3 in the case of softwood and 14.4 in the case of hardwood). It also caused partial loss of lignin content in both the substrates by solubilizing them. These solubilized lignins could be recovered as acid precipitable polymeric lignins (APPL) from extracts of wood powders upon acidification. Extracts of inoculated wood powders yielded higher amounts of APPL than uninoculated controls. Also, the APPLs from Streptomyces-treated wood powders differed from control APPLs in their molecular weight distribution, as observed from their elution pattern using Sephadex G-100.  相似文献
