The bovine leukemia virus (BLV) DNA harbored in the bovine tumor cell genome was cloned in lambda Charon 4A phage. Using either representative or 3' half-enriched BLV cDNA as a blot hybridization probe, clone lambda BLV-1 was shown to carry 9 kb of the BLV genome, flanked by cellular sequences at both ends. Restriction mapping with twelve endonucleases and hybridization of the DNA fragments to BLV cDNA representing a 3'-end portion of the viral genome revealed the presence and precise location of two long terminal repeats (LTRs) and virus-cell junctions. Thus, lambda BLV-1 appears to contain the complete BLV genome and flanking tumor cellular sequences. The restriction map of the cloned BLV proviral DNA closely resembles that previously reported for unintegrated linear proviral DNA, but differs significantly from that of the integrated provirus of another BLV isolate, the difference occurring preferentially in the putative gag and pol genes.  相似文献

Intramuscular fat (IMF) % contributes positively to the juiciness and flavour of lamb and is therefore a useful indicator of eating quality. A rapid, non-destructive method of IMF determination like computed tomography (CT) would enable pre-sorting of carcasses based on IMF% and potential eating quality. Given the loin muscle (longissimus lumborum) is easy to sample, a single measurement at this site would be useful, providing is correlates well to other muscles. To determine the ability of CT to predict IMF%, this study used 400 animals and examined 5 muscles from three sections of the carcass: from the fore-section the m. supraspinatus and m. infraspinatus, from the saddle-section the m. longissimus lumborum and from the hind-section the m. semimembranosus and m. semitendinosus. The average CT pixel density of muscle was negatively associated with IMF% and can be used to predict IMF% although precision in this study was poor. The ability of CT to predict IMF% was greatest in the m. longissimus lumborum (slope −0.07) and smallest in the m. infraspinatus (slope −0.02). The correlation coefficients of IMF% between the five muscles were variable, with the highest correlation coefficients evident between muscles of the fore section (0.67 between the m. supraspinatus and the m. infraspinatus) and the weakest correlations were between the muscle of the fore and hind section. The correlation between the m. longissimus lumborum to the other muscles was fairly consistent with values ranging between 0.34 and 0.40 (partial correlation coefficient). The correlation between the proportion of carcass fat and the IMF% of the five muscles varied and was greatest in the m. longissimus lumborum (0.41).  相似文献

AIMS: The aim of this research was to compare excision sampling with polyurethane and cellulose acetate sponge swabbing for the recovery of total viable counts and Enterobacteriaceae on meat carcasses. METHODS AND RESULTS: Two sample types were used to compare the methods: (i) individual samples, taken from four sites on each carcass and (ii) composite samples, created by pooling the samples from four sites from an additional set of carcasses. When the polyurethane sponge and excision method were compared for individual sites, there were no significant differences in bacterial recovery on beef and pork carcasses and on two of four sites on lamb carcasses. However, when samples from each site were pooled, the excision method was more efficient than either swabbing method across the three animal species. CONCLUSIONS: Sampling using the polyurethane sponge represents an equivalent alternative method to excision for the bacteriological sampling of carcass surfaces which is nondestructive and less labour intensive. SIGNIFICANCE AND IMPACT OF THE STUDY: This study provides the scientific basis for using sponge swabbing instead of excision in compliance with 2001/471/EC.  相似文献

Twenty Assaf lambs fed barley straw plus a concentrate alone (CONTROL group) or enriched with naringin (1.5 g/kg DM, NARINGIN group) were used to assess the effect of this polyphenolic compound on meat quality attributes. Serum samples were collected for 7 weeks, then the animals were slaughtered and the livers and longissimus thoracis et lumborum muscles extracted for analysis. Triacylglycerol levels in the serum samples tended to show (P = 0.087) lower average values for the NARINGIN group when compared with the CONTROL, but no differences were observed when the meat was analysed for the intramuscular fat content. Lower thiobarbituric acid-reactive substances procedure (TBARS) values (P < 0.001) in the liver of the NARINGIN group were detected, probably as a consequence of naringenin accumulation in this organ. No significant differences were observed in the meat samples concerning TBARS or colour evolution during refrigerated storage, as not enough naringenin would have reached the muscle. Independent of naringin administration, the low levels of the most atherogenic oxysterols must be highlighted as the most important quality score in the lamb meat samples studied.  相似文献

The present study was designed to describe the effects of early feed restriction of Merino lambs on feed efficiency during the fattening period by examining ruminal microbiota and fermentation parameters, gastrointestinal morphology, digestibility or liver proteome. In total, 24 male Merino lambs were randomly assigned to two experimental treatments (n=12 per treatment). Lambs of the first group (ad libitum (ADL)) were kept permanently with the dams, whereas the other 12 lambs (restricted (RES)) were milk restricted. When lambs reached a live BW (LBW) of 15 kg, all the animals were offered the same complete pelleted diet (35 g dry matter/kg LBW per day) until slaughter at a LBW of 27 kg. The RES lambs showed poorer feed efficiency during the fattening period when compared with the ADL group (feed to gain ratio, 3.69 v. 3.05, P<0.001). No differences were observed in ruminal microbiota, fermentation parameters or apparent digestibility. However, the proportion of the small intestine and the length of ileal villi were reduced in the RES lambs. In total, 26 spots/proteins were identified in the liver proteomic profile, with significant differences (P<0.05) between experimental treatments, suggesting a higher catabolism of proteins and a reduction in β-oxidation of fatty acids in RES lambs when compared with the ADL animals. In conclusion, early feed restriction of Merino lambs during the suckling period promotes long-term effects on the small intestine and the proteomic profile of the liver, which may influence the metabolic use of nutrients, thus negatively affecting feed efficiency during the fattening phase.  相似文献

The aim of this experiment was to study the effect of total replacement of oat hay by rosemary distillation residues (RR) on growth, carcass characteristics and meat quality of Barbarine lambs. A total of 21 lambs were divided into three groups. The control group (C) was offered 600 g of oat hay; the RR87 and RR60 groups received 600 g of pellets containing 87% and 60% of RR, respectively. The CP content was 9% and 14% for RR87 and RR60, respectively. All animals were supplemented by 600 g of concentrate. After 77 days of fattening, lambs were slaughtered. The DM and CP intakes were significantly increased with RR diets. The average daily gain was higher (P<0.001), while the feed conversion rate was lower for RR60 and RR87 than C group. The dressing percentage was similar for all groups. The tissular (muscle, fat and bone) and the regional (leg, shoulder, etc.) carcass composition did not differ among groups. The bony organs and gut weights were similar among groups, while functional ones (skin, liver, kidney and testicles) were significantly heavier for both RR groups than control. The ultimate pH, water cooking loss and color variables were similar among groups and the chemical composition (protein, fat, myoglobin, collagen and iron) did not differ also among groups. These results revealed the opportunity of RR use in fattening lambs without adverse effects on carcass and meat characteristics. Moreover, 9% CP in RR pellets are enough given the same growth performance recorded as that of RR with 14% CP.  相似文献

Argali sheep (Ovis ammon) are the world's largest wild sheep and are threatened throughout their range in Mongolia. Little is known about the ecology of this highly cursorial species. This project was initiated to develop safe capture techniques in order to learn more about the ecology of argali in the Ikh Nartiin Chuluun Nature Reserve in southeastern Mongolia by using radiotelemetry. To our knowledge no one had ever successfully live‐caught a free‐ranging argali. We developed three techniques for capturing argali. From 2003 to 2007 we hand captured 65 argali lambs, losing one (our first capture) to maternal neglect because we stayed too long in the area and a second that was euthanized after being accidentally injured by a horse. Between 2000 and 2003 we captured six argali by darting, losing one ewe to an abdominal infection after a misplaced dart. Finally, between 2002 and 2007 we captured 32 argali by drive‐netting. We lost one ewe to over‐exertion after a long drive. The remaining 31 animals captured by drive‐netting did not appear to suffer any immediate effects (still alive in 30 days). Interactions between wild carnivore abundance and landscape composition in agricultural landscapes are poorly understood despite their importance to both production and conservation. In Australia, introduced red foxes (Vulpes vulpes) prey on both native species and lambs in the temperate agricultural regions. Historically these areas were extensively cleared of native vegetation. Recently revegetation programmes have been implemented, but there is concern that this may benefit foxes and hence increase their impacts. We used an artificial prey placed in eight revegetated (6–12% cover) and 10 cleared (0–1.5% cover) landscapes of ~700 ha to assess how these landscapes influenced fox predation rates. In June and August 2006 (winter) when we expect fox populations to be relatively stable following juvenile dispersal, predation rates were 1.5–2 times higher in revegetated landscapes than in cleared landscapes. We found no evidence of microhabitat effects on predation rates suggesting these landscape‐level differences were probably due to differences in fox population density. In April 2007 (autumn) the results were more variable, possibly indicating more transient populations including dispersing juveniles at this time. Our results suggest that the impact of foxes on highly vulnerable prey could increase with revegetation. However, the benefits of revegetation to prey may offset negative impacts of foxes and future work is required to assess the likely net effects.  相似文献

选择广东信宜、福建武平、广西岑溪3个不同磷效率特性的马尾松种源,构建同质和异质两种养分环境开展盆栽实验,研究马尾松搜寻利用异质分布养分的获取机制及不同种源觅养行为差异。结果表明,与同质营养环境相比,异质营养环境中马尾松种源具有较高的苗高、地径生长量、较强光合速率和干物质生产能力。研究证实了根系形态可塑性和生理可塑性在马尾松获取异质分布养分中的重要性。马尾松可通过在富养斑块中须侧根的大量增生、对N、P、K等元素的有效吸收提高其觅养能力。马尾松在拓殖富养斑块的初期主要依靠新生侧根的增加和侧根的延长,在拓殖一段时期之后则主要靠新生侧根的生成和须根数量、须根密度的增加来搜寻异质分布养分。异质养分环境中的根系具有较高的养分吸收效率主要缘由在富养斑块中对N、P、K大量的吸收。综合比较分析认为,在3个参试种源中广西岑溪和福建武平种源在异质营养环境中拓殖富养斑块和觅养能力较强,广东信宜种源拓殖和觅养能力相对较弱。  相似文献

A trial was conducted to examine the effect of cinnamon essential oil supplementation on lamb growth performance and meat quality. Sixteen male lambs were randomly assigned to two groups. The first group served as control and was given a basal diet, and the second group was given the same diet supplemented with cinnamon oil (1 ml/kg of concentrated feed) for 35 days. Incorporation of cinnamon oil did not affect growth performance (P>0.05). Meat pH, colour, water-holding capacity, shear force, intramuscular fat and lipid oxidation values of longissimus thoracis muscle were not significantly influenced by cinnamon oil supplementation (P>0.05). The post-inoculation counts of Salmonella enteritidis and Listeria monocytogenes on raw meat during refrigerated storage for 6 days did not differ (P>0.05) between the two groups. The results show that cinnamon oil supplementation may not have the potential to improve lamb growth performance and meat quality characteristics.  相似文献
