Two groups distinguishable on the basis of crystal serology have been identified within Bacillus thuringiensis var kurstaki (serotype 3 ab). The toxicities of these two groups to Trichoplusia ni and Heliothis virescens expressed as TH ratio also differed. It is suggested that serotype 3 ab be separated into two subgroups designated as K-1 and K-73.  相似文献

采用浸叶法测定了2003年秋季、2004年春季采自广东惠州、福建福州、浙江杭州和江苏南京的小菜蛾Plutella xylostella田间种群对Cry1Aa、Cry1Ab、Cry1Ac和Cry2Aa以及Bt制剂kurstaki亚种 (Bacillus thuringiensis subsp. kurstaki, Btk)的抗性水平。与敏感品系PHI-S相比,广东惠州田间小菜蛾种群的抗性水平最高,其对Cry1Ab和Cry1Ac的抗性分别达到了168和120倍,均为高抗水平; 对Btk制剂的抗性有47倍,达到了中抗水平;对Cry1Aa和Cry2Aa具有低水平抗性 (分别为5.8和5.6倍)。福建福州、浙江杭州和江苏南京田间小菜蛾种群抗性水平相近,对Cry1Ab和Cry1Ac具有低至中等水平抗性 (8~28倍),对Btk制剂具有低水平抗性 (3.5~7倍),对Cry1Aa和Cry2Aa还没有产生明显抗性。因此,在我国东南沿海地区要注意Btk制剂与Bt其他亚种制剂或其他生物杀虫剂轮换使用,以减小Bt制剂对小菜蛾的选择压力,延缓小菜蛾对Bt抗性的发展。  相似文献

Trials were conducted to study how spring Bacillus thuringiensis Berliner subsp. kurstaki treatments on apple may be timed to maximize the survival of parasitoids of the obliquebanded leafroller, Choristoneura rosaceana (Harris) (Lepidoptera: Tortricidae), found in the southern interior of British Columbia, Canada. Orchard collections verified that second through fourth instar obliquebanded leafrollers were found in varying proportions from pink through the petal fall stage of apple development when spring B. thuringiensis treatments are applied vs. lepidopteran pests. Laboratory‐reared second through fourth instar obliquebanded leafrollers, unparasitized and parasitized by one of three native parasitoid species, were fed untreated apple leaves or leaves treated with B. thuringiensis. The highest mortality of unparasitized obliquebanded leafrollers occurred when fourth instars were exposed to B. thuringiensis‐treated leaves; B. thuringiensis‐induced mortality in the unparasitized second and third instars was less than 50%. The consumption of B. thuringiensis‐treated leaves by host larvae significantly increased the percentage of dead host larvae in all parasitized and unparasitized treatments. However, because of the low susceptibility of this leafroller species to B. thuringiensis, relatively high numbers (38–43%) of three obliquebanded leafroller parasitoid species were able to survive the consumption of B. thuringiensis by second and third instar host larvae. Fourth instar obliquebanded leafrollers were found at the full bloom and petal fall stage of apple development in the orchard, at which time B. thuringiensis treatments are recommended for optimal leafroller control. The highest parasitoid mortality due to host mortality was recorded in Apophua simplicipes Cresson (Hymenoptera: Ichneumonidae) and Macrocentrus linearis (Nees) (Hymenoptera: Braconidae), when the hosts were treated as fourth instars. Both of these parasitoids emerge from fifth and sixth instar obliquebanded leafrollers. Bacillus thuringiensis did not have as negative an impact on Apanteles polychrosidis Viereck (Hymenoptera: Braconidae), which emerges when the host is in the fourth instar. When leafroller mortality and parasitism were combined, the B. thuringiensis treatment did not significantly increase host elimination above that of parasitism alone, except for larvae parasitized by A. simplicipes that were in the fourth instar. The consumption of B. thuringiensis by unparasitized larvae was shown to slow larval development.  相似文献

Bacillus thuringiensis strains that belong to B. thuringiensis, B. kurstaki and soil-isolated B.t. were assessed in the following phytopathogenic: Rhizoctonia solani, that had their mycelial growth decreased after incubation in the presence of the bacterial strains. The bacteria have also pathogenic effect against the insect pest Spodoptera littoralis. The isolate B.t. D-1 and the B.t. kurstaki HD-203 were found to be inhibiting R. solani, the strain B. kurstaki HD-203 displayed the highest percentage of inhibition (64%) and B.t. D-1 showed 49% of inhibition. Antagonistic activity was maintained up to pH 8.5, and the antifungal activity was stable to heat at 70?°C for 1?h. Minimal inhibitory concentrations were 152 and 131?μl/ ml for B.t. D-1 and B. kurstaki HD-203, respectively. The two strains also have high efficacy against S. littoralis larvae, B.t. D-1 gave 70% and the B. kurstaki HD-203 strain gave 80% mortality after seven days of treatment.  相似文献

Technical powder of DOR Bt-127, a local isolate of Bacillus thuringiensis (Bt) var. kurstaki, containing 105?µm particles was subjected to milling in a planetary ball mill. Dynamic light scattering of samples drawn at 30?min intervals revealed reduced particle sizes ranging 1171–210?nm. Alkali-soluble protein was higher at 153–175?mg/g in milled Bt powders over 140?mg/g in unmilled Bt powder. All milled powders gave higher mortality of Spodoptera litura (Fabricius) larvae in comparison to the unmilled Bt powder. Bt powder from 60?min milling containing 559?nm particles with a low polydispersity index of 0.338 gave a lower LC50 value of 1.35?mg/ml against third instar S. litura larvae in comparison to 2.04?mg/ml for unmilled Bt powder. LC50 value of the suspension concentrate (SC) formulation developed with 559?nm Bt particles was 2.84?µl/ml containing only 0.95?mg Bt. Field evaluation of the SC formulation against S. litura on castor revealed highest per cent larval reduction of 92.4 and 96.2 at concentrations of 2.5 and 3.0?ml/l, respectively.  相似文献

A local strain DOR Bt-1 belonging to Bacillus thuringiensis var. kurstaki (Bt.k) was multiplied through solid state fermentation and the resulting technical powder was milled and sieved to obtain particles of various sizes. Efficacy of Bt.k. against larvae of Helicoverpa armigera was found to increase with the decrease in particle size. Boric acid was found to be synergistic to DOR Bt-1 technical powder. LC50 of the Bt and boric acid mixture (75:25) was lower at 89.63 mg/100 mL in comparison to Bt alone at 116.75 mg/100 mL. A suspension concentrate formulation with DOR Bt-1 technical was developed using boric acid as an adjuvant. The formulation was found to be highly effective against H. armigera in laboratory bioassays with an LC50 of 185 µL (containing 53.36 mg Bt). The formulation gave effective control of H. armigera on sunflower within 3 days after spray even at the lowest dose of 1.0 mL/L under field conditions.  相似文献

Bacillus thuringiensis var. kurstaki has been widely used in caterpillar control programs. Large-scale production of this bacterium is expensive because of the high cost of the raw materials used in the medium. In this study, we attempted to develop an economical medium, based on inexpensive, locally available raw materials using a 3-L fermenter. Parthenium hysterophorus L. extract based culture medium resulted in highest toxicity (LC50 14.628 µg mL–1) against 7-day-old Spodoptera litura (Fab) larvae, spore count (4.1 × 109 spores mL–1) and biomass (4.9 g L–1) within a short fermentation time of 36 h. It was 512 times cheaper than the nutrient broth (standard medium) used for B. thuringiensis production. Hence, this parthenium extract based culture medium was considered most economical with potential for the large-scale industrial production of B. thuringiensis.  相似文献

When microorganisms are used for pest control in vegetable production, the active organisms become part of the microbiota growers are exposed to. The aim of this study was to quantify vegetable growers' exposure to the bacterial strain Bacillus thuringiensis kurstaki strain HD1 (termed HD1) from the biocontrol agent Dipel®, and other airborne mesophilic bacteria. Personal (n=102) and stationary (n=43) measurements of exposure were performed in greenhouses and open fields. Air samples were analysed by plate counts, and total counts with a microscope. Isolates resembling HD1 were identified by PCR analysis. HD1-like bacteria were only detected in environments where Dipel® was used. In a greenhouse with Dipel® treated tomato plants, the growers' exposure to airborne HD1-like bacteria reached 5300 cfu/m3 and 1400 cfu/m3 during harvest and clearing of old plants, respectively. In untreated greenhouses, the highest concentration of total mesophilic bacteria, 1,100,000 cfu/m3, was detected in a cucumber greenhouse. The median concentrations of mesophilic bacteria in tomato greenhouses were significantly lower than the median concentrations in cucumber greenhouses. There was no significant difference in exposure to mesophilic bacteria in tomato greenhouses and in vegetable fields. We found that greenhouse workers, especially in cucumber production, were exposed to high concentrations of total bacteria. Thus, the already present airborne bacteria in greenhouses might have a greater influence on growers' health than applied biocontrol strains. However, further studies are needed to establish an occupational threshold limit for airborne bacteria and to secure a healthy working environment for vegetable growers.  相似文献

To evaluate phylogenetic relatedness as a proxy for susceptibility to Bacillus thuringiensis (Bt) when selecting species to act as surrogates for others in prerelease testing, we examined the responses of 11 laboratory‐reared lepidopteran colonies, comprising members of 2 families, 5 genera, and 9 species, to a commercial Bt preparation. Survival, pupal mass, and timing of pupation and adult emergence of 2 noctuids (Spodoptera litura and Helicoverpa armigera) and 7 tortricids (Cnephasia jactatana, Ctenopseustis obliquana, Ctenopseustis herana,Planotortrix octo, Planotortrix notophaea,Planotortrix excessana [2 different laboratory colonies], and Epiphyas postvittana [2 colonies]) were examined after feeding first instar larvae with artificial diet containing 5 μL/100 mL Dipel® ES (Bt subsp. kurstaki). Bt caused significant larval mortality in all species except S. litura, in which only pupation was delayed compared with untreated controls. Neither of the noctuid species tested would act as a suitable surrogate for the other in tests of Bt impacts on survival. With the exception of the 2 colonies of E. postvittana, which differed from each other not only in their responses to Bt but also in their development times when not treated with Bt, species within each tortricid genus had similar responses to Bt and thus could act as surrogates for each other. Members of different genera within this family could represent each other only if relatively coarse measurement endpoints (e.g., toxic or not) were considered adequate for assessing risks to nontarget species in the field.  相似文献
