Rhizoctonia solani anastomosis group (AG) 11 causes serious damping‐off and hypocotyl rot of narrow‐leafed lupins (Lupinus angustifolius) in the northern grain‐belt of Western Australia. R. solani AG‐11 produced abundant sclerotia in sand overlaid on potato dextrose agar. Sclerotia were produced in larger numbers in natural Lancelin sand than in Geraldton loamy sand collected from the northern grain‐belt of Western Australia. The majority of the sclerotia produced were in >250 to <500 μam size range. The germination levels of sclerotia in the first two cycles of drying and germination were not significantly different. Sclerotia still retained 50% germination after four such cycles, indicating that they may have the ability to withstand the climatic cycles of the Mediterranean environment of southwestern Western Australia. The radial growth of the mycelium from sclerotia, however, declined with each drying and germination cycle. Inoculum potential of the pathogen increased with the size of sclerotia resulting in more severe lupin hypocotyl rot with larger sclerotia. The number of sclerotia produced in soil increased with increasing density of lupin seedlings. The results also indicate that R. solani AG‐11 can produce sclerotia on infected plant tissues as well as in soil. This is the first report of the prolific production of sclerotia by AG‐11 and their significant role in infection of lupins in soil in Western Australia.  相似文献

Air dry soil samples stored at room temperature for more than one and a half years, were used as inocula for actinorhizal plants. Seedlings of Colletia paradoxa, Discaria americana, D. articulata and D. trinervis (Rhamnaceae) cultivated in nitrogen-free nutrient solution, were inoculated by adding dry soil to the solution. All the soil samples tested were able to induce nodulation, showing the presence of infective propagules of Frankia. Healthy growth of nodulated plants suggested the occurrence of nitrogen fixation.  相似文献

The relationship between population densities of race 1 of Meloidogyne incognita and yield of eggplant was studied. Microplots were infested with finely chopped nematode-infected pepper roots to give population densities of 0, 0.062, 0.125, 0.25, 0.50, 1, 2, 4, 8, 16, 32, 64, and 128 eggs and juveniles/cm³ soil. Both plant growth and yield were suppressed by the nematode. A tolerance limit of 0.054 eggs and juveniles/cm³ soil and a minimum relative yield of 0.05 at four or more eggs and juveniles/cm³ soil were derived by fitting the data with the equation y = m + (1 - m)zP⁻T. Maximum nematode reproduction rate was 12,300. Hatch of eggs from egg masses in water or from sodium hypochlorite dissolved egg masses was similar (41% and 39%), but egg viability was significantly greater from egg masses in water (58%) than from sodium hypochlorite dissolved egg masses (12%) after 4 weeks. Greater numbers of nematodes were collected from roots of tomatoes from soil infested with entire egg masses than from tomato roots from soil infested with egg masses dissolved by sodium hypochlorite.  相似文献

Effects of inoculum size and total sugar content on both l-phenylalanine productivity and titre have been investigated using a tyrosine auxotrophic regulatory mutant of Escherichia coli. Fermentations were carried out in a 500 litre pilot fermenter with intermittent feeding of d-glucose plus phosphate. It was found that the productivity was not greatly affected by inoculum size. However, the l-phenylalanine titre was significantly affected by total sugar content. Relatively high productivities of up to 0.35–0.40 g l-phenylalanine l?1 h?1 have been achieved at l-phenylalanine titres of 14–15 g l?1.  相似文献

In intact, uncoupled type B chloroplasts from spinach, added ATP causes a slow light-induced decline (t12 ≈ 3 min) of chlorophyll a fluorescence at room temperature. Fluorescence spectra were recorded after fast cooling to 77 K and normalized with fluorescein as an internal standard. Related to the fluorescence quenching at room temperature, an increase in Photosystem (PS) I fluorescence (F735) and a decrease in PS II fluorescence (F695) were observed in the low-temperature spectra. The change in the F735F695 ratio was abolished by the presence of methyl viologen. Fluorescence induction at 77 K of chloroplasts frozen in the quenched state showed lowered variable (Fv) and initial (F0) fluorescence at 690 nm and an increase in F0 at 735 nm. The results are interpreted as indicating an ATP-dependent change of the initial distribution of excitation energy in favor of PS I, which is controlled by the redox state of the electron-transport chain and, according to current theories, is caused by phosphorylation of the light-harvesting complex.  相似文献

Amphiraphia Chen & Zhu, together with theAmphiraphiaceae andAmphiraphidales, should be abandoned, sinceAmphiraphia cells are the heterovalvar initial cells ofCaloneis. WhenCaloneis reproduces sexually two cells pair and become surrounded by a two-layered capsule of mucilage. Each cell produces two gametes and these become rearranged within the gametangial frustule before plasmogamy. The gametes are amoeboid and fusion is isogamous. The Longwood Complex wildfire in the Siskiyou Mountains of southern Oregon in August 1987 created an opportunity to study erosion and its effects on mycorrhizal fungus inoculum potential of a forest soil on steep slopes. As measured by the erosion-bridge method, most erosion occurred in a single, intense storm in December after the fire and amounted to an estimated 2 to 4 cm of surface soil. Captured eroded soil had a higher pH and P and Mg levels than residual soil. Seedlings ofLibocedrus decurrens andPseudotsuga menziesii were planted on eroded plots with additions of captured eroded soil (ET) or pasteurized eroded soil (PET) transferred to the planting holes. After one growing season,Libocedrus seedlings formed nearly 4 times the vesicular-arbuscular mycorrhizae in ET treatments and more than twice as much in PET treatments than in controls. Survival and basal area growth were significantly better in ET than in the other treatments, and both ET and PET produced more seedling shoot growth than did controls.Pseudotsuga seedlings did not differ in measured characteristics between treatments; ectomycorrhiza formation was slight, evidently the result of reduced inoculum potential resulting from the fire.  相似文献

Mnemiopsin 1 (Mn1) and Mnemiopsin 2 (Mn2) are photoproteins found in Mnemiopsis leidyi. We have tried to answer the question of whether the structural features of photoproteins can explain the observed activity data. According to the activity measurements data, they have the same characteristic wavelength. However, the initial intensity of Mn2 is significantly higher than that of Mn1, and decay time of Mn1 (0.92 s−1) is lower than that of Mn2 (1.46 s−1). The phylogenetic analysis demonstrates that, compared with Obelin and Aequorin from Obelia longissima and Aequorea victoria, respectively, a gene modification event may have caused the expansion of the N-terminal side of all photoproteins from M. leidyi. An in silico study has shown that the stability of the photoprotein–substrate complex of Mn2 is higher than that of Mn1, indicating a higher affinity of the substrate for Mn2 compared with Mn1. It was revealed that the active EF-hand loops 1 and III in Mn2 is locally more rigid compared with those in Mn1. We concluded that different stability of the photoprotein complexes leads to different initial intensity. While different patterns of the local dynamics of loops I and III may influence the decay rate.  相似文献
