Summary Changes in the chord conductanceG and the membrane electromotive forceE m in the so-called breakdown region of large negative potential of theChara plasmalemma were analyzed in more detail. In addition to the increase inG, the voltage sensitivity of the change inG increased, which was the cause of marked inductive current in the breakdown region. The breakdown potential, defined as a critical potential at which both low and high slope conductances of theI–V m relationship cross, almost coincided with the potential at which an inductive current began to appear. This breakdown potential level changed with pH o in a range between 5 and 9. TheChara plasmalemma was electrically most tolerant around pH o 7.In some cellsE m shifted to a positive level as large as +50+70 mV during the breakdown phenomenon. Such a large positive shift ofE m is caused mainly by the increase in conductance of Cl and partly Ca2+ and K+.  相似文献

The rate of exchange of peptide group NH hydrogens with the hydrogens of aqueous solvent is sensitive to neighboring side chains. To evaluate the effects of protein side chains, all 20 naturally occurring amino acids were studied using dipeptide models. Both inductive and steric blocking effects are apparent. The additivity of nearest-neighbor blocking and inductive effects was tested in oligo-and polypeptides and, suprisingly, confirmed. Reference rates for alanine-containing peptides were determined and effects of temperature considered. These results provide the information necessary to evaluate measured protein NH to ND exchange rates by comparing them with rates to be expected for the same amino acid sequence is unstructured aligo- and polypeptides. The effects of various medium sterilization conditions on fermentations of a recombinant, acidic fibroblast growth factor (aFGF) producing Escherichia coli have been studied. Changes in the medium resulting from sterilization were monitored by pH and absorption spectra. This simple experiment provided excellent data for the demonstration of the usefulness of comparative reasoning tools in order to evaluate the effect of sterilization on fermentation performance. The time profiles of the main parameters (e.g., carbon dioxide evolution rate, dissolved oxygen, pH, and aFGF productivity) were simplified into piecewise contiguous linear segments, each of which was sequentially numbered. The length, position, and slope of each tine were then characterized. Application of the comparative reasoning tools confirmed that separate sterilization of the glucose was necessary for the success of the process, despite adding to the cost and complexity. The comparative data analysis also showed that scaleup with longer sterilization holding and cooling times would not be detrimental to aFGF production. (c) 1995 John Wiley & Sons, Inc.  相似文献

There is a continuing controversy over Native American fishing and hunting rights. We show that Native American (Menominee) and European American fish experts have a common knowledge base and share values and attitudes associated with fishing practices (though organized around different ethical principles). Nonetheless, perceived group differences are dramatic (especially European American perceptions of Native Americans). Cultural differences in models of nature and associated inference processes appear to mediate these stereotypes and may hold the key to reducing intergroup conflict over resources.

As an illustration of the complexities involved in connecting physics and molecular biology at the nanoscale, in this paper I discuss two case studies from nanoscience. The first examines the use of a biological structure (DNA) to build nanostructures in a controlled way. The second discusses the attempt to build a single molecular wire, and then decide whether such a wire is indeed conducting. After presenting the central features of each case study, I examine the role played in them by microscopic imaging, the different styles of reasoning involved, and the various theoretical, methodological, and axiological differences. I conclude by arguing that, except for the probe microscopes that are used, there is very little in common between the two cases. At the nanoscale, physics and molecular biology seem to meet in a non-unified way.  相似文献

Exclusion allows the detection of hidden food when confronted with the choice between an empty and a potentially baited food location. However, exclusion may be based on avoidance of the empty location without drawing inferences about the presence of the food in the baited location. So far, such inferences have been demonstrated in the great apes only: after seeing an experimenter eating one of two food types, which both had been hidden previously in two boxes, the apes were able to choose the box that still contained the other food type. African grey parrots are capable of exclusion, and we here assessed if they are capable of inference by exclusion. In our task, two different but equally preferred food items were hidden in full view of the birds under two opaque cups. Then, an experimenter secretly removed one food type and showed it to the bird. Similarly to the apes, one out of seven parrots significantly preferred the baited cup; control conditions rule out that its choice was based on associative learning or the use of olfactory cues. Thus, we conclude that-like the apes-some grey parrots are able to infer the location of a hidden food reward.  相似文献

Aquatic vegetation plays a very important role in providing food, shelter, and nursery habitat and is also regarded as hydraulic resistance in the stream environment. To achieve better ecological restoration, this trade-off should be solved both hydraulically and ecologically. This study quantifies the effect of aquatic vegetation on the spatial distribution of Japanese medaka (Oryzias latipes) to evaluate its importance to fish habitat preference. The preference for aquatic vegetation index is calculated using a fuzzy preference intensity model (FPIM) with interactions among water depth, current velocity and cover ratio in an agricultural canal. In this model, simplified fuzzy reasoning is introduced to explicitly take the essential vagueness of fish behavior into consideration, and a simple genetic algorithm is used to search for an optimum model representation. Uncertainties in measurement errors and dispersions of the physical environment are positively taken into the model using symmetric triangular fuzzy numbers. To overcome the difficulty in model construction with insufficient data observed in an agricultural canal, this model was conjugated with a model developed in a laboratory experiment. The model obtained was then assessed using the AIC (Akaikes Information Criterion) to evaluate the significance of vegetation index with a statistical approach. The results suggest the significance of vegetation index to habitat selection by Japanese medaka and that utilization of the AIC enables us to grasp the validity of an additional factor contributing to habitat prediction with a view to a definite scale  相似文献

We studied human nonverbal transitive inference in two paradigms: with choice stimuli orderable along a physical dimension and with non-orderable choice stimuli. We taught 96 participants to discriminate four overlapping pairs of choice stimuli: A+ B−, B+ C−, C+ D−, and D+ E−. Half of the participants were provided with post-choice visual feedback stimuli which were orderable by size; the other half were not provided with orderable feedback stimuli. In later testing, we presented novel choice pairs: BD, AC, AD, AE, BE, and CE. We found that transitive responding depended on task awareness for all participants. Additionally, participants given ordered feedback showed clearer task awareness and stronger transitive responding than did participants not given ordered feedback. Associative models ( [Wynne, 1995] and [Siemann and Delius, 1998]) failed to predict the increase in transitive responding with increasing awareness. These results suggest that ordered and non-ordered transitive inference tasks support different patterns of performance.  相似文献
