Functional zooplankton bioassays based on ingestion, reproduction and respiration are described, with methods for a new ingestion bioassay included. All bioassays are compared using three indices, including the variability of controls, the range of experimental responses, and a listing of contaminants causing inhibition/stimulation of response. The ingestion bioassay showed the greatest range of response, and was sensitive to pesticides, PCBs and heavy metals. It was also commonly characterized by a hormesis response. The reproduction bioassay showed the lowest variability, illustrated a reduced range of response, and was sensitive to nutrients and heavy metals. In one study, the respiration bioassay was sensitive only to PCBs.  相似文献

A range of surfactants, including the anionic sodium dodecyl sulfate, the cationic cetyltrimethylammonium bromide and the nonionics octylphenoxy polyethoxyethanol (Triton X100) and polyoxyethylene 20 sorbitan monoleate (Tween 80) was studied for effects on proliferation, contractibility and attachment of cultured human fibroblasts. Only ionic surfactants exhibited a stimulatory effect on fibroblast proliferation, whereas all the surfactants tested increased the contraction of collagen gels containing fibroblasts, with the greatest effect from the non-ionic surfactants. This activity was not correlated with an increase of cell population or cell attachment within the collagenous matrix. The activity of the surfactants was seen only at levels close to their LD50 values and in a narrow range of concentrations. Experiments were designed to demonstrate that growth is precisely regulated in crustacean larvae and that cyclic temperatures act as a perturbation on growth regulation. Newly hatched larvae were exposed to either the specific a cyclic temperature, cumulative growth was significantly inhibited. Also, the specific growth rate ( ) of larvae in the cyclic temperature regime was found to oscillate with decreasing amplitude around the of the larvae exposed to constant temperature. This oscillatory pattern has been observed in a wide variety of self-regulating systems. Evidence is presented to demonstrate that this regulatory mechanism also adapts to long-term exposure to cyclic temperatures. These findings increase our understanding of how larvae cope with and adapt to changing environmental temperatures. More significantly, the mechanism could provide a sensitive tool for estimating the effects of specific environmental disturbances on fitness.  相似文献

Recent evidence from studies on DNA repair systems that are implicated in accelerated aging syndromes, have revealed a mechanism through which low levels of persistent damage might exert beneficial effects for both cancer prevention and longevity assurance. Beneficial effects of adaptive responses to low doses of insults that in higher concentrations show adverse effects are generally referred to as hormesis. There are numerous examples of hormetic effects ranging from mild stresses of irradiation to heat stress, hypergravity, pro‐oxidants, or food restriction. Although the notion of hormesis is supported by many observations in various organisms, at least two major caveats have thus far prevented the application of hormesis for disease prevention in humans. First, the very nature of hormesis using toxins as a treatment regimen harbors the inherent danger of detrimental consequences. Second, the molecular mechanisms through which insults might exert beneficial effects have thus far remained elusive. Here, I discuss a mechanistic basis for hormesis and its implications for cancer prevention and healthy aging.  相似文献

Ageing is associated with a decrease in the ability of cells to cope with environmental challenges. This is due partly to the attenuation of a primordial stress response, the so-called heat shock (HS) response, which induces the expression of heat shock proteins (HSPs), composed of chaperones and proteases. The attenuation of the HS response during ageing may be responsible for the accumulation of damaged proteins as well as abnormal regulation of cell death. Maintenance of the HS response by repeated mild heat stress causes anti-ageing hormetic effects on cells and organisms. Here, we describe the molecular mechanism and the state of the HS response as well as the role of specific HSPs during ageing, and discuss the possibility of hormetic modulation of ageing and longevity by repeated mild stress.  相似文献

Despite the long history of radiation hormesis and the public health concerns with low-level exposures to ionizing radiation, there has been surprisingly little formal evaluation of whether hormetic effects are displayed with respect to radiation exposure and cancer incidence (i.e., reduced cancer risk at low radiation doses compared to controls, enhanced cancer risk at higher doses) until relatively recently. This paper reviews data relevant to the question of radiation hormesis and cancer with particular emphasis on experimental studies in animal models exposed to low levels of ionizing radiation. Data exist that provide evidence both consistent with and/or supportive of radiation hormesis. Other biomedical research provides potentially important mechanistic insight: low dose exposures have the capacity to activate immune function to prevent the occurrence of tumor development and metastasis; low doses of radiation have been shown to reduce mutagenic responses and induce endogenous antioxidant responses. These findings are consistent with epidemiological data suggesting an inverse relationship between background radiation and cancer incidence and with occupational epidemiological investigations in which low-dose exposure groups display markedly lower standardized mortality rates than the referent or control group.  相似文献

Hormesis is defined as a dose-response relationship that is stimulatory at low doses, but is inhibitory at higher doses. In a given experiment, it is not unusual to observe enhanced responses at low doses, however, such enhanced responses may not imply hormesis, but the random fluctuation of the data. Statistical tests can be developed to detect hormesis when enhanced responses at low concentrations are observed. We propose the use of a model-based approach to detect the presence of, and estimate the extent of, hormesis. This approach includes two steps: detection and estimation. In the detection step, we compare the full and the reduced models. The full model describes the dose-response relationship incorporating the hormetic effect; the reduced model describes the dose-response relationship without the hormetic effect. The full model is an extension of the reduced model and has an extra parameter that measures the amount of increase in response at low doses. A test of statistical significance of this extra parameter can essentially be a test for detecting hormesis. In the estimation step, we obtain the area under the best-fitted dose-response curve falling within the hormetic zone. Considering both the number of concentrations within the hormetic zone and the magnitude of the stimulatory response, we propose using the ratio of the area under the hormetic zone (AUCH) and the area under the best-fitted curve from zero to zero equivalent point (AUCZEP) as an estimate of magnitude of the hormetic effect. Two numerical examples are used to illustrate the use of this model-based approach.  相似文献

Lung‐cancer mortality (LCM) is elevated in underground miners who chronically inhaled the mutagenic, cytotoxic α‐decay products of radon gas. Epidemiologie studies of LCM rates vs. residential‐radon concentration levels are generally considered inconclusive. However, Cohen (Health Physics 68, 157–174, 1995) has hypothesized that data on LCM vs. residential radon concentrations at the U.S. county level are clearly inconsistent with a linear no‐threshold (LN) dose‐response model, and rather are consistent with threshold or hormesis model. Cohen's hypothesis has been criticized as “ecological fallacy,”; particularly because LN (but not threshold or hormesis) models are generally considered biologically plausible for agents like α radiation that damage DNA in linear proportion to dose. To assess the biological plausibility of Cohen's hypothesis, a preliminary study was made of whether a biologically realistic, cytodynamic 2‐stage (CD2) cancer model can provide a good, joint fit to Cohen's set of U.S. county data as well as to underground‐miner data. The CD2 model used adapts a widely applied, mechanistic, 2‐stage stochastic model of carcinogenesis to realistically account for interrelated cell killing and mutation (both assumed to have a LN dose‐response), cell turnover, and incomplete exposure of stem cells. A CD2 fit was obtained to combined summary data on LCM vs. radon‐exposure in white males in 1, 601 U.S. counties (from Cohen) and in white male Colorado Plateau (CP) uranium miners (from the National Research Council's “BEIRIV”; report). The CD2 fit is shown to: (i) be consistent with the combined data; (ii) have parameter values all consistent with biological data; and (iii) predict inverse dose‐rate‐effects data for CP and other radon‐exposed miners, despite the fact that optimization had not involved any of these dose‐rate data. The latter data were not predicted by a simplified CD2 model in which all stem cells were presumed to be exposed. It is concluded that this study provides preliminary evidence that Cohen's hypothesis is biologically plausible.  相似文献

The Asian multicoloured ladybird beetle, Harmonia axyridis, is utilized as a major natural enemy of aphids in the field, greenhouses and orchards. However, it has been looked as invasive predator distributing in worldwide. To refine integrated pest management (IPM) against aphids, it is important to evaluate the effects of insecticides on physiology and behaviour of the high adapted predators. Beta‐cypermethrin, a broad‐spectrum insecticide, not only kills aphids at lethal concentrations but also affects natural enemy of aphids. In our study, the age‐stage, two‐sex life table was used to evaluating sublethal effects of beta‐cypermethrin on the predatory ladybird beetle H. axyridis. In the parent generation, the pre‐oviposition period of H. axyridis was significantly shortened (8.93 days) after exposure to LC5 beta‐cypermethrin (5% lethal concentration) as compared with control (10.06 days). However, the oviposition period was significantly longer (46.17 days instead of 43.90 days), and fecundity (eggs per female) was significantly increased by 49.64% when compared with control. In the F1 generation, the length of the juvenile stage was not affected, but the oviposition period increased significantly (38.19 days compared to 31.39 days in the control). This positive effect was translated to the fecundity that increased significantly by 62.27% as compared with control. According to the life‐table analysis, the intrinsic rate of increase (rm) was significantly higher in treatment (0.140 per day) than that in the control (0.123 per day). In addition, the net reproductive rate (R0) increased significantly by 91.53%. These results would be useful in assessing the overall effects of beta‐cypermethrin on H. axyridis and even for discussing the ecological mechanism of the unexpected extension of H. axyridis during IPM programme.  相似文献
