Summary— In the sperm nuclei the DNA is packaged into a highly condensed form and is not organized into nucleosome and solenoid but is bound and stabilized mainly by the protamines that arrange the DNA in an almost crystalline state. As demonstrated for somatic cells, the sperm DNA has been reported to be organized in loop domains attached to the nuclear matrix structures. However, the possible role of the sperm head matrix in maintaining the loop organization in absence of a typical nucleosomal structures has not been fully elucidated. By using in situ nick translation at confocal and electron microscope level, we analyzed the organization of the DNAprotamine complex and its association with the sperm nuclear matrix. The data obtained indicate that the chromatin organization in sperm nuclei is maintained during the sperm condensation by means of interactions with the nuclear matrix at fixed sites. The fine stucture of sperm nucleus and of sperm nuclear matrix, investigated on sections and replicas of freeze-fractured specimens, suggests that the lamellar array, observed by freeze-fracturing in the sperm nuclei, could depend on the inner matrix which presents a regular organization of globular structures possibly involved in the maintenance of chromatin domains in highly condensed sperm nuclei also.  相似文献

Bacillus stearothermophilus cells vary the lipid fatty acid composition of cytoplasmic membranes with growth temperature. Spin label studies of such membranes have been interpreted to indicate lateral lipid phase separations at the growth temperature. We have now used freeze-fracture electron microscopy to confirm the spin label studies. Freeze-fracture faces of protoplasts indicate slight but distinct protein aggregation at the growth temperature. Aggregation increases rapidly with decreasing quench temperature in wild-type cells. In contrast we were unable to demonstrate extended protein segregation in membranes of a temperature-sensitive mutant that contains more than 58% branched fatty acids. Storage of protoplasts for prolonged times below the lipid phase transition results in the appearance of corrugated fracture faces with 300- to 500-Å repeat patterns, although this organism does not synthesize lecithins.  相似文献

The sodium channel was studied in osmotically-sensitive membrane preparations from rat brain and in innervated and chronically denervated rat soleus and extensor digitorum longus muscles. These experiments were undertaken in order to define a set of parameters for sodium channel function at the subcellular level to be used as a measure of retention of channel integrity upon subsequent isolation of the channel. Various neurotoxins and drugs were employed to control the permeability of the brain membranes to 22Na and the sodium-conductance properties of the muscles. Batrachotoxin (ED50 = 0.2 μM), veratridine (ED50 = 1 μM), or grayanotoxin I (ED50 = 30 μM) stimulated 22Na uptake in brain membranes is inhibited in an apparently uncompetitive manner by the sodium channel blocking agents tetrodotoxin and saxitoxin in a simple competitive manner by Ca2+ and in a partial or allosteric competitive manner by lidocaine and procaine. This 22Na uptake assay, which can be equated to a measure of equilibrium toxin binding, shows dependence on the concentration of the membranes and is sensitive to pH, temperature, ionic strength, and the ionic composition of the media. Parallel biophysical studies on sodium channels in rat muscle show that the properties of the sodium channel are similarly affected by these agents.  相似文献

Excellent preservation and new structural details can be demonstrated in rust-infected leaf tissue after high pressure freezing and freeze-fracturing. A tubular-vesicular complex was the most remarkable cytoplasmic structure observed in cells of the bean rust fungus Uromyces appendiculatus var. appendiculatus during its establishement in its host Phaseolus vulgaris. In fungal cells undergoing intensive synthesis of wall material, this membranous system extended throughout the cytoplasm; in addition, vesicles were accumulated adjacent to the plasma membrane. Here, membrane-associated configurations were observed which seem to be involved in exo- and/or endocytotic processes. It is assumed that the tubular-vesicular complex belongs to the endomembraneous system of the bean rust fungus and that it is involved in the synthesis and secretion of wall material.  相似文献

Freeze-fracturing of outer wall layers ofCladosporium conidia revealed two types of ultrastructure, coinciding with taxonomic characteristics. The outer conidial layers were essentially smooth in the human pathogenic species,C. bantianum, C. carrionii, andC. trichoides. In contrast, mosaic arrays of rodlets on conidia were observed with freeze-fracturing in the saprobic species,C. cladosporioides, C. coralloides, C. herbarum, C. sphaerospermum, andC. variabile. Conidia ofC. elatum were an exception among the saprobic species as they had smooth surfaces. The present study supports the suggestion that the human pathogenicCladosporium species should be transferred to another genus.  相似文献

Freeze-fractured thylakoids of Fucus serratus L. exhibit three types of faces with a particle density analogous to that of EFs, EFu and PF faces of green plants. However the particle size distribution is unimodal in the three types with a mean of about 8 nm. No obvious distinction between PFs and PFu faces could be detected. The absence on EFs faces of the distinct class of large particles (>13 nm) existing in green plant thylakoids implies a unique organization of pigment proteins, especially of the light-harvesting complexes.  相似文献
