One-week treatment with the benzodiazepine (BZ) flurazepam (FZP), results in anticonvulsant tolerance, associated with reduced GABAA receptor (GABAR) subunit protein and miniature inhibitory post-synaptic current (mIPSC) amplitude in CA1 neurons of rat hippocampus. Because protein kinase A (PKA) has been shown to modulate GABAR function in CA1 pyramidal cells, the present study assessed whether GABAR dysfunction is associated with changes in PKA activity. Two days after 1-week FZP treatment, there were significant decreases in basal (- 30%) and total (- 25%) PKA activity, and a 40% reduction in PKA RIIbeta protein in the insoluble fraction of CA1 hippocampus. The soluble component of CA1 showed a significant increase in basal (100%) but not total PKA activity. Whole-cell recording in vitro showed a 50% reduction in mIPSC amplitude in CA1 pyramidal cells, with altered sensitivity to PKA modulators. Neurons from FZP-treated rats responded to 8-bromo-cAMP with a significant increase (31%) in mIPSC amplitude. Likewise, vasoactive intestinal polypeptide (VIP), an endogenous PKA activator, caused a significant 36% increase in mIPSC amplitude in FZP-treated cells. Neither agent had a significant effect on mIPSC amplitude in control cells. This study supports a role for PKA in GABAR dysfunction after chronic FZP treatment.  相似文献

实验在49只局部麻醉、肌肉麻痹、切断双侧颈迷走神经的家兔上进行。观察到一侧孤束核(NTS)区微量注射氟安定,使对侧膈神经放电平均幅度减低,呼吸频率加快。事先在NTS区微量注射GABA 受体拮抗剂印防己毒素,可阻断氟安定的减低膈神经放电平均幅度的作用。在脑桥头端横断脑干出现长吸式呼吸的免上,一侧NTS区微量注射氟安定,也使对侧膈神经放电平均幅度减低,但呼吸频率减慢。在脑桥结合臂旁内侧核(NPBM)区微量注射氟安定,使呼吸频率明显减慢,吸气和呼气时程延长,这效应可被毒扁豆碱阻断。脊髓蛛网膜下腔注射氟安定,使膈神经放电平均幅度减低,血压下降,这效应可被印防己毒素阻断。结果提示,NPBM 的存在与呼吸频率加快有关;而氟安定作用于NPBM区,却减慢呼吸频率,其机制可能是抗胆碱能作用。氟安定直接作用于NTS 区或脊髓使膈神经放电平均幅度减低、血压下降,其机制可能与激活GABA 受体有关。  相似文献

Using whole cell patch-clamp recordings from pyramidal cells acutely dissociated from rat hippocampal slices, Ro-15 1788 (flumazenil, FLU) was shown to enhance the GABAA-receptor mediated currents evoked by application of -aminobutyric acid (GABA) and to antagonize the enhancing effect of the benzodiazepine agonist flurazepam (FZP) on the GABAA response. Both FLU and FZP increased the peak and the steady-state components of the responses and accelerated the current decay. This suggests that both agents act via a common mechanism on GABA transmission. It is concluded that FLU possesses high affinity for the binding site, but low efficacy on the GABAA-benzodiazepine receptor. This suggests that FLU acts as a partial agonist on GABAA receptors.  相似文献

32只家兔,用α-氯醛糖和氨基甲酸乙酯麻醉,肌肉麻醉行人工呼吸。观察安定或氟安定对刺激中脑导水管周围灰质背侧部所引起升压反应的影响。当静脉注射安定或氟安定注入侧脑室,使刺激中脑诱发的升压反应受抑,表面为收缩压增量和平均压增量减小。微量注射氟安定到双侧延髓腹外侧头端区,也出现升压反应受到抑制,降低△SBP和△MBP;而于双侧腹外端区微量注射相同剂量氟安定则无抑制作用。在rVLM用GABAA受体阻断剂  相似文献

Twenty four hours after mice were forced to swim for up to 10 minutes in cold water, there was a reduction in the ability of MK-801 to antagonize the electrical precipitation of tonic hindlimb extension. Milacemide, a lipophilic prodrug of glycine, restored the antiseizure efficacy of MK-801 to the same level observed in unstressed animals treated with milacemide and MK-801. Stimulation of the glycine-gated chloride ionophore subsequent to the liberation of free glycine could explain milacemide's pharmacologic action as an adjuvant to MK-801. Consistent with this interpretation, milacemide was able to potentiate the antiseizure effects of flurazepam, a benzodiazepine agonist, in stressed and unstressed mice and carbamazepine in unstressed animals.d-cycloserine, a partial glycine agonist with greater specificity for the strychnine-insensitive modulatory site on the NMDA receptor complex, was examined for its effect on MK-801's antiseizure efficacy. At a high dose (320 mg/kg),d-cycloserine alone had an anticonvulsant effect. Moreover, this dose ofd-cycloserine administered with MK-801 showed a significantly greater anticonvulsant efficacy than MK-801 alone. The data support the development of glycinergic interventions as adjunctive agents in the pharmacotherapy of seizure disorders.  相似文献

Copper complexation by the 1,4-benzodiazepines medazepam, diazepam, flurazepam, nitrazepam, and clonazepam was investigated using differential pulse polarography and cyclic voltammetry at a mercury electrode in 0.10 M KNO3 and pH 7.0 +/- 0.1. Because the 1,4-benzodiazepines are easily reduced at a mercury electrode through the two-electron reduction of the 4,5-azomethine functional group, copper reduction, as well as that of the ligands, was analyzed under varying experimental conditions. In most situations adsorption phenomena occurred and their influence on voltammetric signals had to be carefully analyzed. The voltammetric behavior was then interpreted in terms of complex formation. The results showed that all benzodiazepines can act as ligands toward copper(II) ions, forming 1:1 and 1:2 complexes with similar stabilities. The stoichiometric acidity constants of the benzodiazepines under study were also determined by potentiometric titration in water-ethanol medium and 0.10 M KNO3 and then extrapolated to 0% concentration of ethanol.  相似文献
