The results of experiments on the efficiency of periphyton communities for nutrient removal from polluted streams in a continuous flow-through are given. The artifical stream (5 m × 0.7 m × 0.5 m) was made of wood, with silon (a kind of nylon) screens, as a substratum for periphyton growth. The elimination of nutrients was monitored by ammonium, nitrite, nitrate and orthophosphate analyses. In addition, the elimination of organics and the decrease in trophic state were determined. During two field experiments a marked elimination of nitrogen and phosphorus was demonstrated. The maximum efficiency of ammonium and orthophosphate removal was 80% and 70%, respectively. Organic removal reached 35% (C.O.D.Mn-Kubel) and 54% (B.O.D.5). Inorganic and organic nutrient elimination caused significant changes in periphyton community structure in the outflow portion of the through, evaluated by the saprobic index and the similarity coefficient. The experiments confirmed that periphyton communities are a useful means of nutrient removal from polluted streams.  相似文献

Experiments by several authors on the effects of polarized light on phytochromemediated responses in fern gametophytes and in the green alga Mougeotia have earlier been interpreted as showing that the transition moment of phytochrome in the Pr form is parallel to the plasmalemma, but perpendicular to the plasmalemma for the Pfr form of phytochrome. It is now shown that the experimental results can be interpreted differently, and that they are also consistent with a chromophore rotation of about 30° (instead of 90°), as found for immobilized phytochrome molecules in vitro. Thus there is no evidence for a rotation of the whole phytochrome protein. For the gametophyte of Adiantum it is calculated that the Pr transition moment is inclined 17° to the plasmalemma, and the Pfr transition moment ca 50°, corresponding to an in vivo chromophore rotation of ca 33°; however, these values are very approximate.  相似文献

Nonoccluded baculovirus-and filamentous virus-like particles were found in nuclei of hemocytes or midgut cells of field-collected spotted cucumber beetles. Each type of particle was associated with a different type of virogenic stroma containing various viral components similar to those referred to as capsid, nucleocapsid, viroplasm, and viral envelope in other known baculovirus infections. Nucleocapsids of the virus which occured only in hemocytes were rod-shaped particles approximately 230 nm long and 52 nm wide and were enveloped singly by a trilaminar unit membrane. Enveloped and partly enveloped particles appeared to be released from the nucleus to the cytoplasm by budding through the nuclear envelope acquiring additional membranes. The nucleocapsids of the virus which occurred only in nuclei of midgut cells were filamentous particles with an average diameter of 25 nm and variable length up to 2 μm. Some extremely long particles were bent almost 360° near the middle, resulting in a hairpin-like configuration. The particles were always enveloped singly. No particles budding through the nuclear envelope were observed.  相似文献

Summary Two dynamic changes are associated with the phytochrome-regulated phototropic response in tip cells of the mossCeratodon purpureus: a tip-located gradient shift of chlortetracy-cline (CTC)-stained calcium and a structural reorganization of apical microfilaments (MFs). We examined the interdependence of these processes. Cells were treated with the antimicrotubule drug oryzalin, the antimicrofilament drug cytochalasin-D, and the calcium channel blocker nifedipine. respectively. The effects on phototropic growth, on the structural alignment of the cytoskeleton (microtubules, MTs; microfilaments) and on the distribution of CTC-stained calcium were studied under each of these conditions. In gravitropically growing tip cells the apical MFs form a cortical collar-like structure, consisting of actin bundles with a parallel axial alignment. These MFs point towards the presumptive growing point, a weakly stained region in the tip of the cell from which bundles are absent. MTs are present in the cortex and in the endoplasm of the tip, predominantly oriented longitudinally. The MTs converge within the central apex. The cells show a steep tip-to-base CTC-calcium gradient with its highest signal in the central apex. Destruction of MTs by 1 M oryzalin induces several translocational effects: (i) the growing zone and phototropic outgrowth shift from the apex to subapical parts of the cell; (ii) the structural integrity of the apical MFs and the tip-to-base alignment of the CTC-calcium gradient are disturbed; and (iii) the red light induced gradient shift and the reorientation of MFs proceed in an expanded area spanning from the tip to subapical parts of the cell. Cytochalasin-D (10 g/ml) destroys the MFs. Under these conditions tip growth stops and the phototropic outgrowth is suppressed. The apical MT-structure and the CTC-calcium gradient are not influenced by the agent. Unilateral red light still induces the light-directed translocation of the gradient. Tip cells memorize a unilateral irradiation applied during growth inhibition with cytochalasin-D. After recovery in darkness the cells start to grow in the former light direction. The restoration of the MFs precedes the outgrowth. The structural alignment of the rebuilt actin bundles indicates the future growth direction. The calcium channel blocker nifedipine (10 M) also inhibits tip growth and concurrently phototropic outgrowth. Organization of microtubules (MTs) in relation to the behavior of nuclei was examined in dividing binucleate cells ofAdiantum capillus-veneris L. To induce binucleate cells, caffeine, an inhibitor of formation of the cell plate, was applied at 4 mM to synchronously dividing protonemal cells during cytokinesis (Murata and Wada 1993). Formation of the preprophase band (PPB) during the next cell cycle was examined in non-centrifuged and centrifuged cells. The two nuclei were separated or associated with one another in both non-centrifuged and centrifuged cells, although the location of the nuclei in the cylindrical protonemal cells was different (Murata and Wada 1993). Irrespective of centrifugation, a single PPB was formed around the nuclei in cells with associated nuclei. Two PPBs were formed in cells with separated nuclei in centrifuged cells. Patterns of mitosis and cytokinesis varied, depending on the location of the PPB and the distribution of the nuclei. The role of the nucleus in formation of the PPB is discussed.  相似文献

Abstract Production of protoplasts in different genera of filamentous fungi with their own lytic enzymes obtained from autolyzed cultures, as well as the regeneration of these protoplasts, has been studied. The existence of a yield stress in filamentous fermentation broths has important transport phenomena implications in the design and operation of bioreactors. Summary Changes in the organization of cortical actin microfilaments during phytochrome-mediated and blue light-induced photoorientation of chloroplasts were investigated by rhodamine-phalloidin staining in protonemal cells of the fernAdiantum capillusveneris. Low- and high-fluence rate responses were induced by partial irradiation of individual cells with a microbeam of 20 m in width. In the low-fluence rate responses to red and blue light, a circular structure composed of microfilaments was induced on the chloroplast concentrated in the irradiated region, on the side facing the plasma membrane, as already reported in the case of the low-fluence rate response induced by polarized red or blue light. Such a structure was not observed on the chloroplasts located far from the microbeam. Time-course studies revealed that the structure was induced after the chloroplasts gathered in the illuminated region and that the structure disappeared before chloroplasts moved out of this region when the microbeam was turned off. In the high-fluence rate response to blue light, chloroplasts avoided the irradiated site but accumulated in the shaded area adjacent the edges of microbeam. The circular structure made of microfilaments was also observed on the chloroplasts gathered in the area and it showed the same behavior with respect to its appearance and disappearance during a light/dark regime as in the case of the low-fluence rate response. However, no such circular structure was observed in the high-fluence rate response to red light, in which case the chloroplasts also avoided the illuminated region but no accumulation in the adjacent areas was induced. These results indicate that the circular structure composed of microfilaments may play a role in the anchorage of the chloroplast during intracellular photo-orientation.  相似文献
