DNA fingerprints of dogs and cats   总被引:26,自引:0,他引:26  
Human minisatellite probes consisting of tandem repeats of the 'core' sequence, a putative recombination signal in human DNA, cross-hybridize to multiple polymorphic fragments in dog and cat DNA to produce individual-specific DNA 'fingerprints'. Pedigree analysis shows that most of the DNA fragments detected in an individual are heterozygous, and that these fragments are derived from multiple dispersed autosomal loci. DNA fingerprints of cats and dogs should prove suitable for individual identification and for establishing family relationships. They are also suitable for rapid marker generation in large pedigrees and could be applied to linkage analysis in these animals.  相似文献

The primary structures of the alpha chains in hemoglobins from three stocks of mice with theHba w2,Hba w3, andHba w4 haplotypes were determined to establish whether the tentative alpha-chain assignments based on the results of isoelectric focusing patterns were correct. TheseHba haplotypes were identified in laboratory descendants of feral mice captured in different parts of the world. Hemoglobin from Centreville, Maryland,Mus musculus domesticus (Hba w2) contains equal amounts of alpha chains 1 and 3. Hemoglobin from CzechMus musculus musculus (Hba w4) contains equal amounts of alpha chains 3 and 4. Amino acid analysis of the alpha-globins of Skive DanishMus musculus musculus (Hba w3) establishes that its hemoglobin is comprised of about one-third alpha chain 2 as expected plus a greater amount of a unique alpha chain that has not been described previously. This unique alpha chain has glycine at position 25, isoleucine at position 62, and serine at position 68; it is called chain 7. Taurine is an important modulator of neuronal activity in the immature brain. In kittens, taurine deficiency causes serious dysfunction in the cerebellar and cerebral visual cortex. The processes of taurine transport in vitro were now studied for the first time in different brain areas in developing and adult cats. The uptake of taurine consisted initially of two saturable components, high- and low-affinity, in synaptosomal preparations from the developing cerebral cortex and cerebellum, but the high-affinity uptake component completely disappeared during maturation. The release of both endogenous and preloaded labeled taurine from brain slices measured in a superfusion system was severalfold stimulated with a slow onset by depolarizing K+ (50 mM) concentrations. K+ stimulation released markedly more taurine from the cerebral cortex, cerebellum and brain stem in kittens than in adult cats. The responses were largest in the cerebellum. Both uptake and release of taurine are thus highly efficient in the brain of kittens and may be of significance in view of the vulnerability of cats to taurine deficiency.  相似文献

The food habits of the Iriomote catFelis iriomotensis were studied by analyzing 177 feces collected monthly from 1987 to 1988. A total of 26 food items were identified. The frequency of lizards and frogs in the feces was higher than those of mammals and birds. The food habits changed greatly seasonally.Eumeces skinks were fed on most frequently, and found in the feces with a similar high frequency occurrence in March–April and July–September, while their proportion to the total number of food items was larger in March–April than in July–September. The cats fed on larg-sized skinks, adultEumeces kishinouyei, more in March–April than in other seasons. The number of skinks sighted in the course of a road census was greater from March to August, and large-sized skinks were sighted more in March–April than in July–August. The cats fed selectively on large-sized skinks in every season. Therefore, changes in the food habits depended on the food availability. Characteristics of food habits in the Iriomote cat are discussed in comparison with the food habits of other felids in temperate and tropical regions.  相似文献

An extraction and assay procedure to measure fecal estradiol-17β and progestin concentrations in several cat species was developed and validated for use for noninvasive monitoring of ovarian function. Fecal samples were collected over a range of 3–20 months from female tigers (three), lions (three), snow leopards (three), cheetahs (two), caracals (two), and domestic cats (five). Samples were extracted with 90% methanol, lipids removed with petroleum ether, and the estradiol and progestins in the methanol measured by radioimmunoassay (RIA). High Performance Liquid Chromatography (HPLC) fractionation and subsequent RIA of the fractions indicated that the estradiol-17β antiserum cross-reacted primarily with estradiol-17β in the feces of lions and tigers and was assumed to be specific for estradiol-17β in the feces of other species as well. However, there were several immunoreactive compounds, presumably progesterone metabolites, excreted in the feces which varied both quantitatively and qualitatively among species. The behavior of tigers, lions, cheetahs, and caracals was visually monitored during the collection period and frequency of sexual behaviors was positively correlated with increases in fecal estradiol in all species observed. The mean fecal estradiol-17β peaks were as follows: tigers, 128.0 ± 13.1; lions, 186.0 ± 14.8; snow leopards, 136.7 ± 15.9; cheetahs, 140.9 ± 9.0; caracals, 24.5 ± 4.0; and domestic cats 158.9 ± 19.3 ng/gm. Fecal progestin concentrations rose significantly (P < 0.001) only after breeding or during pregnancy and were as follows: tigers, 5.6 ± 0.6; lions, 1.9 ± 0.1; cheetahs, 8.4 ± 1.1; and caracals, 2.4 ± 0.4 μg/gm. Fecal progestins were elevated for one-half to two-thirds of the gestation length during presumed pseudopregnancy but remained elevated throughout successful pregnancies. These results suggest that ovarian function can be monitored noninvasively in the family Felidae by the measurement of fecal estradiol-17β and progestin concentrations. © 1995 Wiley-Liss, Inc.  相似文献

The time course and end products of estradiol metabolism were studied in the domestic cat, which has been chosen as a model for steroid metabolism studies in nondomestic felidae. Radiolabeled estradiol was injected intravenously into three adult female cats; one had a spontaneous estrus, one was induced with follicle-stimulating hormone, and one had been ovariohysterectomized; feces, urine, and blood were collected daily, and the radioactivity content was determined. Feces and urine contained 47 and 1% of the injected dose (0.33 μCi), respectively. Metabolites appeared earlier in the urine than in feces (d 1 vs d 2 postinjection), and excretion was completed on d 5; no radioactivity was detected in plasma 24 h postinjection. Estradiol metabolites were excreted as unconjugated estrogens (22%) and as conjugates hydrolyzable with β-glucuronidase and acid solvolysis (7 and 50%, respectively); the remaining 14% were not recoverable with any of the above methods. The major portion of the conjugates was estradiol-17β (64–80%) while 11–16% appeared as estrone. Endogenous cycles related to the spontaneous and induced ovarian activity were monitored by observation of estrous behavior, vaginal epithelium cornification, and plasma estradiol determination. The reproductive state of each animal had no effect on the time course or type of metabolite excreted. We found low proportions of injected radioactivity excreted in the urine and high residual levels remaining after hydrolysis and extraction in the feces. These findings suggest that although feces are an abundant source of estradiol metabolite in the cat, and probably in the exotic felidae, development of noninvasive methods for monitoring ovarian cycles in these species will depend on more efficient methods for urine hydrolysis, on the resolution of problems encountered in fecal steroid analysis, or on the identification of metabolites which may be measured directly in the urine without hydrolysis or extraction.  相似文献

Cats were exposed to 200 Brugia malayi larvae on one hind foot over a 3 week period. Six weeks after the initial exposure to B. malayi, 10 of the cats were challenged on both hind legs with a Group G streptococcus. The remaining 10 cats were not exposed to the streptococcus. Following bacterial challenge, the B. malayi-infected leg of 9 of 10 cats displayed sequelae including erysipelas and abscesses. In addition, 5 of the affected legs had an elephantoid appearance, both by gross observation and as seen at necropsy 10 weeks after the initial B. malayi infection. The contralateral, uninfected leg of each cat remained normal in appearance. Histologic processing and examination of the elephantoid tissue showed it to be collagen; eosinophils and mast cells were plentiful in the collagen matrix. In the controls, only 1 animal displayed erysipelas and no abscesses were seen. Lymphedema seen in the B. malayi-infected leg of 5 control cats was less extensive than in uninfected cats challenged with streptococci and at necropsy no significant collagen matrix was evident. The location and number of worms in the lymphatics were noted. This study demonstrated that secondary microbial infections can contribute to the causation of elephantiasis under certain circumstances and that developing B. malayi were in some way adversely affected by the streptococcal involvement of the filaria-infected lymphatics.  相似文献

Studies on feral cat diet offer important ecological information and are the first step towards determining their impact upon endangered species. However, in comparing seasonal changes in diet with seasonal prey availability, the scarce amount of research into oceanic islands worldwide must be considered when deciding if a specific population is actually affected by cat predation. Cat diet was analysed on Santa Luzia (Cabo Verde Islands) since this invasive predator is considered one of the main threats to native endangered species that require conservation measures. These previous studies were carried out in different seasons, providing contrasting results, skinks being more preyed upon in the rainy season and mice in the driest periods. To check these different results, we focussed on how cat diet varied seasonally in response to changes in prey abundance. Saurians were the most important prey group, followed by mice, invertebrates and birds. No seasonal differences were, however, observed in the different prey groups consumed, saurians being the main prey in both seasons. All cases reflected their respective abundances. Results corroborate the generalist and opportunistic trophic ecology of feral cats, providing important information to assess their impact on prey populations and design future eradication programmes.  相似文献
