目的:探讨长期四氢生物蝶呤(BH4)治疗对高脂血症(HL)大鼠血管脂质过氧化水平及血管力学性质的影响。方法:选用8周龄雄性Wistar大鼠54只,随机分为3组(n=18):对照组、高脂饮食组(HE组)和高脂饮食并腹腔注射BH4组(HL+BH4组),于第8、16和24周龄时每组各杀死6只大鼠测定血脂和脂质过氧化终产物丙二醛(MDA)水平,主动脉血管测量零应力状态张开角、压力-直径关系。结果:至B地治疗后的第16和24周龄,血脂无明显变化,但MDA明显降低(P〈0.01);HL+BH4组和HL组比较胸主动脉张开角显著减小(P〈0.01)、压力-直径(P-D)关系曲线上移。结论 BH4可以减轻由于长期高脂血症导致的脂质过氧化,恢复血管弹性,降低血管的结构异常改变。  相似文献

目的:探讨长期四氢生物喋呤(BH4)治疗对自发性高血压大鼠(SHR)血管形态及血管力学性质的影响;方法:选用4周龄雄性SHR36只,随机分为实验组和对照组,每组18只。实验组每周2次腹腔注射BH420mg/kg,对照组注射等容量生理盐水,于实验第4、16和26周龄时各取6只测量动脉收缩压(SBP),并使用计算机图像分析的方法分别测量主动脉血管零应力状态张开角、压力-直径关系及肠系膜动脉血管的壁/腔比值。结果:至BH4治疗后的第16和26周龄,SHR的SBP明显降低(P〈0.01);实验组SHR胸主动脉张开角显著减小(P〈0.01),压力-直径(P-D)关系曲线上移;实验组肠系膜动脉三级分支血管壁/腔(W/L)值减小(P〈0.05)。结论:BH4可以减弱由于长期高血压所导致的血管肥厚和管腔狭窄,恢复血管弹性。  相似文献

Guanine‐rich DNA sequences that may form G‐quadruplexes are located in strategic DNA loci with the ability to regulate biological events. G‐quadruplexes have been under intensive scrutiny owing to their potential to serve as novel drug targets in emerging anticancer strategies. Thermodynamic characterization of G‐quadruplexes is an important and necessary step in developing predictive algorithms for evaluating the conformational preferences of G‐rich sequences in the presence or the absence of their complementary C‐rich strands. We use a combination of spectroscopic, calorimetric, and volumetric techniques to characterize the folding/unfolding transitions of the 26‐meric human telomeric sequence d[A3G3(T2AG3)3A2]. In the presence of K+ ions, the latter adopts the hybrid‐1 G‐quadruplex conformation, a tightly packed structure with an unusually small number of solvent‐exposed atomic groups. The K+‐induced folding of the G‐quadruplex at room temperature is a slow process that involves significant accumulation of an intermediate at the early stages of the transition. The G‐quadruplex state of the oligomeric sequence is characterized by a larger volume and compressibility and a smaller expansibility than the coil state. These results are in qualitative agreement with each other all suggesting significant dehydration to accompany the G‐quadruplex formation. Based on our volume data, 432 ± 19 water molecules become released to the bulk upon the G‐quadruplex formation. This large number is consistent with a picture in which DNA dehydration is not limited to water molecules in direct contact with the regions that become buried but involves a general decrease in solute–solvent interactions all over the surface of the folded structure. © 2013 Wiley Periodicals, Inc. Scanning microcalorimetry has been used to study the high pressure effect on the main transition from the ripple gel P′β phase to the liquid crystal (Lα) phase in DPPC (dipalmitoylphosphatidylcholine). It has been demonstrated that an increase of the pressure by 200 MPa shifts the transition to higher temperatures by 36.4 degrees. The pressure increase does not affect the cooperativity of transition but reduces noticeably its enthalpy. The changes of the molar partial volume, isothermal compressibility as well as volume thermal expansibility during transition in DPPC suspension have been estimated. It has been shown that monovalent ions (Na+, Cl) in solution slightly affect the main thermodynamic parameters of the transition. Calcium ions significantly decrease distinction in compressibility and thermal expansibility between liquid-crystal and ripple gel phases of lipid suspension, which in its turn reflects less difference in their volume fluctuations.  相似文献
