Abstract

The antioxidant effects of ellagic acid (EA) and hesperidin (HES) against skeletal muscle ischemia/reperfusion injury (I/R) were performed. Hindlimb ischemia has been induced by tourniquet occlusion for 2?h on left hindlimb. At the end of ischemia, the tourniquate has been removed and initiated reperfusion for 2?h. EA (100?mg/kg) has been applied orally before ischemia/reperfusion in the EA?+?I/R group. HES (100?mg/kg) has been given orally in the HES?+?I/R group. The left gastrocnemius muscle has been harvested and stored immediately at??80?°C until assessed for the levels of MDA and antioxidant enzymes activities. MDA level has statistically increased in I/R group (p?<?0.05) compared to other groups. The muscle tissue antioxidant enzymes activities were lower than the other groups in the I/R group (p?<?0.05). EA and HES treatments significantly reversed the damage level in I/R, also activity of tissue SOD increased in the EA?+?I/R and HES?+?I/R groups.  相似文献

Parkinson's disease (PD) is the second most prevalent central nervous system (CNS) degenerative disease. Oxidative stress is one of key contributors to PD. Nuclear factor erythroid‐2‐related factor 2 (Nrf2) is considered to be a master regulator of many genes involved in anti‐oxidant stress to attenuate cell death. Therefore, activation of Nrf2 signalling provides an effective avenue to treat PD. Ellagic acid (EA), a natural polyphenolic contained in fruits and nuts, possesses amounts of pharmacological activities, such as anti‐oxidant stress and anti‐inflammation. Recent studies have confirmed EA could be used as a neuroprotective agent in neurodegenerative diseases. Here, mice subcutaneous injection of rotenone (ROT)‐induced DA neuronal damage was performed to investigate EA‐mediated neuroprotection. In addition, adult Nrf2 knockout mice and different cell cultures including MN9D‐enciched, MN9D‐BV‐2 and MN9D‐C6 cell co‐cultures were applied to explore the underlying mechanisms. Results demonstrated EA conferred neuroprotection against ROT‐induced DA neurotoxicity. Activation of Nrf2 signalling was involved in EA‐mediated DA neuroprotection, as evidenced by the following observations. First, EA activated Nrf2 signalling in ROT‐induced DA neuronal damage. Second, EA generated neuroprotection with the presence of astroglia and silence of Nrf2 in astroglia abolished EA‐mediated neuroprotection. Third, EA failed to produce DA neuroprotection in Nrf2 knockout mice. In conclusion, this study identified EA protected against DA neuronal loss via an Nrf2‐dependent manner.  相似文献

Six selected antioxidants (catechin, quercetin-3-galactoside, cyanidin-3-glucoside, pelargonidin-3-glucoside, ellagic and gallic acids) were evaluated in vitro for their antifungal activities and mode of action on Botrytis cinerea Pers., one of the most important pathogens of strawberries. Inhibitory effects were found for all the tested antioxidants, but varied at different fungal developmental stages. Catechin and quercetin-3-galactoside showed linear inhibitory effects on germ tube elongation, with the highest suppression ratios of 54.8% and 58.8% respectively. No significant effect was found on spore germination between treatments and control. Gallic acid showed very strong and linear inhibition on spore germination (r = ?0.95), but the effect diminished after spore germination. Cyanidin-3-glucoside and pelargonidin-3-glucoside provided effective control on the fungi as concentrations increased. The arresting effect of ellagic acid on development of B. cinerea was quadratic. Ellagic acid inhibited germ tube elongation and mycelial growth at its highest and lowest concentrations, while no effects were observed at its medium concentration used in this study.  相似文献

Ellagitannin-derived ellagic acid (EA) and colonic metabolite urolithins are functional dietary ingredients for cancer prevention, but the underlying mechanism need elucidation. Mucin-type O-glycosylation, initiated by polypeptide N-acetyl-α-galactosaminyltransferases (ppGalNAc-Ts), fine-tunes multiple biological processes and is closely associated with cancer progression. Herein, we aim to explore how specific tannin-based polyphenols affect tumor behavior of colorectal cancer cells (CRC) by modulating O-glycosylation. Utilizing HPLC-based enzyme assay, we find urolithin D (UroD), EA and gallic acid (GA) potently inhibit ppGalNAc-Ts. In particular, UroD inhibits ppGalNAc-T2 through a peptide/protein-competitive manner with nanomolar affinity. Computational simulations combined with site-directed mutagenesis further support the inhibitors’ mode of action. Moreover, lectin analysis and metabolic labelling reveal that UroD can reduce cell O-glycans but not N-glycans. Transwell experiments prove that UroD inhibits migration and invasion of CRC cells. Our work proves that specific tannin-based polyphenols can potently inhibit ppGalNAc-Ts activity to reduce cell O-glycosylation and lead to lowering the migration and invasion of CRC cells, suggesting that disturbance of mucin-type O-glycosylation is an important mechanism for the function of dietary polyphenols.  相似文献

High-speed countercurrent chromatography (HSCCC) has been successfully applied to the preparative separation of corilagin and ellagic acid in one step from the Chinese medicinal plant Phyllanthus urinaria L. by use of direct and successive injections of a crude methanolic extract. 红背山麻杆(Alchornea trewioides )为山麻杆属植物,作为传统的中药被用来减轻疾病和身体不适,但它的药效物质基础尚未完全清楚。为了全面掌握红背山麻杆药效物质基础,采用80%丙酮对其新鲜叶进行提取,并利用 MCI gel CHP 20P、Sephadex LH-20等色谱柱进行分离共得到9个化合物。这些化合物的结构经氢谱与碳谱比较分析鉴定为鞣花酸(1)、3-O-甲基没食子酸(2)、decarboxyellagic acid(3)、1-O-没食子酰基-β-D-葡萄糖(4)、1,6-二-O-没食子酰基-β-D-葡萄糖(5)、柯里拉京(6)、叶下珠鞣质 D(7)、furosonin(8)、老鹳草素(9)。其中化合物2~9均为首次从该属植物中分离得到。  相似文献

利用柱层析方法,从湖丹皮70%乙醇提取物的氯仿萃取部位分离得到5个化合物。运用理化及波谱方法,它们分别鉴定为丹皮酚(1)、3,6-二羟基-4-甲基苯乙酮(2)、去甲基丹皮酚(3)、3,3’-二甲氧基鞣花酸(4)和胡萝卜苷(5)。其中化合物4、5为首次从该种植物中分离得到。HPLC研究表明,湖丹皮中的去甲基丹皮酚含量高于国内其它产地。  相似文献

Divalent metal ions (Fe2+, Cu2+, Zn2+, Ni2+, and Mn2+) induced lipid oxidation in cooked, but not in raw fish. The extent of lipid oxidation, measured by the production of thiobarbituric acid reactive substances (TBRS), was increased with higher concentrations of iron, zinc, and nickel, but was decreased with increasing concentrations of copper and manganese. The natural products: ellagic acid, tannic acid, myricetin, and quercetin, inhibited lipid oxidation in cooked fish. The enhanced lipid oxidation caused by cupric ions (103 pmol/100 g fish) was also inhibited by the natural products. The degree of inhibition in copper-treated fish, however, was less than that in fish that had no added copper. The inhibition was concentration dependent. The antioxidative potency of the various natural products was independent of the type of metal ion-induced lipid oxidation. Ellagic acid was the most potent antioxidant (75.7–83.9%), followed by tannic acid (60.4–77.3%), myricetin (52.9–70.4%), and quercetin (32.6–44.2%).  相似文献
