Human pancreatic stellate cells (HPSCs) are an essential stromal component and mediators of pancreatic ductal adenocarcinoma (PDAC) progression. Small extracellular vesicles (sEVs) are membrane-enclosed nanoparticles involved in cell-to-cell communications and are released from stromal cells within PDAC. A detailed comparison of sEVs from normal pancreatic stellate cells (HPaStec) and from PDAC-associated stellate cells (HPSCs) remains a gap in our current knowledge regarding stellate cells and PDAC. We hypothesized there would be differences in sEVs secretion and protein expression that might contribute to PDAC biology. To test this hypothesis, we isolated sEVs using ultracentrifugation followed by characterization by electron microscopy and Nanoparticle Tracking Analysis. We report here our initial observations. First, HPSC cells derived from PDAC tumors secrete a higher volume of sEVs when compared to normal pancreatic stellate cells (HPaStec). Although our data revealed that both normal and tumor-derived sEVs demonstrated no significant biological effect on cancer cells, we observed efficient uptake of sEVs by both normal and cancer epithelial cells. Additionally, intact membrane-associated proteins on sEVs were essential for efficient uptake. We then compared sEV proteins isolated from HPSCs and HPaStecs cells using liquid chromatography–tandem mass spectrometry. Most of the 1481 protein groups identified were shared with the exosome database, ExoCarta. Eighty-seven protein groups were differentially expressed (selected by 2-fold difference and adjusted p value ≤0.05) between HPSC and HPaStec sEVs. Of note, HPSC sEVs contained dramatically more CSE1L (chromosome segregation 1–like protein), a described marker of poor prognosis in patients with pancreatic cancer. Based on our results, we have demonstrated unique populations of sEVs originating from stromal cells with PDAC and suggest that these are significant to cancer biology. The question of whether the appearance of a novel deleterious chemical in a preferred host plant could alter the relative degree of preference by a polyphagous insect during its lifetime was considered. The behavioral preference of fall armyworm, Spodoptera frugiperda (J. E. Smith), for corn (Zea mays L.) over red kidney bean Phaseolus spp. was quantified in a two-way choice test, and followed by a no-choice test in which the same larvae were fed either treated or untreated corn. Each treatment consisted of one of 15 compounds, including 13 naturally occurring allelochemicals and two synthetic insecticides. None of the test chemicals had any inherently deterrent effect at the rates tested. Deleterious effects on the larvae during exposure included reduced growth, reduced size, irritancy, and in some cases death. After this exposure the two way choice test was repeated. All test compounds failed to elicit a change in relative preference during the final two-way choice test. However, larvae exposed to gossypol and chrysanthemum monocarboxylic acid ester fed less than expected during the latter stages of no-choice conditions. The degree of aversion learning by each larva was related to the degree of prior sickness it had experienced.
Résumé Nous avons examiné si l'introduction d'une substance chimique nocive dans la plante-hôte préférée pouvait modifier la hiérarchie des choix durant le reste de la vie d'un insecte phytophage. Le comportement de préférence de Spodoptera frugiperda pour le maïs (Zea mays) vis à vis des Phaseolus, a été quantifié dans un dispositif à choix binaire, ont suivi des expériences au cours desquelles les chenilles devaient s'alimenter, soit sur maïs sain, soit sur maïs traité. Chaque essai concernait un composé chimique (15 au total), 13 étant des substances allélochimiques naturelles, 2 des insecticides de synthèse. Aux concentrations étudiées, aucun des produits n'a eu d'effet dissuasif. Les effets nocifs pendant l'exposition sur les chenilles comprenaient une croissance et une taille réduites, une irritation et, dans quelques cas, la mort. Aucune des expériences de choix binaire, avec des chenilles ayant subi cette exposition, n'a modifié les préférences. Cependant les chenilles, qui avaient été exposées au gossypol ou à l'acide monocarboxylique éthyl ester de chrysanthemum, ont consommé moins pendant les stades ultérieurs que ce que l'on pouvait attendre en absence de choix. Eleven cases of feeding on driver ants (Dorylus sp.) by mountain gorillas (Gorilla gorilla beringei) are described. Ant eating provides the gorillas with more animal protein and other nutrients per unit feeding time than
do other forms of insectivory that contribute to their diet, but it is so rare that it is unlikely to be of real nutritional
significance. Gorillas obtain ants with their hands and do not use tools. Immature individuals (except infants) ate more ants
than did adult females, and silverbacks were not seen to eat ants. These differences are more likely to reflect differences
in individual taste and interest in novelty than differences in nutritional strategy. Not all gorillas in the Virungas population
eat ants. Intra-population variability may be ecologically contingent, but ant eating appears to be a socially acquired and

Although various proteins and some electrolytes have been measured in human saliva, little systematic data about the major
and minor elemental components of this body fluid have been obtained. In order to obtain such data, concentrations of C, Na,
P, Cl, K, Ca, Sc, Cr, Fe, Co, Zn, Se, Br, Rb, Sb, I, and Cs in human parotid saliva were measured by instrumental nuclear
methods. The data obtained confirmed the relative lack of Zn in saliva of patients with hypogeusia (decreased taste acuity)
and suggested that concentrations of Na, Cl, Br, and Ca followed the order: normals > hypogeusia > hyposmia (decreased smell
acuity). To compare concentrations of elements in saliva with those in blood and urine, absolute concentrations were normalized
to that of Na through the use of a concept called an enrichment factor. On this basis, parotid saliva is relatively depleted
in Se, Zn, and Fe and enriched for most other elements relative to blood plasma indicating that the fluid is not simply a
transudate of blood plasma. Using this same technique, saliva composition was found more similar to urine than blood plasma,
being relatively depleted in Se, Cs, and Co, being enriched in I, Br, and Cr and having about the same relative concentrations
of P, Cl, Zn, Fe, Ca, K, and Rb. As the total body concentrations of many of the enriched elements in saliva are extremely
small, their enrichment in saliva suggests special roles for these elements in the oral cavity. Because of its accessibility,
ease of collection, and interaction with some body constituents, saliva represents a useful, albeit neglected, tool in the
diagnosis of some physiological and pathological changes in body function and in understanding important aspects of trace

Summary Colonies of HeLa cells cultured in media supplemented with human or bovine serum or both can be morphologically described
as three types: diffuse, intermediate, and compact, with their modal distribution depending on the serum or sera added to
the growth medium. We have observed that for a particular medium or serum system, the percentage of compact colonies remains
fairly constant under normal culture conditions, 0.2%, whereas the diffuse and intermediate colonies vary over a much wider
range. The presence of certain substances as trypsin, heparin and Darvan in the medium favor the increase of compact colonies
at the expense of other types. Furthermore, we have discovered that colonial morphology is influenced by cocultivation of
the HeLa cells with human fibroblastlike cells, the compact colonies increasing as the density of the fibroblast element introduced
into the mixed cultures is increased. Subsequent investigation revealed that conditioned medium from confluent fibroblast
and HeLa cell cultures contained a factor(s), that significantly increased the percentage of compact colonies. The factor
is nondialyzable, heat-stable and can be neutralized by serum. Recorded in this presentation are preliminary observations
on the kinetics of colony formation and the interaction among the three HeLa cell colony types, the diffuse, the intermediate,
and the compact. The factor's effect on HeLa cell colonial morphology is time dependent and rapidly reversed if the factor(s)

Summary New cell lines, designated as ML-DmDl≈10, were established from dissociated imaginal discs ofDrosophila melanogaster. The culture medium was prepared by mixing in a 1:1 ratio Cross and Sang’s M3(BF) medium, supplemented with 10% heat inactivated
fetal bovine serum (FBS), with the supernatant of a primary embryonic cell culture made in the M3(BF) medium and supplementing
this mixture with insulin. One cell line was established in the medium containing larval hemolymph instead of the primary
culture supernatan, and another was established in fresh M3(BF) medium supplemented with insulin and FBS. In these mediums,
imaginal disc cells first formed aggregates and cellular vesicles within a few weeks followed by the proliferation of thin-layered
cells around them after about 1 mo. Ten cell lines have so far been established from two kinds of imaginal discs and disc
mixtures. The ploidy of these cell lines was predominantly diploid. Population doubling time was about 50 to 70 h at 3 to
10 mo. after initiation of the culture. When the cell aggregates formed in vitro were implanted in metamorphosing larvae,
they differentiated at high frequency into adult cuticular strutures in the early phase of the primary culture. This differentiation
of aggregates was also observed, though at low frequency, in a culture maintained by dilution-transfer for 6 to 15 mo. in

The monoterpene derivative, 2-methylisoborneol (MIB), is produced by many blue-green algae and often is responsible for the “musty” taste/odor in aquaculturally raised finfish and potable water supplies. Although previous researchers have suggested that taste/odor metabolites are partitioned among cell constituents and that coregulation with pigment biosynthesis occurs, no structural evidence for these hypotheses exists. MIB was localized in cells of Oscillatoria limosa (Roth) Agardh at the ultrastructural level using standard gold-labeled antibody techniques. There was no apparent relationship between age of the cells and MIB synthesis; cells that were and were not undergoing active division had similar amounts of MIB label. There was no consistent partitioning of MIB label within cells. However, occasionally, specific label was observed along photosynthetic lamellae, suggesting a potential linkage of MIB synthesis and/or binding to the pigment systems. 相似文献

Psychophysical functions for the odor, taste, and flavor offive common flavorings were obtained by the method of magnitudeestimation. The stimuli included three simple compounds (vanillin,piperonal, and benzaldehyde) and two complex ones (natural vanillaextract and artificial almond essence). The odor intensity ofall the flavorings grew much less rapidly with concentrationthan did taste intensity. The growth of flavor for the complexsubstances and piperonal behaved very much like taste. For vanillinand benzaldehyde, the flavor functions resembled taste functionsat high concentrations but showed a tendency to flatten at lowerconcentrations. These findings implied that, at least for someflavorings, the growth of flavor reflects the most salient featureon the particular concentration range studied. At low concentrationsodor seems to be the most important feature and so flavor functionsare generally flat, but at high concentrations taste becomesthe salient feature and so flavor functions steepen. 相似文献

Summary Apical membrane currents were recorded from the taste pore of single taste buds maintained in the tongue of the rat, using a novel approach. Under a dissection microscope, the 150-m opening of a saline-filled glass pipette was positioned onto single fungiform papillae, while the mucosal surface outside the pipette was kept dry. Electrical responses of receptor cells to chemical stimuli, delivered from the pipette, were recorded through the pipette while the cells remained undamaged in their natural environment. We observed monophasic transient currents of 10-msec duration and 10–100 pA amplitude, apparently driven by action potentials arising spontaneously in the receptor cells. When perfusing the pipette with a solution of increased Na but unchanged Cl concentration, a stationary inward current (from pipette to taste cell) of 50–900 pA developed and the collective spike rate of the receptor cells increased. At a mucosal Na concentration of 250mm, the maximal collective spike rate of a bud was in the range of 6–10 sec–1. In a phasic/tonic response, the high initial rate was followed by an adaptive decrease to 0.5–2 sec–1. Buds of pure phasic response were also observed. Amiloride (30 m) present in the pipette solution reversibly and completely blocked the increase in spike rate induced by mucosal Na. Amiloride also decreased reversibly the stationary current which depended on the presence of mucosal Na (inhibition constant near 1 m). During washout of amiloride, spike amplitudes were first small, then increased, but always remained smaller than the amiloride-blockable stationary current of the bud. This is understandable since the stationary current of a bud arises from a multitude of taste cells, while each current spike is presumably generated by just one taste cell. We suggest that, in a Na-sensitive receptor cell, (i) the apical amiloride-blockable Na inward current serves as a generator current causing cell depolarization and firing of action potentials, and (ii) each current spike recorded from the taste pore arises mainly from a modulation of the apical Na inward current of this cell, because the action potential generated by the taste cell will transiently decrease or abolish the driving force for the apical Na inward current. Rat fetuses exposed to an odor stimulus and an aversive stimulus in utero showed an aversion to the odor when tested 16 days postnatally. Fetuses that also received 80 μg/kg Met-enkephalin showed a greater aversion to the odor stimulus than those subjects that did not receive the peptide. The difference between these groups was marginally significant. Control subjects did not show an aversion. But, subjects exposed to the odor and Met-enkephalin without the aversive stimulus, when tested, showed a significant preference for the odor over other control groups. These data show that associative learning in rat fetuses at 20 days of gestation may be enhanced by administration of Met-enkephalin. 相似文献
