A review of in vitro mutagenesis assessment of metal compounds in mammalian and nonmammalian test systems has been compiled.
Prokaryotic assays are ineffective or inconsistent in their detection of most metals as mutagens, with the notable exception
of hexavalent chromium. Mammalian assay systems appear to be similarly inappropriate for the screening of metal compounds
based upon the limited number of studies that have employed those compounds having known carcinogenic activity. Although of
limited value as screening tests for the detection of potentially carcinogenic metal compounds, the well-characterized in
vitro mutagenesis systems may prove to be of significant value as a means to elucidate mechanisms of metal genotoxicity. 相似文献

Bioaccessibility measurements have the potential to improve the accuracy of risk assessments and reduce the potential costs of remediation when they reveal that the solubility of chemicals in a matrix (e.g., soil) differs markedly from that in the critical toxicity study (i.e., the key study from which a toxicological or toxicity reference value is derived). We aimed to apply this approach to a brownfield site contaminated with chromium, and found that the speciation was CrIII, using a combination of alkaline digestion/diphenylcarbazide complexation and X-ray absorption near edge structure analysis. The bioaccessibility of Cr2O3, the compound on which a reference dose for CrIII is based, was substantially lower (<0.1%) than that of the CrIII in the soils, which was a maximum of 9%, giving relative bioaccessibility values of 13,000% in soil. This shows that the reference dose is based on essentially an insoluble compound, and thus we suggest that other compounds be considered for toxicity testing and derivation of reference dose. Two possibilities are CrCl3·6H2O and KCr(SO4)2·12H2O, which have been used for derivation of ecological toxicity reference values and are soluble at a range of dosing levels in our bioaccessibility tests. 相似文献

AbstractIn this paper, we describe a series of laboratory experiments which quantify the rate of Cr6+ reduction by Fe0. The main goal of these experiments was to determine the removal efficiency of Cr6+ by iron. The results indicate that Fe0 reduces Cr6+ to Cr3+ under alkaline and slightly acidic conditions. The removal efficiency rises with an increase of the initial concentration of Cr6+ (1 mg/L to 10 mg/L) when the quantity of Fe0 is stable. The removal efficiency increases as the quantity of Fe0 is raised when other conditions are constant. The removal efficiency would not be affected by other inorganic ions unless they were present at very high concentrations. When the initial concentration Cr6+ is 10mg/L and pH is 6.5–7.7, the final concentration of Cr6+ in effluent is less than 0.05 mg/L and the total Fe is less than 0.3 mg/L in effluent. 相似文献

Chromium is an essential trace element and is associated with some biological pathways, especially with glucose tolerance.
For these reasons, we decided to determine the concentration of chromium in two sets of Brazilian medicinal plants. The first
group consisted of plants that are considered as antidiabetic, whereas the second included plants that do not have this therapeutic
property. The concentration of chromium was determined by flameless atomic absorption. All the plants analyzed contain chromium
in the normal range for this element, but the hypoglycemic plants contain more chromium than the others (1–4 μg/g compared

Grass shrimp, Palaemonetes pugio, were exposed for 1 month to subacute concentrations of hexavalent chromium (0.5, 1.0, 2.0, 4.0 ppm) after which the gills, midgut, hepatopancreas, and antennal glands were examined for histopathological and ultrastructural changes. Pathological changes were greatest in the antennal glands, followed by hepatopancreas, gills, and midgut. Severe changes occurred in some shrimp, even at 0.5 ppm chromium. Cells of all tissues frequently had both swollen mitochondria and rough endoplasmic reticulum. Small, spherical or ring-like intranuclear inclusions, possibly indicative of cellular hyperactivity or manifestions of chromium and/or protein complexes, were most prevalent in the hepatopancreas and antennal glands but also occurred in the midgut and gills. Other major degenerative changes in the antennal glands were restricted to the labyrinth and included diminution of basal plasmalemmal infoldings and cytoplasmic density, nuclear hypertrophy followed by widespread nuclear pyknosis and epithelial desquamation. In severely altered hepatopancreas hypertrophy was indicated for the basal laminae, nuclei, and possibly for the nucleoli. There was an apparent reduction in mitotic events and many observed mitotic nuclei were abnormal. Abnormal midgut hypertrophy was present in only 8 of 20 examined shrimp, exposed to 0.5 and 1 ppm chromium. Further, the gills of only 10 of the 40 examined chromium-exposed shrimp possessed abnormal features detectable with ligh microscopy. Ultrastructural analysis of the latter indicated an increase in lysosomes and a decrease in cytoplasmic density. In addition, there was a pronounced diminution in the degree of lamellar, subcuticular plasmalemmal infolding. This latter feature is postulated to be a mechanism for the regulation of chromium influx. Possible explanations for most observed alterations in the above tissues are proposed. 相似文献

Adult grass shrimp were exposed to four concentrations (0.5, 1.0, 2.0 and 4.0 ppm) of hexavalent chromium for 28 days. At the end of the exposure period, over 50% of the surviving shrimp possessed cuticular lesions that had many of the gross characteristics of “shell disease.” These lesions were usually associated with articulations of the appendages and abdomen. Furthermore, it was found that at increasing levels of chromium exposure, there was a proportionate increase in the loss of limbs such that nearly 50% of the limbs were lost in grass shrimp exposed to the highest test concentration of chromium. Histological and ultrastructural examination of numerous lesions demonstrated a range of degenerative features within the subcuticular epithelium that included cytoplasmic vacuolization, mitochondrial swelling, chromatin emargination, and the presence of unusual nuclear inclusions that appear to indicate direct chromium toxicity. Additionally, a marked retardation in new epicuticle and exocuticle formation was observed in viable tissues associated with lesions in late premolt shrimp. It is proposed that chromium interferes with the normal functions of subcuticular epithelium, particularly cuticle formation, and subsequently causes structural weaknesses or perforations to develop in the cuticle of newly moted shrimp. Because of these chromium-induced exoskeletal deficiencies, a viaduct for pathogenic organisms (e.g., bacteria) and direct chromium influx is formed that perpetuates lesion development. 相似文献

Uncultivated plants growing on disturbed sites may be useful for assessing the bioavailability of some metals in soils, and thus the potential for metal mobilization up the terrestrial food chain, an important element in ecological risk assessment. A planted chicory cultivar (Cichorium intybus L. var. foliosum Hegi.) and the uncultivated plants horseweed (Canada fleabane) (Erigeron canadensis L.) and dogfennel (Eupatorium capillifolium (Lam.) Small) were evaluated for their ability to act as index plant species for soil Cd, Cr, Ni, and V at two field sites where these metals had been applied five yr previously to two highly weathered sandy Ultisols. Soil Cd was available to all analyzed plant tissues of all three plant species at both sites, particularly on the sandier Blanton soil. Chicory was an effective index plant for Cd on the finer textured Orangeburg soil but functioned as an indicator plant (toxicity symptoms were observed) on the sandier Blanton soil. Horseweed and dogfennel were effective index plants for Cd in both contaminated soils. Soil Cr, Ni, and V were less bioavailable than soil Cd and plant metal uptake was more sensitive to residual soil Cr, Ni, and V than was soil extraction with double acid. Horseweed and chicory may have potential as index plants for soil Cr. Chicory may have potential as a Ni index plant. Chicory and dogfennel may have potential as V index plants. 相似文献

Chromium can be removed from ground water by the unicellular yeast, Saccharomyces cerevisiae. Local ground water maintains chromium as CrO4

conditions (8.2 and +343 mV, respectively). In laboratory studies, we used commercially available, nonpathogenic S. cerevisiae to remove hexavalent chromium [Cr(VI)] from ground water. The influence of parameters such as temperature, pH, and glucose concentration on Cr(VI) removal by yeast were also examined. S. cerevisiae removed Cr(VI) under aerobic and anaerobic conditions, with a slightly greater rate occurring under anaerobic conditions. Our kinetic studies reveal a reaction rate (Vmax) of 0.227 mg h-1 (g dry wt biomass)-1 and a Michaelis constant (Km) of 145 mg/l in natural ground water using mature S. cerevisiae cultures. We found a rapid (within 2 minutes) initial removal of Cr(VI) with freshly hydrated cells [55–67 mg h-1 (g dry wt biomass)-1] followed by a much slower uptake [0.6–1.1 mg h-1 (g dry wt biomass)-1] that diminished with time. A materials-balance for a batch reactor over 24 hours resulted in an overall shift in redox potential from +321 to +90 mV, an increase in the bicarbonate concentration (150–3400 mg/l) and a decrease in the Cr(VI) concentration in the effluent (1.9-0 mg/l). 相似文献

The influence of chromium concentration on ethylene production in bean plants ( Phaseolus vulgaris L. cv. Contender) was investigated. A Cr ion-induced inhibition of ethylene synthesis from endogenous 1-aminocyclopropane-1-carboxylic acid (ACC) was observed within both leaf discs floated on 2 m M CrO2− 4 or Cr3+ and leaf discs from plants cultured in nutrient solutions containing 10, 20 or 40 μ M CrO2− 4 . However, Cr ions supplied either to plants with the nutrient solution or to discs with the incubation medium rather increased the conversion of exogenous ACC to ethylene. Primary leaves of plants exposed to CrO2− 4 -containing nutrient solutions showed a statistically insignificant decrease of ACC-synthase activity. In the trifoliolate leaves of plants exposed to 10 μ M CrO2− 4 , in which a significant decrease of ethylene production from endogenous ACC was observed, a substantial increase of ACC synthase was found. These results indicate that Cr ion-induced inhibition of ethylene production is not due to a breakdown of membrane integrity, which is necessary for ethylene forming enzyme activity, but caused by metabolic alterations leading to decreased ACC availability. Chromium ions may act by inhibiting ACC synthase activity or by diverting a metabolic step prior to the ACC synthase catalyzed reaction. 相似文献
